Objective To investigate the gene expression profiles of type Ⅳ collagen, matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) at the onset of radiation pulmonary fibrosis, so as to explore their possible roles in the pathogenesis of pulmonary radiation injury. Methods The whole lung was irradiated with 20Gy of 60Co ?-ray in C57 mice. At 1, 2 and 4 weeks after irradiation, both irradiated mice and corresponding number of normal control mice were sacrificed. Alterations in mRNA transcription of type Ⅳ collagen, MMP-9 and TIMP-1 were observed by RT-PCR at different time points post-irradiation. The mean optical density of electrophoretic bands was measured and analyzed by image analyzer. Result The mRNA expression of type Ⅳ collagen was up-regulated gradually after irradiation with the passage of time. The mean optical density of electrophoretic band was 0.202 at 1 week post irradiation and reached its peak value (0.340) at 4 weeks post irradiation; the mRNA expression of MMP-9 was up-regulated significantly and reached its peak value (0.730) at 1 week post irradiation. It began to lower at 2 weeks (0.592). The mRNA transcription of TIMP-1 began to increase slightly 1 and 2 weeks post irradiation (0.987) and reached its peak value (2.027) at 4 weeks post irradiation. Conclusion 60Co ?-ray can stimulate mRNA expression of type Ⅳ collagen, MMP-9 and TIMP-1 with different profiles at different time points. Complex interactions among them may result in early tissue remodeling after pulmonary radiation injury, and it bears close relation with subsequent pulmonary fibrosis.

AIM: To investigate the mechanisms of augmenter of liver regeneration (ALR) in promoting damaged hepatocyte proliferation.METHODS: The effects of Kupffer cell condition medium (KCCM+) stimulated by ALR on damaged hepatocyte proliferation were studied by MTT. The localization of ALR binding to Kupffer cell membrane and in intact rat liver was studied by immunohistochemistry. The IL-6 expression in Kupffer cells stimulated with ALR was observed by immunohistochemistry. RESULTS: The proliferation of damaged hepatocytes stimulated with KCCM+ was increased significantly. ALR immunostaining particles in plasm of hepatocyte were found in intact liver. The rough immunostaining particles of ALR were seen on the surface of Kupffer cell membrane. Immunostaining particles of IL-6 in Kupffer cells induced by ALR increased. CONCLUSION: ALR promotes proliferation of damaged hepatocytes indirectly by stimulating Kupffer cells.

Objective To observe the effects of functional ambulation training in a realistic environment based on the activities of daily living among stroke patients with hemiplegia. Methods Thirty-two stroke survivors with hemiplegia were randomly divided into an experimental group and a control group, each of 16.Both groups were given routine rehabilitation training, while the experimental group was additionally given functional ambulation training based on the activities of daily living in a realistic environment for 60 min per day, five days a week for six weeks. Both groups were evaluated using the mini-mental state examination (MMSE),the Holden walking functional class assessment (HWFCA),the Berg balance scale (BBS),functional gait assessment (FGA),the 5 times sit to stand test (FTSST),the timed up and go test (TUGT),and rated using the specific activity balance confidence scale (ABC) and the modified Barthel index (MBI).Results Before the training,no significant differences between the two groups were found in terms of any of the measurements. After the six weeks of training, significant improvement was observed in all of the outcome measures except the MMSE and the HWFCA,with the experimental group scoring significantly better,on average, than the control group. Conclusion When combined with routine rehabilitation training, functional ambulation training based on the activities of daily living in a realistic environment can significantly enhance the functional gait, balance and postural control of stroke survivors. This should facilitate their activities in daily life and improve their confidence in maintaining their balance.

Total 3 483 students in 4 to 9 grade from primary and middle school in Xiling District of Yichang city were selected by stratified cluster random sampling method between September and October 2016 to participate in screening for abnormal angle of trunk rotation(ATR),including 1 797 boys and 1 686 girls aged 9-15 years. The ATR was measured with Scoliometer and the ATR >5°was defined as abnormal. The overall abnormal ATR rate was 7.60%(265/3 483), that in boys was 4.73%(85/1797)and in girls was 10.68%(180/1 686)(χ2=43.75,P<0.05).The abnormal ATR rate in age group 13-15 was higher than that in age group 9-12[10.62%(159/1 496)vs.5.33%(106/1 987),χ2=36.92,P<0.01].Students with low BMI had higher abnormal ATR rate than that with normal or higher BMI(χ2=30.00,P<0.01);and students with left handedness had lower abnormal ATR rate than that with right handedness or double handedness(χ2=6.59,P=0.04).The abnormal ATR rate was not associated with dietary habits or sports in the daily life of students. The results show that gender, age,BMI and handedness affect abnormal rate of ATR.

Objective To investigate the protective effect of the compound drug Weng-Li-Tong(WLT)against cisplatin(CDDP)-induced hepatocyte injury. Methods A cellular injury model was established by treating murine hepatocyte line BNL CL.2 with CDDP(80 μmol/L).Experimental groups were divided as follows:CDDP group(modeling only),WLT group(intervention with 1 g/L WLT),WLT+CDDP group(co-administration of CDDP and 1 g/L WLT),and a control group(normal culture). The protective effect of the compound drug WLT on CDDP-mediated hepatocyte injury was evaluated using CCK-8 assay,PI staining, crystal violet staining,Western blotting, reactive oxygen species(ROS)detection, and apoptosis analysis. Results Compared with the control group, the number of dead cells increased significantly(P<0.001)in the CDDP group, but no cytotoxicity was observed in the WLT group. The hepatocyte morphology in the WLT+CDDP group showed improvement with no obvious shrinkage compared to the CDDP group,as evidenced by the reduced proportion of PI-positive cells.Crystal violet staining results also indicated a higher cell count in the WLT+CDDP group than in the CDDP group, suggesting the protective effect of WLT against CDDP-mediated liver injury.Under CDDP intervention, the expression of the apoptosis-related protein Cleaved Caspase-3 increased. However,in the WLT+CDDP group, the expression of Cleaved Caspase-3 decreased, while the expression of the anti-apoptotic protein Bcl-2 increased.Additionally, compared to the CDDP group, the WLT+CDDP group showed a reduction in ROS production[DCFH-DA staining positive rate(%):56.20±1.65 vs.44.57±0.31]and a decrease in the proportion of apoptotic cells[proportion of early and late apoptotic cells(%):43.60±0.44 vs.19.57±0.78;33.30±1.02 vs.14.83±0.57].Conclusion The compound drug WLT exhibits a protective effect against CDDP-mediated hepatocyte injury, suggesting potential therapeutic value in acute liver injury models.
