1.Advances in antidiabetic small molecule ABHD6 inhibitors

Type 2 diabetes,a glucose and lipid metabolism disorder accompanied by chronic multiple organ damage,has become a huge threat to human health,α/β hydrolase domain-6 (ABHD6) regulates the insulin release negatively by hydrolyzing monoacylglycerol.Small molecule ABHD6 inhibitors have been proven to lower bloodglucose and regulates energy homeostasis,which is a potential candidate for the treatment of type 2 diabetes.This paper introduced the ABHD6 signaling pathway and its mechanism,then reviewed the progress of small molecule ABHD6 inhibitors with different structures in recent years,and analyzed the structure activity relationship.
2.Establishment of a Stable Cell Line Expressing"Toxic" Transient Receptor Potential A1 Channel

Transient receptor potential A1 (TRPAl) is a cold sensitive cation channel, which could also be activated by various pungent compounds. As a transduction channel in a number of sensory modalities, TRPAl expressing in heterogonous systems serves to provide great convenience in pharmacological analysis and functional investigation. Due to cellular toxicity, establishment of stable TRPAl cell line has always been challenging. Nevertheless, the first stable human embryonic kidney (HEK-293) cell line with un-controlled expression of TRPAl was successfully established. It was also confirmed that this stable cell line retained TRPAl expression for more than 25 passages in culture. The functional analysis of the cell response verified the stability and specificity of this novel recombinant TRPAl cell line. Altogether, the data indicated this TRPAl-HEK cell line would be a useful tool for functional analysis of TRPAl and for the development of high throughput screening (HTS) compatible assay in the effort to identify TRPAl modulators.
3.Practice of Syndrome Differentiation of Eight Principles Theory in TCM Medication Consultation Services

OBJECTIVE:To provide reference for the clinical pharmacists to apply syndrome differentiation of eight principles theory for TCM consultation services. METHODS:Several examples were given to illustrate the practice of the theory guiding patients'medication consultation in respect of Chinese herbal medicine differential medication,Chinese patent medicine differential medication,dietary taboo,etc. According to the author's many years of work experience,from the three aspects of the counselor, the consultant,and the consulting conditions,the reasons that TCM consultation window couldn't provide the medical staff and patients with rational TCM use suggestions and appropriate diet were analyzed. RESULTS & CONCLUSIONS:The syndrome differentiation of eight principles theory is based on four diagnostic methods to collect and analyze symptoms,signs and other information comprehensively. The information were divided into eight types of syndromes as yin and yang,exterior and interior, cold and heat,deficiency and excess,so as to determine the theory of basic syndromes of disease. The suggestions on rational drug use and appropriate diet were provided for the patients by using syndrome differentiation of eight principles theory. In view of the lack of knowledge and ability of the clinical pharmacists on syndrome differentiation of eight principles theory,the unfamiliar service scope of the clinical pharmacists in TCM consultation service,and the lack of consultation conditions for the use of TCM, it is suggested that the clinical pharmacists should learn more about the syndrome differentiation of eight principles theory;TV, WeChat and other media guide public dialect use of Chinese herbal medicines,Chinese patent medicines and appropriate diet;hardware conditions for TCM consultation by establishing TCM consultation rooms and TCM consultation clinics are establish so as to improve the level of clinical pharmacists'TCM consultation services.
4.Development trajectory and influencing factors of pediatric medical traumatic stress in parents with children undergoing surgery: a latent growth mixture model

Objective:To explore the development trajectory and influencing factors of pediatric medical traumatic stress in parents of children undergoing surgery, so as to provide reference for targeted intervention measures in the later stage.Methods:Adopting prospective research design, from January 2022 to August 2022, convenience sampling was used to recruit 199 parents of children undergoing surgery in the pediatric surgery ward of the First Affiliated Hospital of University of Science and Technology of China as subjects. The level of medical traumatic stress symptoms in parents was investigated by the PTSD Checklist for DSM-5 at one day before surgery, during postoperative recovery, after discharge and 3 months after discharge. The general data questionnaire, Parents′ Perception of Uncertainty Scale, Hospital Anxiety and Depression Scale, Simplified Coping Style Questionnaire and Perceived Social Support Scale were used to investigate the influencing factors of medical traumatic stress symptoms in parents one day before the operation.Latent growth mixture model and binary Logistic regression were used to analyze the data.Results:The development trajectory of pediatric medical traumatic stress in parents of children undergoing surgery was divided into two categories: the first was low-level decline (90.5%, 180/199) and the second was high-level stability (9.5%, 19/199). Binary Logistic regression analysis showed that postoperative complications, surgical type and negative coping styles of parents were the main influencing factors of pediatric medical traumatic stress trajectory in parents of surgical children(all P<0.05). Conclusions:The pediatric medical traumatic stress in parents of children undergoing surgery have group heterogeneity trajectory, medical staff can formulate targeted and individualized intervention programs according to their trajectories and influencing factors.
5.The impact on the delivery quality of the midwife accompanied

Objective To explore and analyze the impact on the delivery quality of the midwife accompanied. Methods One hundred and twenty puerperal from November 2012 to November 2013 in the thematernity department of our hos-pital were selected as the research objects,they were randomly divided into the experiment group and the control group,the control group were given the normal maternal care personnel shift system, while the experiment group ma-ternal were given the one-to-one accompanied by midwives , assisting in childbirth puerperal. The women complica-tions and two modes of nursing satisfaction of two groups were compared. Results The pain, bleeding, abnormal labor, fetal asphyxia probability and negative emotions in the experiment group were significantly lower than that of the con-trol group, while the maternal satisfaction rate of the experiment group was significantly higher than that of the control group(P<0.05). Conclusion Implements of the one-to-one for maternal the whole nursing model can effectively avoid the maternal complications and more beneficial to the health of the fetus birth and improve maternal satisfaction ,it is beneficial to the improvement of the overall quality of care for maternal and infant and worty of proper clinical promo-tion.
6.Effect of attachment on coping styles in patients with advanced lung cancer: the chain-mediated role of disease perception and hope

Objective:To explore the mediating chain effect between attachment and coping style of disease perception and hope in patients with advanced lung cancer, and to provide theoretical basis for improving coping style in patients with advanced lung cancer.Methods:From October 2021 to June 2022, 354 patients with advanced lung cancer in the First and Second Affiliated Hospitals of Anhui Medical University were selected by convenience sampling. The general information questionnaire, the Experiences in Close Relationships Inventory, the Brief Illness Perception Questionnaire, the Herth Hope Index, and the Medical Coping Modes Questionnaire were used to conduct cross-sectional questionnaire survey. SPSS 25.0 software and Bootstrap method were used to construct and verify the chain mediation model.Results:Finally, 336 patients with advanced lung cancer were included, including 214 males and 122 females, aged 27-79(59.43 ± 8.61) years old. Attachment avoidance score was (3.31 ± 1.01) points, attachment anxiety score was (3.86 ± 1.17) points, illness perception score was (40.07 ± 12.01) points, hope score was (34.05 ± 5.87) points, and face coping score was (18.75 ± 5.34) points in patients with advanced lung cancer. The avoidance coping score was (15.47 ± 1.97) points, and the yielding coping score was (9.62 ± 3.85) points. In patients with advanced lung cancer, attachment avoidance and attachment anxiety were positively correlated with yield coping ( r=0.448, 0.747, both P<0.01), positively correlated with illness perception ( r=0.356, 0.627, both P<0.01), and negatively correlated with hope ( r=-0.406, -0.670, both P<0.01). Illness perception was positively correlated with yield coping ( r=0.744, P<0.01), and negatively correlated with hope ( r=-0.628, P<0.01). Hope was negatively correlated with yield response ( r=-0.769, P<0.01). The mediation model showed that the chain mediating effect of attachment avoidance, illness perception, hope and yield coping was significant in patients with advanced lung cancer, with an effect value of 0.009 and an effect size of 13.95%. The chain mediating effect of attachment anxiety, illness perception, hope and yield coping were significant, with an effect value of 0.010 and an effect size of 8.27%. Conclusions:Attachment can not only directly predict submission coping in advanced lung cancer patients, but also indirectly predict submission coping through the chain mediation of illness perception and hope.
7.Expression and characterization of a keratinase encoding gene gm2886 in Streptomyces pactum ACT12 strain.

By bioinformatics analysis, a putative keratinase gene gm2886 (Accession number: KY368946) was discovered in the genome of a feather-degrading strain, Streptomyces albidoflavus Fea-10. gm2886 was ligated into integrative Escherichia coli-Streptomyces shuttle vector pSET152 under the promoter PermE and added with C-terminal His-tag. The expression vector was transformed into Streptomyces pactum ACT12 by conjugal transfer and the recombinant protein GM2886-His6 was detected in fermentation broth. GM2886-His6 was purified and characterized. Its size was nearly 36 kDa. GM2886-His6 showed proteolytic activity towards a variety of substrates and could even degrade insoluble substrates, such as azure keratin and chicken feathers. The optimal pH and temperature of GM2886-His6 for proteolysis of casein was pH 10.0 and 50 ℃, respectively. The enzyme activity was inhibited by PMSF, but not EDTA, indicating that GM2886-His6 was a serine proteinase. Our results laid the foundation for the research of the molecular biological mechanism on feather-degrading and for the further utilization of Fea-10.
8.Study on in vitro Inhibitory Activity of the Extracts from 9 Kinds of TCM for Dredging Collaterals and Dispelling Wind on Xanthine Oxidase

OBJECTIVE:To study in vi tro inhibitory act ivities of 9 kinds of TCM for dredging collaterals and dispelling wind on xanthine oxidase (XO),and to screen TCM with outstanding activity. METHODS :Using xanthine as substrate and xanthinase as reaction enzyme ,allopurinol as positive control ,with water extract and methanol extract (hereinafter referred to as “ethanol extract”)from the stem and leaves of Hedera nepalensis ,the whole plant of Piper wallichii ,the fruits of Rubus corchorifolius ,the root of Caragana sinica ,the root of Wisteria sinensis ,the root of Rubus crataegifolius ,the bark of Catalpa ovata ,the root of Campsis grandiflora ,the stem of P. hancei (hereinafter referred to by plant name )and petroleum ether ,dichloromethane,ethyl acetate,n-butanol and water fraction of active extract as the objects ,inhibition rate of each sample to XO was detected by spectrophotometry;IC50values were calculated with Graphpad prism 6.0 software to screen active extract/fraction. Double reciprocal method was used to determine the type of enzyme inhibition. RESULTS :Among 9 kinds of TCM and 18 kinds of the extracts ,the inhibitory rates to XO of 500 μg/mL extracts from each TCM(except for ethanol extract of P. wallichii ),250 μg/mL water extract and ethanol extract of H. nepalensis ,P. wallichii ,R. corchorifolius and P. hancei ,250 μg/mL water extract of C. ovata ,250 μ g/mL ethanol extract of C. sinica ,R. crataegifolius and C. grandiflora ,125 μ g/mL ethanol extract of C. sinica and R. crataegifolius,62.5 μg/mL ethanol extract of C. sinica were more than 50%. The IC 50 value of the ethanol extract from C. sinica was 43.43 μg/mL,which was lower than the extracts of other TCM ,and which was the active extract. The IC 50 values of petroleum ether,dichloromethane,ethyl acetate ,n-butanol and water fracti on of ethanol extract from C. sinica were >200,193.35,7.67, 14.80 and >200 μg/mL,respectively. The IC 50 value of ethyl XO was competitive-noncompetitive inhibition , which was different from competi tive inhibition of positive control. CONCLUSIONS:The ethanol extracts of C. sinica ,R. crataegifolius ,P. wallichii ,R. corchorifolius ,P. hancei ,H. nepalensis and the water extracts of H. nepalensis ,P. wallichii ,C. ovata show certain inhibitory activity in vitro to XO ,especially ethanol extract of C. sinica . The ethyl acetate fraction of the ethanol extract of C. sinica has similar inhibitory activity to allopurinol but their inhibition types are different.
