1.Effect of gemcitabine combined with cisplatin on ER, MMP-9 and estrogen in patients with stage III breast cancer

Objective To explore the effect of gemcitabine combined with cisplatin on ER , MMP-9 and estrogen in patients with stage III breast cancer.Methods 93 patients with breast cancer were collected and randomly divided into control group and experimental group.Two groups were all under breast cancer surgery, control group was treated with cisplatin chemotherapy,1st day of intravenous infusion of cisplatin 90 mg/m2 , 21 days for a course of treatment, continuous treatment of 6 courses.Experimental group were treated with cisplatin combined with gemcitabine chemotherapy , 1st and 8th days of intravenous infusion of 1000 mg/m2 gemcitabine, 3 ~5d, intravenous drip of cisplatin 90 mg/m2 , every 28 days a cycle, continuous administration of 2 cycles,After treatment, the clinical efficacy of the two groups, the level of ER, MMP-9, estrogen and the incidence of adverse reactions were compared.Results Compared with before treatment,the levels of serum MMP-9,E1, E2, ER were decreased (P<0.05), and the level of LH and FSH were increased (P<0.05) of two group post-treatment.Compared with control group post-treatment, the levels of serum ER, MMP-9, E1, E2 were lower in experimental group (P<0.05), and the LH and FSH levels were higher (P <0.05).The incidence of adverse reactions of experimental group was 6.38%, and control group was 15.22%, the incidence of adverse reactions in the two groups was no significant difference . Conclusion Gemcitabine combined with cisplatin in the treatment of patients with breast cancer has a significant effect, and can improve the clinical symptoms, reduce the serum ER, E1, MMP-9, E2 levels, improve FSH, LH level.
2.Expression of VEGF and its correlation with clinicopathological factors of cervical carcinoma ABSTRACT

Objective This study was designed to examine the expression of VEGF and its correlation with clinicopathological factors of cervical carcinoma. Methods mmunohistochemistry S-P method was used to detect the expression of VEGF in 10 cases of nomal cevical epithelium( NCE), 18 cases of cervical intraepithelial neoplasm (CIN) and 77 cases of early invasive carcinoma of cervix (ICC). Results The positive rate of VEGF protein in NCE,CIN,ICC was 10% (1/10) ,44.4% (8/18) ,74. 0% (57/77), respectively. The defference was significant (P <0. 05). Expresson of VEGF was not related with tumor stage, histologic types and stroma infiltration, but related with cell differentiation and intravascular involvement、lymph node metastasis. Conclusion VEGF protein had a positive correlation with cell differentiation and intravascular involvement 、 lymph node metastasis. the expression of VEGF gene result in the occurrence and development of cervical carcinoma by inducing the angiogenesis.
3.Unnecessity of Integrating Metabolic Indices into the Diagnosis of Polycystic Ovary Syndrome

Objective To discuss whether it is necessary to integrate metabolic indices into diagnosis of polycystic ovary syndrome (PCOS).Methods Taking ESHRE/ASRM diagnosis as the gold standard,51 women with PCOS and 47 women without PCOS were selected and divided into the intervention group and control group respectively from the Department of Gynecology in the Second Affiliated Hospital of Kunming Medical University between January 2013 and July 2014.Logistic regression based on principal component analysis and significant variables chosen through single factor analysis were used to establish the new diagnostic model which combined reproductive indices and metabolic indices.We evaluated the validity and reliability of the new diagnostic model by using ROC curve analysis.Finally,we analyzed the consistence and difference between the new diagnostic model and the gold standard.Results Thirteen significant variables were chosen using single factor analysis.ROC analysis showed that an area under the curve was 0.976 (P<0.001) and the optimal cut-off point was 0.526 with a sensitivity of 96.08%,a specificity of 93.62% and a consistency of93.88%.The new diagnostic model had superior validity and reliability.The two diagnostic methods had strong consistence (Kappa=0.877,P<0.001) and no difference (x2=0.167,P=0.688).Conclusion Considering that the integration of metabolic indices does not change the diagnosis result,we come to a conclusion that it is unnecessary to integrate metabolic indices into diagnosis of PCOS.
4.Anti-tumor effect and its mechanism of co-administration of fusion proteins hVEGF121/βhCG and mGM-CSF/βhCG

This study aimed at investigating the inhibitory effects and the anti-tumor mechanisms of co-adminis-tration of fusion proteins mGM-CSF/βhCG ( GC ) and hVEGF121/βhCG ( VC ) on RM-1 prostatic cancer and B16 F10 melanoma in the C57 BL/6 J mouse model. Two recombinant stains containing pET-28 a-mGM-CSF-X10-βhCGCTP37 and pET-28 a-VEGF-M2-X10-βhCG-CTP37 were induced by lactose to express fusion proteins. The fusion proteins were separated and purified to prepare the anti-tumor protein vaccines ( VC protein vaccine and GC protein vaccine) , which were then mixed to prepare a combined protein vaccine named VGC protein vac-cine. The prostatic cancer and melanoma tumor-bearing mice C57 BL/6 J were immunized with described vac-cines, then the growth of each tumor was measured;splenocyte proliferation of immunized mice was detected;and the cytotoxic effects of the vaccine on tumor cells were tested. After that, the in vivo concentrations of IFN-γ and anti-hVEGF antibodies were investigated by ELISA. The difference between each experimental group and normal saline group ( NS) was statistically significant in both tumor-bearing mouse models ( P <0. 05) respectively. Besides, VGC group exhibited significantly better anti-tumor effect compared with the GC and VC groups with the anti-tumor rate ( 41. 7 ± 0. 83)% and ( 46. 4 ± 1. 27)% for prostatic cancer and melanoma tumor, respectively. The co-administration of the two proteins, VC and GC, could inhibit the growth of RM-1 prostatic tumor and B16F10 melanoma effectively via anti-tumor immunity and anti-tumor angiogenesis.
5.Abnormal distribution of laminin α2 chain in glomerular basement membrane in membranous nephropathy

To study the abnormal distribution of type Ⅳ collagen and laminin chains in glomerular basement membrane (GBM) in membranous nephropathy (MN).Methods 52 cases of MN were collected and staged according to electron microscopic morphological characteristics,and 10 cases of kidney tissues of minimal change disease were used as normal GBM control.Distribution pattern of or5 (Ⅳ) chain,laminin α5and β2 chains,and laminin α2 and β1 chains were detected using immunofluorescence method.Results In minimal change disease,α5 (Ⅳ) chain,laminin α5 and β2 chains all showed continuously linear positive expression along GBM,and laminin α2 and β1 chains were negatively expressed in GBM.In stage Ⅰ MN,α5 (Ⅳ) chain,laminin α.5 and β2 chains all showed continuous linear positive expression along GBM.In stage ⅡMN,the expression of α5 (Ⅳ) chain was increased and showed abundant spikes on the basis of continuous linear positive staining along GBM,and the expression of laminin α5 and β2chains was increased,and segmental spikes were seen on the basis of continuous linear positive staining along GBM.In stage ⅢMN,the expression of α5 (Ⅳ),laminin α5 and β2 chains was also enhanced and segmental double tracks were seen.The expression of laminin α2 chain was negative in GBM in stage ⅠMN,but granular positive expression along GBM was seen in stage Ⅱ and stage Ⅲ MN.No positive expression of laminin β1chain was seen in GBM in different stages in MN.Conclusion The GBM thickness in MN originates not only from intrinsic type Ⅳ collagen chains and laminin chains,but also from laminin α2chain,which only exist in glomerulus mesangium in normal condition.
6.Application progress of fullerene and its derivatives in medicine

Fullerene is an effective free radical scavenger and antioxidant. The fullerene derivatives obtained by chemical modification of fullerene have good water solubility and biological activities. Fullerene and its derivatives have many advantages in cell protection and antioxidant properties, antibacterial activity, antiviral activity, photodynamic activity, drug delivery and anti-tumor activities, playing an important role in the field of medicine. In recent years, great progress has been made in this field. In this review, we summarized the latest research progress and applications of fullerene and its derivatives in medicine field at home and abroad from four aspects of regulating tumor microenvironment, drug delivery, photodynamic therapy and anti-oxidative stress. At last, the future development and application of fullerene and its derivatives in the domain of medicine are prospected.

7.Association of human epicardial adipose tissue volume and inflammatory mediators with atherosclerosis and vulnerable coronary atherosclerotic plaque.

OBJECTIVETo investigate the relation of epicardial adipose tissue volume (EATV) determined by dual-source CT (DSCT) cardiac angiography and EAT-derived inflammatory factors to coronary heart disease (CHD) and vulnerable plaque.
METHODSA total of 260 patients underwent cardiac computed tomography to evaluate stenosis of coronary artery, and blood samples were obtained from each patient. CHD was confirmed in 180 patients by DSA and CHD was excluded in the remaining 80 patients (NCHD). Vascular remodeling index and plaque vulnerability parameters (fatty volume, fibrous volume and calcification volume and fiber volume) were measured in CHD patients and correlation with EATV was analyzed. Epicardial adipose tissue (EAT) and intrathoracic adipose tissue (TAT) were collected from 40 CHD patients undergoing CABG surgery, and, mRNA and protein expressions of leptin and MMP9 were detected by RT-PCR and Western blot analysis.
RESULTS(1) The EATV was significantly higher in the CHD group than in NCHD group ((121.2 ± 40.6) mm³ vs. (74.7 ± 18.1) mm³, P = 0.01). (2) Subgroup analysis of the CHD patients demonstrated that EATV was significantly higher in patients with positive remodeling than in patients without positive remodeling ((97.6 ± 42.0) cm³ vs. (75.5 ± 25.4) cm³, P = 0.01). Lipid plaque volume was positively correlated with EATV (r = 0.34, P = 0.002); however, fiber plaque volume was negatively correlated with EATV (r = -0.30, P = 0.008). (3) Logistic regression analysis indicated that EATV was an independent risk factor for positive vascular remodeling (OR = 2.01, 95% CI: 1.30-2.32, P = 0.01). (4) mRNA and protein expression of leptin and MMP9 in EAT was significantly upregulated in 40 CHD patients who received CABG surgery compared to 40 NCHD patients (P < 0.01). However, there was no significant difference (P > 0.05) in mRNA and protein expression of leptin and MMP9 from the SAT between CHD and NCHD patients. (5) In the CHD group, leptin and MMP9 levels in EAT and EATV were positively correlated with lipid plaque volume and fibrous plaque volume (P < 0.05).
CONCLUSIONSEATV is an independent risk factors of coronary heart disease and plaque vulnerability; EAT secretion of inflammatory cytokines from CHD patients is significant increased compared to NCHD patients, EAT secretion of inflammatory cytokines are positively correlated with EATV, both of which are determinants affecting vascular remodeling. Reducing EATV might help to attenuate inflammation and plaque vulnerability and reduce the risk of coronary heart disease.

8.Study on Clinical Term Semantic Relationship of Traditional Chinese Medicine Based on Clustering

The construction of clinical ontology of traditional Chinese medicine (TCM) is one of the important components of TCM internationalization.Among them,the study of clinical term entity has been quite successful.But the research on semantic relation is still lacking.This paper presented a method based on the combination of clustering and syntax pattern to study the semantic relations between TCM conceptual entities.By extracting the feature words around the entity,K-means was used as the clustering algorithm to perform the first round of clustering for all corpora.Based on results of the first round of clustering,the longest common subsequence was extracted in the same cluster and generalized as syntax pattern.According to the sentence after manual adjustment,it was automatically judged that each sentence in the corpus has the most suitable syntax pattern of semantic relations,and the second round of clustering is characterized by the syntax pattern.The result was the final clustering result.The experimental results showed that the accuracy of this method was 88.23% for the classification of semantic relations in corpus.
9. Analysis on the incidence of occupational pneumoconiosis of migrant workers in Shiyan city from 2013 to 2017

Objective:
To explore the incidence regularity and characteristics of pneumoconiosis among migrant workers in Shiyan city, and to provide a basis for making policies about pneumoconiosis prevention and treatment among migrant workers.
Methods:
In September 2018, 338 cases of pneumoconiosis among migrant workers and 354 cases of pneumoconiosis among non-migrant workers were collected between 2013 and 2017. The proportions of pneumoconiosis types, the age and seniority of onset workers, the severity grade of pneumoconiosis, the type of pneumoconiosis, the type of work and regional distribution, etc. were analyzed.

10.Application and anaylsis of discussion class in the teaching of Cellular and Molecular Basis of Medicine for undergraduates

Cellular and Molecular Basis of Medicine is a basic module course newly established by our school that is compulsory for medical students. This module course overthrows traditional disciplinary boundaries, and adopts the forms of combining classroom teaching with students' self-learning and teachers' instruction with topic discussion of students guided by teachers. In the teaching of the module course, discussion was adopted among students which involves teachers giving out the range of topics to students so that they can prepare for the topic of their intertest. After group discussion during class, the head of the teaching group gave comment on students' discussion and prize for whom had the best performance. Then, students were asked to summarize and analyze the questions raised during discussion after class. In conclusion, discussion class has stimulated students' interest in learning, improved their ability to think independently and encouraged them to demonstrate their abilities, which has achieved good teaching outcomes.
