Cancer stem cells located in specific microenvironment,that play an important role in the proliferation and metastasis of tumor cells.Hypoxia is one of the important features of tumor tissue microenvironment.A series of studies results show that the hypoxic microenvironment of tumor stem cells could promote tumor metastasis,which provides new clues for understanding tumor pathogenesis and tumors treatment.
2.PTEN and gastric cancer

The development and progression of gastric cancer is a multi-factor,multi-gene,multi-stage and multi-approach sequential process.Phosphatase and tensin homologue deleted on chromosome 10 (PTEN)is so far the first discovery of a dual specificity phosphatase activity of the tumor suppressor gene,which regulates the cell cycle and multiple signaling pathways of gastric cancer and plays an important role in the development and progression of gastric cancer.Further studies on PTEN will provide a new way for the early diagnosis,treatment and prognosis of gastric cancer.
3.Researches on Lgr5 the newest biological marker of stem cells and the relationship with tumour

Lgr5 is one of the G protein-coupled receptors families, the newest research displays that it has the potency as the special marker of stem cell, it also has a great relationship with tumor formation and development. Lgr5 may be a marker of tumor stem cells, and can be the new treatment target.
4.The diagnostic value of CT angiography on diabetic lower extremity vascular disease

Objective To investigate the value of vascular disease of lower extremity of 64 slice spiral CT angiography in patients with diabetic foot.Methods 48 patients with diabetic lower limb underwent arterial CT angi-ography,Using CT a variety of post -processing techniques and compared with the original image.Results 48 patients were successfully obtained CTA image,41 patients had peripheral artery disease,40 cases were bilateral,39 cases of unilateral or bilateral had severe stenosis or occlusion;25 cases had obvious collateral circulation.Lower extremity arterial segments CTA and DSA showed good agreement (Kappa Value =0.887).Positive predictive rate was 92.5%,the negative predictive value was 98.6%.After delayed scanning,CTA on the foot artery count develo-ping increased,checked with the DSA consistency.Conclusion 64 slice spiral CTA can accurately and comprehen-sive assessment of lower extremity arterial disease of diabetic foot.Diabetic lower limb vascular lesions can be used as the preferred method of examination.
5.Effect of GM_1 on rCBF and microvessel after cerebral vasospasm in rats

Objective To investigate the effect of GM 1 on vasospasm after subarachnoid hemorrhage.Methods Twenty-five wistar rats receiving intracisternal injection of 0.15 ml autologous nonheparinized blood or 0.15 ml saline two times were divided into five groups as follows: control group, cerebral vasospasm (CVS) group, Nimodipine group, GM 1 therapy group and GM 1 prevent group. BACKGROUND: Generally speaking, neuroendocrine cells have been not observed in normal breast tissue but found in breast carcinoma tissue which was affected by local microenvironment and hormone level during differentiation of breast epithelial stem cells.OBJECTIVE: By detecting expressions of breast cancer resistance protein (BCRP), cytokeratin-8 (CK8), and chromogranin-A (CgA) in breast carcinoma tissue, to explore the possible mechanism of neuroendocrine cells observed in breast carcinoma tissue during differentiation of multi-lineage potential of bone marrow mesenchymal stem cells.METHODS: BCRP, CK8, and CgA were used as markers for SP stem cells, glandular epithelium differentiation, and neuroendocrine differentiation, respectively. Immunohistochemistry was used to detect the expressions of BCRP, CK8, and CgA in breast tissues of 89 subjects and analyze their correlation. RESULTS AND CONCLUSION: Both BCRP and CK8 expressions were observed in normal breast tissues, hyperplastic tissues, and breast carcinoma tissue. BCRP expression was increased in the breast carcinoma tissue; CK8 expression was decreased with the abnormal differentiation of breast tissue; CgA expression was only detected in breast carcinoma tissue. BCRP expression was significantly correlated with positive CgA expression (P < 0.01), but it was no correlation with positive CK8 expression in normal breast tissues, hyperplastic tissues, and breast carcinoma tissue (P=0.069). The results suggested that neuroendocrine cells were not observed in both normal breast tissues and hyperplastic tissues but in breast carcinoma tissue, which possibly correlated to differentiation of multi-lineage potential of bone marrow mesenchymal stem cells.
7.Undergraduate course of general practice in higher medical education

Data were analyzed from self-filled questionnaires for the students who took the elective course of general practice in China Medical University. 95.5% of students thought it was necessary to study general practice in the college. 0-10 was used to score the degree of how they want to choose general practice as a career, the median was 0 before they had the class and the median was 5 after the class (Wilcoxon rank test, P<0.01). But there were still some places need to be improved in terms of curriculum design, teaching methods and community practice. The General Practice teaching need to be innovated and the teaching mode should be different from the traditional one.
8.Establishment and application of dual real-time fluorescent RT-PCR method for detection of Enterovirus

Objective To develop a dual real-time fluorescent RT-PCR method for rapid detection of enterovirus(EV)and en terovirus type 71(EV71).Methods Specific primers and probes were designed and the dual real-time fluorescent RT-PCR reaction system was established.The quantitative standard curve was drawn;its sensitivity and precision were evaluated.Feces and throat swab specimens of 109 clinical patients with hand foot and mouth disease were collected and tested by using this method.Then the obtained results were compared with those detected by commercial EV71 PCR kit.Results The relative coefficient(2)of EV and EV71 standard curve established by the dual real-time fluorescent RT-PCR method were both 0.998.Its sensitivity reached 0.5 TCID50/mL for detecting EV and 0.05 TCID50/mL for detecting EV71.The within-run precision for detecting EV and EV71 was <3% and total precision≤4%.The results showed good specificity for the detection of enterovirus and non-enterovirus.In 109 detected clinical samples,84 cases of EV positive samples were detected,in which 56 cases were EV71 positive with the total positive rate of 51.4 %,which was consistent with the result of simple fluorescent RT-PCR commercialization kit(P=1.000).Conclusion The established dual real-time fluorescent RT-PCR method has high sensitivity and good stability,which has an important significance for early high throughput rapid diagnosis of hand foot and mouth disease.
9.Protective effect and mechanism of ulinastatin on rats with hemorrhagic shock

AIM: To investigate the protective effect of ulinastatin on rats with hemorrhagic shock. METHODS: A prospective, controlled animal study was designed. The model of hemorrhagic shock in rats was produced by Chaudry method. After 60 min, rats were resuscitated by transfusion of shed blood and normal saline, but a half of them were treated with ulinastatin. At different time points after reperfusion, the levels of tumor necrosis factor-alpha (TNF-?), interleukin-6 (IL-6), malondialdehyde (MDA) and superoxide dismutase (SOD) in serum were detected. RESULTS: The levels of TNF-?, IL-6 and MDA significantly increased and the activity of SOD decreased. In the ulinastatin-treated groups, the blood pressure and heart rate were obviously improved; the levels of TNF-?, IL-6 and MDA significantly decreased and the activity of SOD had little change after hemorrhagic shock and reperfusion. CONCLUSION: Ulinastatin has a protection effect on rats with hemorrhagic shock by suppressing the production of inflammatory factors and reducing oxidative damage.
