1.UPLC Determination of Four Active Components in Ziziphi Spinosae Semen

This study was aimed to establish an analytical method to determine spinosin, jujuboside A, jujuboside B and betulinic acid content of Ziziphi Spinosae Semen quickly and accurately. The UPLC determination of Ziziphi Spinosae Semen method was established on an HSS T3 Column (2.1 mm × 100 mm, 1.8 μm) eluted with the mobile phase consisted of water containing 0.03% phosphoric acid and acetonitrile in gradient mode with the flow rate of 0.5 mL?min-1 and the detection wavelength was set at 204 nm. The standard curves of spinosin, jujuboside A, jujuboside B, betulinic acid showed a good linearity in 29.70 - 594.0 μg?mL-1, 10.68 - 213.6 μg?mL-1, 6.175 - 123.5 μg?mL-1, 22.00 - 440.0 μg?mL-1, respectively. The mean recoveries (n = 9) of the four components were between 98 . 3% and 99 . 4%, RSD < 1 . 4%. This method which is quick and accurate can be used to determine spinosin, jujuboside A, jujuboside B and betulinic acid in Ziziphi Spinosae Semen.
2.Effect of Shui Medicine Asarum insigne Polysaccharide on Glycolipid Metabolis,Renal Function and Oxi-dative Stress in Model Rats with Experimental Type 2 Diabetes

OBJECTIVE:To study the effect of Shui medicine Asarum insigne polysaccharide on glycolipid metabolism,renal function and oxidative stress in rats with experimental type 2 diabetes,and provide reference for its development and use. METH-ODS:60 SD rats were randomly divided into normal control group,model control group,positive control group (Irbesartan tab-lets,0.02 g/kg) and A. insigne polysaccharide low-dose,medium-dose,high-dose groups (calculated by crude drug as 2,4,8 g/kg),10 in each group. Except for normal control group,rats in other groups were intraperitoneally injected streptozotocin 75 mg/(kg·d)to induce model with diabetes. After modeling,rats in each administration group were intraperitoneally injected relevant medicines,and rats in normal control group and model control group were intragastrically administrated 5% carboxymet hylcellu-lose sodium solution,twice a day,for 42 d. After administration,UV spectrophotometer was used to detect the glycogen content in liver tissue. Automatic biochemical analyzer was used to detect 24 h urine output,24 h urinary protein content of rats,levels of tri-glyceride (TG),cholesterol (TC),low-density lipoprotein cholesterol (LDL-C),high-density lipoprotein cholesterol (HDL-C), creatinine (Cr),urea nitrogen (BUN) in serum,and levels of superoxide dismutase (SOD),glutathione peroxidase (GSH-Px), catalase(CAT),superoxide dismutase(ROS),malondialdehyde(MDA)in kidney tissue. RESULTS:Compared with normal con-trol group,glycogen content in liver tissue of rats in model control group was decreased(P<0.05);24 h urine output and 24 h uri-nary protein content were increased(P<0.05);levels of TG,TC,LDL-C in serum were increased(P<0.05),and HDL-C level was decreased (P<0.05);levels of SOD,CAT,GSH-Px in kidney tissue were decreased (P<0.05),and levels of ROS,MDA were increased(P<0.05). Compared with model group,except that there were no significant differences in the improvement of uri-nary protein content,HDL-C level in serum,24 h urine out-put in A. insigne polysaccharide low-dose group,above-men-tioned indexes in each administration group were obviously im-proved (P<0.05). CONCLUSIONS:A. insigne polysaccha-ride can regulate lipid metabolic disorders,and improve renal function and antioxidant capacity of rats.
3.Effect of controlled low central venous pressure with milrinone on laparoscopic hepatectomy

Objective:To evaluate the effect of controlled low central venous pressure with milrinone on laparoscopic hepatectomy in the patients.Methods:Fifty American Society of Anesthesiologists physical statusⅠ-Ⅲ patients of both sexes, aged 18-64 yr, with body mass index of 18-30 kg/m 2, of Child-Pugh grade A or B, undergoing elective laparoscopic hepatectomy, were divided into 2 groups ( n=25 each) using a random number table method: milrinone group (group M) and nitroglycerin group (group NG). After the start of surgery, milrinone 0.5 μg·kg -1·min -1 was continuously infused in group M, and nitroglycerin was continuously infused with the initial dose of 0.5 μg·kg -1·min -1 to maintain central venous pressure (CVP)≤5 mmHg in group NG.Mean arterial pressure and heart rate were recorded on admission to the operation room (T 0), at skin incision (T 1), at the beginning of liver resection (T 2), at completion of liver resection (T 3), at the end of operation (T 4), and CVP, cardiac index and stroke volume variation were recorded at T 1-4.Internal jugular vein blood samples were collected to determine the concentrations of hemogloblin, blood lactate at T 1 and T 4, and serum alanine aminotransferase, aspartate aminotransferase and creatinine concentrations at 1, 3 and 7 days after surgery.The score of blood oozing in hepatic surgical field, amount of norepinephrine used, blood loss, postoperative recovery and occurrence of complications within 7 days after operation were recorded. Results:Compared with group NG, cardiac index was significantly increased at T 2, 3, the CVP was decreased at T 2, the blood oozing score, blood loss, consumption of norepinephrine, and concentrations of blood lactate were decreased, and the postoperative drainage indwelling time was shortened in group M ( P<0.05). There was no significant difference in the serum alanine aminotransferase, aspartate aminotransferase and creatinine concentrations and incidence of postoperative complications at 1, 3 and 7 days after operation between the two groups ( P>0.05). Conclusions:Milrinone is better than nitroglycerin in decreasing central venous pressure, reducing blood loss, maintaining stable circulatory function and tissue perfusion in laparoscopic hepatectomy.
