1.Detection of cell apoptosis induced by 125I brachytherapy in nude mice bearing lung adenocarcinoma using 99Tcm-Annexin V imaging combined with diffusion weighted MRI

Objective To detect apoptosis of transplanted lung adenocarcinoma cells in nude mice after 125I brachytherapy by 99Tcm-Annexin V combined with diffusion weighted MRI (MR-DWI).Methods Twenty-five BALB/c-nu nude mice models subcutaneously transplanted with A549 cells were divided into experimental group (EG,n=13) and control group (CG,n=12) by random number table method.One 125I seed with apparent activity of (24.8±6.3) MBq was implanted into each mouse in EG,while CG underwent cold seed implantation.Both of 99Tcm-Annexin V imaging and MR-DWI were performed within 7-10 d after brachytherapy,then all mice were sacrificed and tumor cell apoptosis was detected by terminal oxynucleotidyl transferase mediated dUTP biotin nick end labeling (TUNEL) immunofluorescence,Survivin expression was assayed by SP.Two-sample t test,x2 test and Pearson correlation analysis were used for data analysis.Results Positive rate of cell apoptosis by 99Tcm-Annexin V imaging was 69.2%(9/13) and 8.3%(1/12) respectively in EG and CG (x2 =12.73,P<0.01).The uptake ratio of 99Tcm-Annexin V,apparent diffusion coefficient(ADC) value and apoptosis index(M) of the tumor in EG were 2.91±0.85,(2.03±0.44)×10-3 mm2/s and (49± 18) %,respectively,which were significantly higher than those of CG (1.26 ± 0.37,(1.29 ± 0.21) ×10-3 mm2/s and (11±4)% respectively,t=5.930,5.452,7.606,all P<0.05).Survivin expression in EG and CG was (46± 13) % and (15±7) % respectively (t =5.158,P<0.05).The value of ADC was correlated with AI and uptake ratio(r=0.756,0.788,both P<0.05).Uptake ratio was correlated with AI (r=0.754,P<0.05),while Survivin expression was negatively correlated with AI (r =-0.772,P<0.05).Conclusions Down-regulation of Survivin expression may play an important role in apoptosis induced by 125I brachytherapy.99Tcm-Annexin V combined with MR-DWI could effectively evaluate apoptosis of lung adenocarcinoma cells in a non-invasive way,thus it might be helpful in evaluation of early efficacy of 125I brachytherapy.
2.Assessment of reliability and validity of assessment of multiple systematic reviews in Chinese systematic reviews on stomatology.

OBJECTIVETo measure the agreement, reliability and validity of assessment of multiple systematic reviews (AMSTAR) to assess Chinese systematic reviews on stomatology.
METHODSA comprehensive electronic search was made through Chinese BioMedical Literature Database, VIP Database for Chinese Technical Periodicals and China National Knowledge Infrastructure electronically on March 1st 2011 together with handsearch through 19 stomatological journals to identify published Chinese systematic reviews on stomatology. Each systematic review was assessed by two reviewers with overview quality assessment questionnaire (OQAQ) and AMSTAR. And reliability (interobserver Kappa of the 11 AMSTAR items), interclass correlation coefficient (ICC) of the sum scores and construct validity (ICC of the sum scores of AMSTAR compared with those of the OQAQ) were reported.
RESULTSA total of 52 systematic reviews on stomatology were eligible. The reviewers agreement of the individual items of AMSTAR was with a mean Kappa of 0.81 [95% CI(0.73, 0.89)] while the OQAQ was 0.74 [95% CI (0.66, 0.83)]. The ICC of the total score for AMSTAR was 0.98 [95% CI (0.97, 0.99), P = 0.000]. Cronbach' alpha was 0.69 [95% CI (0.56, 0.80), P = 0.000]. And ICC of the sum scores of AMSTAR compared with those of the OQAQ was 0.94 [95% CI (0.90, 0.97), P = 0.000].
CONCLUSIONAMSTAR has good agreement, reliability and validity. AMSTAR can be well used in Chinese stomatology and can bring dentists much convenience when assess the methodological quality of systematical reviews on stomatology.

3.Relationship between expression levels of TTF-1 and Galectin-3 in differentiated thyroid carcinoma tissues and clinical manifestations and prognosis of patients

Objective To investigate the relationship between the expression levels of thyroid transcription factor-1(TTF-1)and Galectin-3 in differentiated thyroid carcinoma(DTC)tissues and clinical manifestations and prognosis of patients.Methods A total of 76 DTC patients admitted to the hospital from January 1,2017 to May 30,2020 were selected as the study objects.Cancer tissue specimens obtained during surgery were in-cluded in the DTC group(n=76),and corresponding paracancer tissue specimens were included in the para-cancer group(n=76).The expressions of TTF-1 and Galectin-3 in DTC group and paracancer group were de-tected by immunohistochemistry,and the relationship between the expression levels of TTF-1 and Galectin-3 and the clinicopathological characteristics of DTC patients was analyzed.Multivariate Cox regression analysis was used to investigate the prognostic factors of DTC patients.Results The positive expression rates of TTF-1 and Galectin-3 in DTC group were higher than those in paracancer group,and the difference was statistically significant(P<0.05).The TTF-1 positive expression rate and Galectin-3 positive expression rate in DTC pa-tients with TNM stage Ⅲ to Ⅳ,low differentiation,tissue type of papillary thyroid carcinoma and lymph node metastasis were higher than those in DTC patients with TNM stage Ⅰ to Ⅱ,medium/high differentiation,tis-sue type of thyroid follicular carcinoma and no lymph node metastasis.The difference was statistically signifi-cant(P<0.05).The 3-year overall survival rate of TTF-1 negative and Galectin-3 negative DTC patients was higher than that of TTF-1 positive and Galectin-3 positive DTC patients,and the difference was statistically significant(P<0.05).Multivariate Cox regression analysis showed that lymph node metastasis,positive TTF-1 and positive Galectin-3 were prognostic factors in DTC patients(P<0.05).Conclusion TTF-1 and Galectin-3 are related to TNM stage,differentiation degree,tissue type,lymph node metastasis and 3-year sur-vival rate of DTC patients,and have important reference value for the diagnosis and prognosis evaluation of DTC patients.
