Objective To investigate the effect of strong ion-irradiation and active macrophage on vascular endothelial cells. Methods The vascular endothelial cells(ECV-304) were divided into 3 groups: ECV304(A), ECV-304 with radiation (B)and ECV-304 with active macrophage U937 and radiation(C). The survival rates, cell cycle and apoptosis of ECV-304 were determined 48 hours after irradiation. The concentrations of TGF-?1 and VEGF in the cultured supernatant were determined by ELISA method.The protein expression of the endothelial receptor KDR in ECV-304 cells was detected by immunofluorescence and flow-cytometry while its mRNA expression was detected by RT-PCR. Results After high dose of ion-irrdiation, the cellular proliferation activity decreased, apopototic cells increased and the cell ratio in G2/M phase increased.In addition,The secretion of VEGF and TGF-?1 was promoted with the increased KDR expression. Active macrophages could alleviate the decrease of cellular proliferation and the increase of apoptotic cells and cell ratio in G2/M phase induced by ion-irriation.Furthermore,the VEGF secretion and the KDR expression were enhanced and the secretion of TGF-?1 was inhibited by the active macrophages. Conclusions Ion irradiation could induce the decrease of cellular proloferation, the increase of apoptotic cells and the cells in G2/M phase.This might partly be related to the inhibiting roles of TGF-?1 secreted by the ECV-304 cells.Active macrophages might protect the endothelial cells from radiation injury.

Objective To research the effects of treating hemoptysis due to bronehiectasis and bronchography of selective filling bronchus with meglumine diatrizoate-Yunnan Baiyo colloidal solution. Methods In 30 cases of recurrent hemoptysis due to bronchiectasis which had been diagnosed by clinic, chest photos and bronchofibroscopy, selective filling bronchus with meglumine diatrizoate-Yunnan Baiyao colloidal solution were done in 15 cases for treatment and bronchography { experiment group) , and only internal medicine were used in other 15 cases (comparison group) ,and all 30 cases were followed up for 5 years. Results 15 cases of selective bronchography of experiment group were all successful, and the effects of curing hemoptysis and relaxing the recurrence were better than comparison group { P < 0. 05).Conclusion Meglumine diatrizoate-Yunnan Baiyao colloidal solution is not only a effective medicine for treating hemoptysis due to bronchiectasis,but also a expectant contrast medium for bronchography,and has the practicality of research and development.

Objective To investigate molecular typing and drug resistance patterns of 98 Klebsiella pneumoniae (K.pneumoniae) isolated from type 2 diabetes patients complicated with maxillofacial infection,to research the virulence and resistance mechanisms.Methods The study was a prospective study that adopted the method of continuous sampling from fixed location,from March 2010 to October 2012.The maxillofacial surgery patients diagnosed with type 2 diabetes complicated with maxillofacial infection were chosen in 7 hospitals in Zhengzhou as the research object,and a total of 431 pus sample were collected continuously,in which 98 strains K.pneumoniae were isolated and identified.The Kirby-Bauer disk diffusion test was conducted in 98 strains to determine the resistance to 19 antibacterial agents.K.pneumoniae chromosomal DNA were digested by restriction endonuclease Xba Ⅰ and analyzed by pulsed-field gel electrophoresis (PFGE).PFGE patterns of K.pneumoniae strains were analyzed using Fingerprinting software.The relationship between the molecular types and resistance phenotype was observed.The extended spectrum β-1actamase-producing K.pneumoniae were screened out by the double disc synergy test (DDST)Polymerase chain reaction (PCR) was used to detect resistant genotypes,serotype and virulence genes.The purified PCR products of resistant genes were cloned and sequenced.Hypermucoviscosity phenotype of all strains were determined by string test.Results Much severer drug-resistance for K.pneumoniae was identified and the result of extended-spectrum beta-lactamase producing rate was 57.1 ％.Ninety-eight strains were dispatched into 13 groups by PFGE.No dominant bands and specific extended-spectrum beta-lactamase DNA bands were found.The results of PCR showed that among the 56 strains of extended spectrum β-lactamase-producing K.pneumoniae,40 were positive for blaSHV (accounting for 71.4％),28 positive for blaTEM (accounting for 50.5％),21 positive for blaCTX-M (accounting tor 37.5％).The sequencing results were as follows:TEM-1,CTX-M-3 and a variety of SHV.Serotype K1,K2,K3,K5,K20,K54 and K57 and 3 kinds of virulence genes were detected,but not in strong toxicity-based.Hypermucoviscosity positive rate was 31.6％ (31/98).Conclusion Much severer drug resistance of K.pneumoniae in this study was identified and resistant mechanism was complex,in which strong toxicity serotype and virulence geues exist,which need more attention from clinical.

BACKGROUND:Skin and soft tissue defects of hands are easy to appear due to different factors, such as trauma. Flap transplantation is a clinical treatment for soft tissue defects of hands. Dorsal foot flap, which is thicker with constant vessel traveling, is very suitable for repair of hand soft tissue defects. OBJECTIVE: To explore the success rate and survival rate of dorsal foot flap transplantation in the repair of hand skin and soft tissue defects. METHODS: A retrospective analysis was performed on the clinical data of 86 patients with skin and soft tissue defect of hands. These patients were respectively given conventional skin wound skin grafting (control group) and dorsal foot flap transplantation (observational group). Hand appearance, infection rate, perception recovery and complications were observed during the postoperative folow-up. RESULTS AND CONCLUSION:Skin flaps al survived after grafting, and the texture of flaps was good. Patients were satisfied with hand shape and the recovery of hand function. After flap transplantation, one patient had local skin necrosis at the donor region, and the scar healed after dressing; another patient had venous crisis, and was given vascular re-anastomosis. In the observation group, the postoperative appearance was better than that of the control group, the infection rate was lower than that of the control group, the perception recovery was better, and there were fewer complications (P < 0.05). These findings suggest that the dorsal foot flap transplantation is better to repair skin and soft tissue defects in the hand.

AIM To study the protective effect of total glucosides of paeony(TGP)on complete cerebral ischemia reporfusion injury and its mechanism of action.METHODS A complete cerebral ischemia[FQ(10。46,X-WZ]- reperfusion model of rats was induced by clipping three arteries. The changes in eye colour, SOD activity, MDA contents and histopathological characteristics were observed. RESULTS TGP (TGP 10、20、40 mg?kg -1 ) markedly reduced the change of eye colour; TGP(20 mg?kg -1 ) obviously elevated SOD activity decreased and inhibited the MDA of increased contents of cerebral cortex, hippocampus and corpus striatum of cerebral ischemia reperfusion in rats; TGP(20 mg?kg -1 )possessed protective effects on the histopathological changes of ischemia cerebral tissues in rats. CONCLUSION TGP possessed obvious protective effects on the cerebral ischemia reperfusion injury, suggesting that the protective action of TGP is relative to anti-oxidative action.-

OBJECTIVE To investigate the effect of quercetin on primary cultured newborn rat cortex neuron cell which is estrogen depletion, and discuss the possible mechanism, to provide new ideas and strategies for developing a drug of neurodegenerative disease. METHODS Rat cortex neurons were isolated from one day old Sprague Dawley rats and treated with estrogen, quercetin and estrogen receptor antagonists (ICI182,780). Cell viability was determined by MTT assay, neurite outgrowth was measured by fluorescent microsope and estrogen receptors were determine by Western blot. RESULTS Quercetin functions like estrogen to increase cortex neuronal cell viability, the Que (50, 100 μmol·L-1) group compared with the control group could significantly improve the activity of the cortical neurons(P<0.05). It can also increase neurite out growth, the Que (50,100 μmol·L-1) group significantly promoted the formation of synapse, most of the neurons were full, and the synapses of neurons became thick, growth, and connect to a dense neural network. And in the Western blot experiments, Que (50, 100 μmol·L-1) group could obviously increase the expression of estrogen receptor alpha protein, in addition, the neural protective effect of quercetin can be inhibited by ICI182,780. CONCLUSION Quercetin like estrogen can protected cortex neuronal and the effect of quercetin on cortex neuronal cells was mediated by estrogen receptor alpha.

Objective To make a preliminary study on the results from qualitative and quantitative detection of low concentra-tion hepatitis B surface antigen(HBsAg).Methods 85 HBsAg low concentration serum samples (ELISA method test re-sults were 0.60.05)and 0.60.05).The positive detection rate showed statistically difference between ELISA (70.6%)and CMIA,TRFIA in 0.6TRFIA>ELISA.②The contents of HBsAg showed statistically difference among 0.6TRFIA>ELISA.③There was a positive correlation between the three methods of HBsAg content (t=2.939,2.928,60.915,P<0.05).The correlation between CMIA method and TRFIA method was the best (r=0.989).Con-clusion CMIA was the first choice for testing low concentration HBsAg,TRFIA was the second.For the specimens of the low concentration HBsAg detected by ELISA should be suggested to clinical and retested by CMIA or TRFIA in order to a-void missing detection.And it was not recommended to clinical that different methods of quantitative or half-quantitative re-sults were transverse compared in order to avoid misdiagnosis.
