1.Distribution and Drug Resistance of Enterococcus in Hospital:Analysis and Prevention

OBJECTIVE To study the status of the distribution and drug resistance of Enterococcus in hospital,in order to further provide the effective reference for rational use of antibacterial and the prevention of Enterococcus in clinical therapy. METHODS The drug sensitivity and resistance of 201 strains of Enterococcus were determined by MicroScan WalkAway40 automatic microorganism system. RESULTS The highest incidence rate of Enterococcus was E. faecium (48.8%),the second was E. faecalis (37.3%),and the others were E. gallinarum (13.9%). The drug resistance of E. faecium was much higher than that of E. faecalis. CONCLUSIONS The most of Enterococcus infection is due to E. faecium,and there is a lot of difference for drug resistance in the strains of Enterococcus. It is necessary to rationally use antibacterials on the basis of the drug resistance,infection sites and grade of infection.
2.Effects of 3 Kinds of Serum Containning Blood-activating and Stasis-eliminating TCM Compound Formu-las on JAK/STAT Signal Pathway of Glioma U251 Cells

OBJECTIVE:To evaluate the effects of 3 kinds of serum containing blood-activating and stasis-eliminating TCM compound formulas on aggressive behavior,Janus kinase (JAK)/signal transduction and transcriptional activator (STAT) pathway of glioma U251 cells. METHODS:Rats were randomly divided into normal saline group (5 mL/kg),Taohong Siwu decoction group(5.7 g/kg),Xuefu Zhuyu decoction(8.5 g/kg)and Didang decoction(2.8 g/kg),calculated by crude drug,intragastrically administrated once a day,for 10 d. 10% drug-containing serum culture medium was prepared after 2 h of last administration. After 10% drug-containing serum culture medium intervening U251 cells for 1 week,Transwell method was conducted to detect the cell invasion rate, Western blot was adopted to detect the metal matrix protease 2 (MMP-2), MMP-9, phosphorylated JAK2 (p-JAK2),phosphorylated STAT3(p-STAT3)protein expression;and real-time fluorescence quantitative polymerase chain reaction method was used to detect MMP-2,MMP-9 mRNA expression. RESULTS:Compared with blank serum,Xuefu Zhuyu decoction drug-containing serum can reduce cell invasion rate (P<0.05),decrease the MMP-2,MMP-9 mRNA and its protein expression, and p-JAK2,p-STAT3 protein expression in cells (P<0.05 or P<0.01),while there was no significant difference in above-men-tioned indexes in Taohong Siwu decoction drug-containing serum group and Didang drug-containing serum group(P>0.05). CON-CLUSIONS:In the 3 kinds of blood-activating and stasis-eliminating TCM compound formulas,Xuefu Zhuyu decoction shows sig-nificant invasive effect on inhibiting U251 cells;the mechanism may be related to inhibiting the activation of JAK2/STAT3 signal pathway and decreasing MMP-2,MMP-9 gene and protein expressions.
3.Reliability and validity of adolescent sedentary behavior scale ( Chinese version) in junior middle school students

Objective To evaluate the reliability and validity of adolescent sedentary behavior scales ( Chinese version) in junior middle school students. Methods A total of 782 junior students were tested through multistage sampling method. The reliability was assessed, using Cronbach's αcoefficient and split-half reliability,while its validity was tested through ICC, correlation analysis and confirmatory factor analysis. Results The internal consistency reliability for total scale score ( Cronbach's α = 0.822 ) ,change strategies score ( Cronbach's α =0.882 ), decisional balance/pros score( Cronbach's α = 0.478 ), decisional balance/cons score ( Cronbach's α =0.631 ) ,and self-efficacy score (Cronbach's α = 0.645 ) ;the split-half reliability for total scale score was 0.857,four domains split-half reliability ranged between 0.488 ~ 0.896; the correlations between items and the domains ranged 0.226 ~ 0.768, except the correlations coefficients between decisional balance (cons score) and change strategies score, decisional balance ( cons score) and decisional ( pros score) were - 0.052,0.090 ( P < 0.05 ),respectively. Low to moderate correlations between domains ranged from -0.210 to 0.394 ( P < 0.01 ), while moderate to high correlations between domain and total scale. Correlation coefficients ranged between 0.365 to 0.554( P < 0.01 ). Conclusion The reliability and validity of adolescent sedentary behavior scales ( Chinese version) were good, and adapt to measure junior middle school students' sedentary behavior.
4.Association of Body Mass Index, Waist Circumference and Serum Lipid Levels with Essential Hypertension

Objective To explore the relationship of different body mass index, waist circumference and serum lipid levels with essential hypertension among residents of Ningbo. And to explore the predictive value of BMI and WC (Waist Cir-cumference) for hypertension. Methods Adopting cluster sampling, 2 303 people (1 081 cases of hypertension and 1 222 people with normal blood pressure) were collected in the four townships of Ningbo City. We conducted questionnaires, physi-cal examination and blood lipid testing for all of subjects. WC group:WC≥85 cm in male or WC≥80 cm in female is central obesity, and the others are normal. BMI groups:BMI≥24 kg/m2 is overweight, and BMI<24 kg/m2 is normal. The relationship of BMI, WC and serum lipid levels with hypertension was analyzed by multivariate unconditional logistic regression model. The predictive value of BMI and WC for hypertension was analyzed by receiver operating characteristic (ROC) curve. Results WC, BMI, total cholesterol (TC), triglyceride (TG) and low density lipoprotein cholesterol (LDL-C) in hypertension group were higher than those in control group (P<0.01). And WC, BMI, TG, LDL-C is the impact factor for hypertension. The overweight rate and central obesity rate in hypertension group were higher than control group (P<0.01). TG and LDL-C level abnormality, overweight and central obesity were the risk factors of hypertension. As a risk factor of hypertension, over-weight with central obesity group is higher than overweight with normal waistline group, simple central obesity group and nor-mal group. In the area under the ROC curve, BMI is higher than WC in both male and female population. Conclusion WC, BMI, TG and LDL-C abnormalities were risk factors for hypertension, and BMI was a better predictive factor for hyper-tension than WC.
5.Experimental study of three different β-receptor blockers in treatment on murine hemangioendothelioma

Objective:To comparison of three different beta blockers on murine hemangioma (EOMA cells) cells in vitro and in vivo effects.Preliminary study on the therapeutic effect of propranolol on vascular tumor in mice and possible mechanisms , provide a reference for beta blockers in the treatment of infantile hemangioma .Methods: Comparative study on the effects of three kinds of different β-receptor blockers---metoprolol, propranolol and butoxamine , on the proliferation and apoptosis of Mouse Hemangioendothelioma Endothelial cell (EOMA cells) was conducted in vitro.EOMA cells were cultured in vitro,randomly divided into different groups,propranolol and timolol were added into the medium respectively ,after 24 h intervention.MTT assay and acridine orange staining assay were conducted respectively to detect cell viability and apoptosis level .EOMA cells were transplanted into nude mice in vivo.Tumor volume growth to 100 mm3 ,animals were randomly divided into 4 groups respectively ,the control group ,metoprolol group,Bhutto Samin group and propranolol group ,drug group according to 2 mg/( kg? d) oral gavage ,control group were given an equal volume of saline ( NS ) , every two days measurement tumor volume size .Serum levels of tumor necrosis factor alpha ( TNF-α) and vascular endothelial growth factor ( VEGF ) were detected by enzyme linked immunosorbent assay ( ELISA ) in the end of the experiment.Results:For propranolol,after 24 h treatment,significant differences of cell viability and apoptosis were noted (P<0.05) at the concentration of 50 μmol/L,while continuing to increase to 800 μmol/L,the cell survival rate decreased sharply to close to 10%. Acridine orange staining at the 50 μmol/L group after 24 h revealed many apoptotic cells .For metoprolol and butoxa mine ,significant differences of cell viability and apoptosis were noted ( P<0.05 ) at the concentration of 100 μmol/L,while continuing to increase to 800μmol/L,the cell survival rate decreased sharply to close to 20%.It was significantly higher than propranolol group at the same concentration ( P<0.05 ) .It showed a similar trend in acridine orange staining .In vivo experiments showed that the end of the experiment of metoprolol , butoxamine group and propranolol drugs in mice tumor volume , respectively ( 1 642.8 ±89.3 ) , ( 1 529.3 ± 119.1) and (752.7±46.5)mm3,significantly lower than the control group of mice tumor volume of (2 023.3±123.0) mm3(P<0.001).Metoprolol,butoxamine mice and propranolol drugs group ,serum VEGF levels for (606.5±105.8 ) pg/ml,(534.3±243.2 ) pg/ml and (420.1±123.7) pg/ml, significantly lower than the PBS control group [(825.8±145.7) pg/ml,(P<0.05)],the TNF alpha result was followed by(301.3±62.3) pg/ml,(305.1±53.8) pg/ml and (288.8±59.5) pg/ml,significantly lower than the normal control group [(444±100.4) pg/ml,P<0.05].Conclusion:Three kinds of beta-blockers can effectively inhibit EOMA cells proliferation and induce apoptosis in vitro, the role of propranolol more significantly than butoxamine and metoprolol .Three kinds of beta blockers restrain the growth of the hemangioma in vivo ,in which the inhibitory effect of propranolol is stronger than the metoprolol and butoxa mine.Three kinds of beta blockers can lower the levels of VEGF and TNF-αin vivo.Indicating that propranolol on vascular tumor in mice may be one of the mechanisms of β1 and β2 receptor synergy effect and its mechanism in the treatment of hemangioma may be associated with VEGF and TNF-α.
6.CT three-dimensional reconstruction in percutaneous balloon compression of the semilunar ganglion of trigeminal nerves

Objective:To explore the CT three-dimensional reconstruction in percutaneous balloon compression of the semilunar ganglion of trigeminal nerves.Methods:Thirty-one patients with trigeminal neuralgia, admitted to our hospital from May 2018 to August 2018, were treated by percutaneous balloon compression of the semilunar ganglion of trigeminal nerves. All patients underwent intraoperative mobile CT scan; the distance from the external aperture of foramen ovale to the petrous ridge, the angle from puncture needle and the petrous ridge, and the angle from puncture needle and the clivus were analyzed by three-dimensional reconstruction technology. Prognoses were recorded immediately after surgery and 10-12 months after follow-up.Results:The distance from the external aperture of foramen ovale to the petrous ridgea was (2.14±0.17) cm, ranged from 1.83 cm to 2.42 cm. The angle from puncture needle and the petrous ridge was (102.03±7.60)°, ranged from 91.00° to 116.00°. The angle from puncture needle to the clivus was (68.03±7.06)°, ranged from 52.00° to 80.00°. The puncture succeeded in all patients under the help of CT three-dimensional reconstruction. The volume of the inflated balloon ranged from 0.30 cm 3 to 0.47 cm 3 (averaged 0.42±0.05 cm 3). The compression time ranged from 3.00 min to 5.00 min (averaged 4.72±0.53 min). The incidence of diplopia was 3.23%, and the incidence of dry eyes/decreased corneal sensation was 12.90%. During the follow-up, one patient relapsed 10 months after surgery; the left patients recovered with disappeared complications. Conclusion:The CT 3D reconstruction can reduce the difficulty of ovale foramen puncture during operation, assist the judgment and adjustment of intraoperative balloon volume, and improve the surgical efficacy.
7.Expression of microRNA-6768-5p in lung cancer tissue and its effect on malignant biological behavior of lung cancer cells

Objective To investigate the expression of miR-6768-5p in lung cancer tissue and its effect on the proliferation and invasion of lung cancer cells through targeted regulation of carboxypeptidase A4(CPA4).Methods The expression of miR-6768-5p in lung cancer tissues and adjacent tissues was analyzed u-sing the TCGA database.Quantitative real-time PCR(qPCR)was used to detect the expression of miR-6768-5p in human lung cancer cell lines(HCC1588,H1650,H1299,A549,HCC827)and normal alveolar epithelial cells(HPAEpiC cells).Lung cancer cells were transfected with NC mimics and miR-6768-5p mimics,respec-tively,and divided into NC group and miR-6768-5p group.The MTS assay and Matrigel invasion assay were used to detect the cell proliferation and invasion ability of each group,respectively.The putative binding sites of miR-6768-5p and CPA4 were verified using RNAhybrid software and dual-luciferase reporter gene experi-ment.The expression of CPA4 mRNA in each group of cells was detected by qPCR.The expression of AKT/c-MYC signaling pathway proteins in the cells of each group was analyzed by Western blot.Results Com-pared with the adjacent tissues,the relative expression level of miR-6768-5p in lung cancer tissues was signifi-cantly decreased,and the difference was statistically significant(P<0.05).Compared with HPAEpiC cells,the relative expression level of miR-6768-5p was significantly decreased in lung cancer cell lines,and the differ-ence was statistically significant(P<0.05).Compared with the NC group,the cell proliferation rate of miR-6768-5p group was significantly decreased(P<0.05).The number of invasive cells in NC group and miR-6768-5p group was(131.30±12.55)and(37.45±7.77),respectively,and the number of invasive cells in miR-6768-5p group was significantly lower than that in NC group(P<0.05).The relative expression level of CPA4 mRNA in H1299 cells of miR-6768-5p group was significantly lower than that in NC group(t=4.93,P<0.05).Compared with the NC group,the expressions of AKT/c-myC signaling pathway proteins p-AKT,p-mTORC1,XIAP,MDM2 and C-myC proteins in miR-6768-5p group were significantly decreased.Conclusion The expression of miR-6768-5p is decreased in lung cancer tissues,and miR-6768-5p may inhibit the activation of AKT/c-MYC signaling pathway by targeting CPA4,and reduce the proliferation and invasion ability of lung cancer H1299 cells.
8.Study on the mechanism of LncRNA AC132217.4 regulating the proliferation and invasion of liver cancer cells

Objective:To investigate the effect of lncRNA AC132217.4 on the proliferation and invasion of liver cancer MHCC97-H cells and its molecular mechanism.Methods:The TCGA database was used to analyze the differential expression of AC132217.4 in liver cancer tissue and adjacent tissue, and to analyze the relationship between the expression level of AC132217.4 and the overall survival of liver cancer patients. Transfection of pcDNA-AC132217.4 plasmid into MHCC97-H cells was defined as AC132217.4 group, transfection of pcDNA plasmid into MHCC97-H cells was defined as negative control (NC) group, respectively. The proliferation and invasion ability of MHCC97-H cells were detected by MTT method and Matrigel invasion assay. The binding site between AC132217.4 and miR-18a-5p was analyzed by starBase v2.0 software and dual luciferase reporter gene assay. Real-time quantitative PCR (RT-qPCR) detected the differential expression of miR-18a-5p in the two groups of MHCC97-H cells. The expression of epithelial-mesenchymal transition protein was detected by Western-blotting. Measurement data with normal distribution were expressed as mean±standard deviation ( ± s), and t-test was used for comparison between the two groups. Results:Compared with adjacent tissues, the expression of AC132217.4 was down-regulated in liver cancer tissues ( P<0.01). Compared with liver cancer patients with low expression of AC132217.4, the overall survival of liver cancer patients with high AC132217.4 expression was longer ( P<0.05). The pcDNA-AC132217.4 plasmid significantly inhibited the proliferation of MHCC97-H cells ( P<0.05). The number of invasive cells in the NC group and AC132217.4 group were (131.30±12.55) and (37.45±7.77), respectively. The pcDNA-AC132217.4 plasmid significantly inhibited the invasive ability of MHCC97-H cells ( t=6.36, P<0.01). AC132217.4 directly complemented miR-18a-5p ( P<0.01). The expression of miR-18a-5p in MHCC97-H cells in AC132217.4 group (1.04±0.30) was significantly lower than that in NC group (6.13±0.75) ( t=6.27, P<0.01). Compared with the NC group, the expressions of epithelial phenotype proteins Cytokeratin and Claudin-1 in MHCC97-H cells in AC132217.4 group were up-regulated, while the expressions of mesenchymal phenotype proteins Vimentin, Slug and Snail were down-regulated. Conclusions:The expression of AC132217.4 is low in liver cancer tissue, and it is related to the overall survival of liver cancer patients. AC132217.4 might inhibit the proliferation and invasion of liver cancer MHCC97-H cells by sponge miR-18a-5p.
9.Protective effect of compound drug Weng-Li-Tong on cisplatin-induced hepatocyte injury

【Objective】 To investigate the protective effect of the compound drug Weng-Li-Tong (WLT) against cisplatin (CDDP)-induced hepatocyte injury. 【Methods】 A cellular injury model was established by treating murine hepatocyte line BNL CL.2 with CDDP (80 μmol/L). Experimental groups were divided as follows: CDDP group (modeling only), WLT group (intervention with 1 g/L WLT), WLT+CDDP group (co-administration of CDDP and 1 g/L WLT), and a control group (normal culture). The protective effect of the compound drug WLT on CDDP-mediated hepatocyte injury was evaluated using CCK-8 assay, PI staining, crystal violet staining, Western blotting, reactive oxygen species (ROS) detection, and apoptosis analysis. 【Results】 Compared with the control group, the number of dead cells increased significantly (P<0.001) in the CDDP group, but no cytotoxicity was observed in the WLT group. The hepatocyte morphology in the WLT+CDDP group showed improvement with no obvious shrinkage compared to the CDDP group, as evidenced by the reduced proportion of PI-positive cells. Crystal violet staining results also indicated a higher cell count in the WLT+CDDP group than in the CDDP group, suggesting the protective effect of WLT against CDDP-mediated liver injury. Under CDDP intervention, the expression of the apoptosis-related protein Cleaved Caspase-3 increased. However, in the WLT+CDDP group, the expression of Cleaved Caspase-3 decreased, while the expression of the anti-apoptotic protein Bcl-2 increased. Additionally, compared to the CDDP group, the WLT+CDDP group showed a reduction in ROS production [DCFH-DA staining positive rate (%): 56.20±1.65 vs. 44.57±0.31] and a decrease in the proportion of apoptotic cells [proportion of early and late apoptotic cells (%): 43.60±0.44 vs. 19.57±0.78; 33.30±1.02 vs. 14.83±0.57] . 【Conclusion】 The compound drug WLT exhibits a protective effect against CDDP-mediated hepatocyte injury, suggesting potential therapeutic value in acute liver injury models.
10.Analysis on the relations between levels of change and the mental decisive factors on the physical exercise behavior among middle school students.

OBJECTIVETo study the situation of sports and its influencing factors among students in order to improve health related education and promotion programs.
METHODSA multistage random sampling method was used on 3 600 students from three cities to understand their physical exercise behavior. Both t and χ(2) test were used to measure the scale of psychology and to describe the time spent on exercise. Single variance factor was used to measure the levels of change on behavior of physical activities, psychological and physical exercise behaviors.
RESULTSThe weekly exercise time for students was 2.66 ± 1.801 days, with 81.4% of the students less than four days. Data showed that 37.3%, 23.6%, 20.5% of the high school students were in pre-contemplation stage, in contemplation stage, or in preparation stage respectively, with only 18.6 percent of the high school students in the action phase and maintaining phase. Students in the stages of change increase the amount of physical exercises. Scores with statistically significant differences were seen in the following areas: between five stages of behavior change on strategies (F = 77.442, P < 0.001), forward effects on the balance of decision-making (F = 29.498, P < 0.001), having negative effects (F = 14.784, P < 0.001)and self-efficacy (F = 135.544, P < 0.001). Changing strategy on the intention stage of front maintenance phase, positive effects on decision-making balance and self-efficacy scores were increasing along with the increasing stages of changing. The effect of balancing the negative effects increased when the change of phase decreased.
CONCLUSIONStudents were in lack of physical exercise per week. The differences related to the behavior on physical exercise did exist but most of the high school students were in the primary stage. Psychological factors played important role in the different stages of changing, suggesting that high school students should receive different health education and psychological intervention measures to enhance the effectiveness of physical exercise.
