Objective To analyse the clinicopathological characteristics of synovial sarcoma (SS) from histomorphology,immunohistochemical (IHC) and fluorescence in situ hybridization (FISH),and to investigate the related factors influencing the prognosis of SS patients.Methods 94 cases were collected,including 60 SS cases and 34 in dined to SS cases.The expressions of common antibodies related to the diagnosis of SS Vimentin,CD99,AE1/AE3,EMA,CD34,Calponin,Ki-67 were detected by IHC,and the SYT-SSX fusion gene was detected by FISH.The clinical pathologic factors that affecting the prognosis of patients were analyzed through the statistical method.Results The positive rates of Vimentin,CD99,AE1/AE3,EMA,CD34,Calponin,Ki-67 were 100.00 ％,74.47 ％,36.17 ％,28.72 ％,17.02 ％,90.43 ％ and 60.64 ％ respectively.FISH results showed that 81 patients were tested with SYT gene translocation,including 54 cases who were considered as SS,and 27 cases who were suspected/inclined to SS,and the number of positive cells surpassed 80 ％.The gene translocation was not related with the patient' s age,sex,tumor position,histological type,differentiation of the cancer (all P ＞ 0.05).Single factor survival analysis showed that the degree of differentiation,tumor diameter,the recurrence and metastasis had certain influences on patients' survival time (all P ＜ 0.05).Multiple factors regression analysis showed that the degree of differentiation and tumor diameter were the risk factors influencing the prognosis of SS (risk coefficients ＞ 1,P ＜ 0.05).Conclusions IHC combined with FISH SYT-SSX fusion gene detection have the vital significance in the diagnosis of SS.The degree of differentiation and tumor size are the risk factors influencing the prognosis of SS.

To compare the effects of laparoscopic total or subtotal hysterectomy and myomectomy on ovarian reserve by measuring the serum level of anti-Mullerian hormone (AMH).A total of 96 patients with uterine fibroids underwent total laparoscopic hysterectomy (TLH group,n =32),laparoscopic subtotal hysterectomy (LSH group,n =31) and laparoscopic myomectomy (LM group,n =33).Changes in ovarian reserve were examined by measuring the level of AMH pre-operation and at 1 and 4 months post-operation.No significant differences existed in AMH in LM group[(1.42 ±0.65),(1.31 ±0.53) & (1.33 ±0.61) μg/L,P ＞ 0.05].The levels of AMH in both groups were significantly lower than those at pre-operation [(1.17 ± 0.11),(1.01 ±0.10),(0.48±0.54) & (1.18±0.93),(0.45 ±0.39),(0.14±0.0) μg/L,P＜0.001].The mean percentage decrease in AMH was lower in TLH group than that in LSH group at 4-month follow-up [(-37.4 ± 37.6) ％ vs.(-27.0 ± 23.0) ％,P =0.017].The results showed that LM had no effect on ovarian reserve.Both LSH and TLH had adverse effects on ovarian reserve and TLH was more marked.

AIM: To obtain the high expression of recombinant human stem cell factor - thrombopoitin (SCF-TPO) fusion gene and predict its structure property. METHODS: Tour primers were designed according to known sequence of TPO and SCF to amplify the functional amino acid domain of TPO and SCF by RT- PCR, respectively from fetus hepatocytes. The expression plasmid pET32a/SCF- TPO was constructed by VOE gene fusion technique and expressed in BL21(DE3)plysS. The fusion protein property, such as second structure, flexibility, and hydrophilicity were predicted by DS Gene and Protscale software. RESULTS: The expression vector, pET32a/SCF - TPO was constructed and the high expression of the SCF/TPO fusion protein was obtained, with the expression amount of up to 40% of the total cellular protein. DS Genel .5 and Protscale predict no new antigenicity in fusion protein, and the second structure and ioelectric point have no changes except four amino acids change in first structure. There are high flexibility and low hydrophilieity in the linker peptide. CONCLUSION: High expression of SCF- TPO fusion protein has been obtained and protein prediction shows that the fusion protein design is reasonable, which lay foundation for further study of biological fundation of SCF - TPO fusion protein.

Objective:Identification of some biochemical and physical properties for a new recombinant B7-2-PE40KDEL exotoxin fusion protein.Methods:12%SDS-PAGE separating and gel imaging analyzing,peptide mass fingerprinting,Western blot and MTT assasying were used respectively for identification of the protein.Results:Molecular weight of the recombinant B7-2-PE40KDEL was 72 628,5% of the difference to its theoretical value 69 561.The result of Western blot indicated that the purified recombinant B7-2-PE40KDEL could specifically bind with mAb anti-human B7-2 and the antibody against PEA,while the negative control did not.The recombinant B7-2-PE40KDEL digested with trypsin and then detected by MOLTI-TOF-MS.It was shown that the detected 15 peptides lied in the extracellular part of B7-2 and the truncated Pseudomonas extoxin PE40KDEL.Searching in the peptident data bank of Expasy website,we did not find any known proteins which was accordant with the above terms.The cytotoxic activity of the recombinant toxin with MTT method showed that the B7-2-PE40KDEL selectively killed Jurkat cell line which expressesed CD28 receptor well and had no killing effect on the Raji cell line unexpressing CD28 receptor.Conclusion:Recombinant B7-2-PE40KDEL exotoxin fusion protein we construct proves to be a new one with targeted killing bioactivity to B7:CD28 system.

Objective To investigated the value of intraoperative ultrasonography in guiding the surgical approach,improving the total resection rate of the tumor,and the survival time of patients who underwent resection of cerebral gliomas.Methods One hundred and ninteen patients with cerebral gliomas (pathologically confirmed) comprised the study groups.Patients who underwent intraoperative ultrasoundguided surgery were followed up for 6-60 months.In addition,randomly selected low and high grade cerebral glioma patients (all 30 cases of which were without intraoperative ultrasonography guidance) served as the control groups.Follow up included survival time,and the differences in survival time between the study groups and control groups were statistically analyzed.Results Complete follow-up data were obtained for 101 of 119 cases (84.9％) for a follow-up time of 6 60 months.Total removal of the lesion was achieved in 68 cases (67.3％) and subtotal removal in 33 cases (32.7 ％).Sixty-five cases had partial recurrence (64.4％) and 60 patients died due to tumor recurrence.Survival in patients with low grade gliomas (LGG) was significantly higher than that in patients with high grade gliomas (HGG).Patients with total tumor resection had a longer survival time than patients who had subtotal tumor resection.In the control LGG group,6-month survival was 96.7％,1-year survival was 73.3％,2-year survival was 53.3％,4-year survival was 36.7％ and 5-year survival was 26.7％.In the control HGG group,survival at 6 months,1 year,2 years,4 year and 5 years was 83.3％,36.7％,13.3％,3％,0 respectively.When comparing survival between the control groups and study groups,there was no significant difference at 6 months (P ＞0.05),but survival at 1 year,2 years,4 year and 5 years was different (P ＜0.05).Conclusions Intraoperative ultrasonography-guided resection of cerebral gliomas helps the surgeon to understand the relationship between the lesion and the surrounding structures,helps to protect normal brain tissue and increase the safety of surgery.It is of value in improving the total resection rate of tumors and the patient's survival time.

Objective: Pervious RT-PCR results revealed the possibility of effective regulation channel of Xiaoyao San in treating rats of syndrome of liver invading the spleen. This experiment was designed to explore CIS rats’ immunohistochemistry findings in hippocampal subregion CA1 and amygdaloid subregion BLA after suppressing both amygdaloid AMPA receptors by microinjection of CNQX. To compare Xiaoyao San group with CNQX group, and to analysis of these findings may prove the possibility of effective regulation channel of Xiaoyao san treating this syndrome. Methods: 75 male SD rats were randomly divided into 5 equal groups: normal group, CIS group, sham-operation group, CNQX group and Xiaoyao San group. The number of GluR2 positive cells in hippocampal subregion CA1 and amygdaloid subregion BLA in these 5 groups were examined by immunohistochemistry test. Results: Compared with control group, the number of GluR2 positive cells decreased in hippocampal subregion CA1. Statistical analysis showed that no difference existed in control, CNQX and Xiaoyao San groups in CA1 region. Except CNQX group, tendency of changes were contradictory in BLA and CA1 region. In addition, the least in BLA in CNQX group. This finding revealed that Xiaoyao San may act the same as CNQX, thus Xiaoyao San afforded effective protection against this depression syndrome through suppression of both amygdaloid excitability. Conclusion: The balance coordination between hippocampus and amygdala may be the key in transformation from adaption to damage in a state of stress, was one of the central mechanisms of liver governing smoothing qi flow (LGSQF) on regulating the stress. Xiaoyao San may effectively regulate the balance of the excitability of hippocampus and amygdala. This hypothesis may reveal the regulation channel on Xiaoyao San affording effective protection against liver invading the spleen syndrome.

Although previous reports showed drug-eluting stent (DES) could effectively inhibit neointima formation, in-stent restenosis (ISR) remains an important obstacle. The purpose of this study was to investigate different effects of paclitaxel on proliferation and cell cycle regulators between vascular smooth muscle cells (VSMCs) and vascular endothelial cells (VECs) of rats in vitro. The cultured VSMCs and VECs of rats from the same tissues were examined by using immunohistochemistry, flow cytometry and Western blotting in control and paclitaxel-treated groups. The results showed paclitaxel could effectively inhibit proliferation of VSMCs and VECs. However, as compared with VECs, proliferation of VSMCs in paclitaxel-treated group decreased less rapidly. The percentage of cells in G0-G1 and G2-M phases was reduced, and that in S phase increased after treatment for 72 h. The expression of cyclin D1 and B1, p27 and PCNA in VSMCs of paclitaxel-treated group was up-regulated, but that of p21 down-regulated as compared with VECs. It is concluded that there are significant differences in the expression of cell cycle regulators and proliferation rate between paclitaxel-treated VSMCs and paclitaxel-treated VECs, suggesting that the G1-S checkpoint regulated by paclitaxel may play a critical role in the development of complications of DES, which provides new strategies for treatments of ISR.

Objective: To determine the factors influencing unprotected sexual behavior among college students，so as to provide the reference and basis for better sex safety education in college. Methods: From March to August, 2022, 3 188 college students from three universities, namely "double first class" universities, ordinary public undergraduate universities and ordinary private universities, were selected by stratified random cluster sampling method. Cognitive Style Graphic Test, Adolescent Health related Risk Behavior Questionnaire and deographic information questionnaire were used to investigate. Results: Cognitive style and unprotected sexual behavior were positively correlated ( r=0.13, P <0.05). A total of 18.23% ( n =314) of participants reported engaging in unprotected sexual behavior. Binary Logistic regression analysis of participants grouped by cognitive style indicated major factors affecting unprotected sexual behavior among college students. In the field independence group, male students, senior students, and those with frequent romantic encounters (1, 2, and ≥3 times) had more unprotected sexual behavior ( OR =1.28, 1.79, 1.35, 1.68 , 1.84, P <0.05). Four major factors were associated with unprotected sexual behavior in the field independence group: junior and senior college students, those that study liberal arts, urban student source, and those with frequent romantic encounters (1,2 and ≥3 times) ( OR =1.91, 2.01, 1.26, 1.40, 1.46, 1.78, 1.99, P<0.05). Conclusion College students unprotected sexual behavior is associated with multiple factors. Cognitive style is an important reference variable. The factors influencing this behavior have differing effects according to the different cognitive styles among college students.

Objective To observe the expression of vimentin (Vim) after silence of interleukin-1β (IL-1β) in rats with spinal cord contusion (SCC). Methods The model of SCC was established in 30 Sprague-Dawley rats with Allen's method. The rats were randomized into vector group (n=15) and silence group (n=15), which were injected blank lentivirus vector and vector of IL-1β siRNA, respectively; and divided in three, seven and 28 days subgroups. The relationship between IL-1βand Vim was predicted with GeneMANIA bioinformatics. The expression of Vim protein and mRNA in spinal cord was detected with immunohistochemistry and real-time quantitative polymerase chain reaction. Results GeneMANIA bioinformatic analysis indicated that there was some direct and indirect relationship between IL-1β and Vim. The Vim protein and mRNA expressed in the spinal cord, and was less in the silence group than in the vector group (t>2.875, P<0.05). Conclusion Silence of IL-1β can inhibit the expression of Vim in SCC rats, which may promote the recovery of spinal cord function.
