1.Development and application of medical equipment maintenance management module

Objective To realize fine management of medical equipment maintenance,effectively improve the efficiency and save the cost of equipment maintenance.Methods A medical equipment maintenance management module was developed with hospital resource planning system and SQL Server 2008 platform so as to implement informatized management for equipment acceptance check,maintenance,processing and etc.Results The module realized fine management,controlled cost and enhanced social and economic benefits of the hospital.Conclusion The module contributes to medical equipment maintenance management informatization,and is of great significance for the improvement of the hospital.
2.The efficacy of the cicatrisation-fumigant Chinese herbal on the postoperative complications of the mixed hemorrhoids

Objective To observe the efficacy of Chinese herbal fumigant sitz bath for different exposure durations on patients for the complicatiors after mixed hemorrhoidectomy.Methods One hundred and twenty patients with mixed hemorrhoidectomy were randomized into four groups(2 treatment groups and 2 control groups) with 30 cases in each group.The treatment groups were treated with the Chinese Herbal fumigant sitz bath for 10 minutes and 15 minutes respectively;while the control groups were treated with the hemorrhoids lotion for 10 minutes and 15 minutes respectively.The severity of pain,edema,bleeding,constipation and urinary disturbance were scored on the second,seventh and fourteenth days after treatment.Data were analyzed statistically.Results Chinese herbal fumigant Sitz baths for 10 minutes could significantly reduce postoperative pain,edema and bleeding symptoms[(1.72±1.23) points,(0.72±1.25) points,(0.91±1.37) points,respectively,P <0.05],followed sequentially by groups of Chinese herbal fumigant Sitz baths for 15 minutes [(1.79±1.21)points,(0.89±1.28) points,(1.03±1.24) points]; The hemorrhoids lotion for 10 minutes ((1.86±1.25) points,(1.14±1.47) points,(1.49±1.56)) and 15 minutes[(2.03±1.48) points,(1.05±1.29) points,(1.39±1.47) points,respectively,P < 0.05]With the successful cases of 27,21,18,and 6 in the groups,respectively.Though the overall response rates of for the groups of 10 min's treatment,15min's treatment and 10 min's control were not significantly different between each other(P =0.0634,P =0.2560),they showed a superior efficacy in the overall response rate to the 15 min's control group(P =0.0064,P =0.0365,P =0.0089),respectively.Conclusion Chinese herbal fumigant Sitz baths for 10minutes have a success rate of 100% on alleviating complications after mixed hemorrhoidectomy.It has a prominent efficacy on reducing postoperative edema and bleeding symptoms than other treatments studied in this program.
3.Effects of pretreatment with dimethyloxalylglycine on the survival of multi-territory perforator flap in rat and related mechanism

Objective To observe the effects of pretreatment with dimethyloxalylglycine (DMOG) on the survival of multi-territory perforator flap and the vessels of choke zone (CZ) 2 in rat,and to explore related mechanism.Methods Sixty adult SD rats were divided into group DMOG and normal saline group (NS) according to the random number table,with 30 rats in each group.Perforator flap with three angiosomes was made on the right dorsal side of rat,including deep iliac circumflex artery perforator,intercostal artery perforator,thoracodorsal artery perforator,as well as CZ 1 and CZ 2.Rats in group DMOG were intraperitoneally injected with 2 mL NS containing DMOG (40 mg/kg) 2 days before operation,2 hours before operation,and 2 days after operation.Rats in group NS were intraperitoneally injected with equivalent volume of NS at the same time point.On post operation day (POD) 7,gross observation was conducted,and the survival rate of flap was calculated.On POD 7,the vascularity in CZ 2 and potential zone of flap was observed using angiography.On POD 7,new vessel in CZ 2 of flap was observed with HE staining,and the microvessel density (MVD) was calculated.On POD 7,the expression of vascular endothelial growth factor (VEGF) in CZ 2 of flap was detected by immunohistochemistry and Western blotting (respectively denoted as integral absorbance values and ratio of gray value),and blood flow volume of vessel in CZ 2 of flap was examined by laser Doppler perfusion imager.The sample number of each index was 6 in each group.Data were processed with t test.Results (1) On POD 7,rats in two groups all survived,and the flaps were not infected.In group DMOG,the necrotic area of flaps of rats with dark yellow crust and soft texture was observed approximately at the distal end of skin entry point of thoracodorsal artery perforator.In group NS,the necrotic area of flaps of rats with brownish black crust and hard texture was observed approximately at the distal end of CZ 2.The survival rate of flap of rats in group DMOG was (88 ± 3) %,which was significantly higher than that in group NS [(82 ± 3) %,t =3.38,P < 0.01].(2) On POD 7,there were clear vascular structure and many new vessels in CZ 2 of flaps of rats in group DMOG,with intact vascular structure in potential zone.On POD 7,there were unclear vascular structure and few new vessels in CZ 2 of flaps of rats in group NS,with disorder vascular structure in potential zone.(3) On POD 7,MVD in CZ 2 of flaps in rats of group DMOG was (29.2 ± 2.2)/mm2,which was significantly higher than that of group NS [(20.3 ± 3.6)/mm2,t =5.10,P <0.01].(4) On POD 7,the expressions of VEGF in CZ2 of flaps in rats of group DMOG detected by immunohistochemistry and Western blotting were 5 060 ± 432 and 0.48 ± 0.04 respectively,which were significantly higher than those of group NS (2 811 ± 382 and 0.26 ± 0.06,with t values respectively 9.54 and 5.67,P values below 0.01).(5) On POD 7,blood flow volume of vessel in CZ 2 of flaps in rats of group DMOG was (58 ±4) perfusion units (PU),which was significantly more than that of group NS [(46 ± 4) PU,t =5.20,P < 0.01].Conclusions DMOG can increase the survival rate of multi-territory perforator flap through promoting angiogenesis in CZ 2 of flap on the back of rat and improving blood supply of flap.
4.Improvement of memory in healthy elderly subjects by Shentai Tea Polyphenols

Objective:To investigate the effect of Shentai Tea Polyphenols on memory and its underlying mechanism in healthy elderly adults.Methods:According to the randomized, double blind and placebo-controlled prospective experimental design, 240 healthy middle-aged or elderly subjects with normal objective cognition were recruited in our hospital from April to December 2019 through advertising, and they were randomly divided into an experimental group ( n=120) and a control group ( n=120). Oral administration of Shentai Tea Polyphenols or placebo capsules (lasting for 90 d, 2 times/d, 2 capsules/time) was given to subjects in the two groups. The neuropsychological scale scores and near-infrared brain imaging data of all subjects were collected before and after intervention, and the mean time from baseline data collection before intervention to follow-up data collection after medication was controlled within 6 months. Results:In patients from the experimental group, as compared with those before intervention, scores of delayed recall and recognition memory in the auditory verbal learning test (Huashan version) and Boston naming test, scores of Montreal cognitive assessment scale (basis version) were significantly higher, and scores of Hamilton anxiety scale, shape trails test A and rapid-eye-movement sleep behavior disorder screening questionnaire were significantly lower after intervention ( P<0.05); however, only scores of delayed recall in the auditory verbal learning test (Huashan version) were significantly increased in the control group ( P<0.05). A total of 23 pairs of differences of brain function connection before and after intervention in the experimental group were significantly higher than those in the control group ( P<0.05), mainly involving the function connection of forehead peak network (FPN), and function connection of FPN-default network, function connection of FPN-somatosensory movement network, function connection of FPN-dorsal attention network and function connection of FPN-visual network, as well as the function connection of default network and somatosensory movement network. Conclusion:Shentai Tea Polyphenols can improve cognitive performances including memory, language and executive function, anxiety mood and sleep quality in healthy middle aged or elderly subjects by affecting the functional connections of the networks in the brain.
5.Expression and characterization of a keratinase encoding gene gm2886 in Streptomyces pactum ACT12 strain.

By bioinformatics analysis, a putative keratinase gene gm2886 (Accession number: KY368946) was discovered in the genome of a feather-degrading strain, Streptomyces albidoflavus Fea-10. gm2886 was ligated into integrative Escherichia coli-Streptomyces shuttle vector pSET152 under the promoter PermE and added with C-terminal His-tag. The expression vector was transformed into Streptomyces pactum ACT12 by conjugal transfer and the recombinant protein GM2886-His6 was detected in fermentation broth. GM2886-His6 was purified and characterized. Its size was nearly 36 kDa. GM2886-His6 showed proteolytic activity towards a variety of substrates and could even degrade insoluble substrates, such as azure keratin and chicken feathers. The optimal pH and temperature of GM2886-His6 for proteolysis of casein was pH 10.0 and 50 ℃, respectively. The enzyme activity was inhibited by PMSF, but not EDTA, indicating that GM2886-His6 was a serine proteinase. Our results laid the foundation for the research of the molecular biological mechanism on feather-degrading and for the further utilization of Fea-10.
