3.CHARGE SELECTIVITY OF PROTEINURIA IN DIABETES

The clearance of proteins differing in charge and/or size (IgG4, total IgG and albumin) was evaluated in 95 diabetic patients. Urinary IgG4 values increased in group mi-croalbuminuria. The increased urinary IgG4 excretion correlated significantly with the degree of albuminuria and diabetic retinopathy. Urinary total IgG levels increased obviously only in group with massive macroalbuminuria. The IgG4/total IgG ratio was highest in group mi-croalbuminuria, and its change had a special pattern. Urinary IgG4 and IgG4/total IgG ratio may be sensitive parameters in assessing early protein charge-selectivity impairment and in detecting incipient diabetic nephropathy.

Postherpetic neuralgia (PHN ) is one of the most frequent complications of herpes zoster, with the feature of intractable chronic pain. The higher incidence of PHN has been found in elder people and the people with low immunity. Because its pathogenesy is not clear, the clinical treatment appears to be very difficult, and all kinds of the treatments just relieve pain. Neurotropin combined with nerve block is one of the new therapies recently, which is deserved to be used in clinic and to be generalized.

6.Type 2 inflammatory responses in atopic dermatitis

Studies have shown that the type 2 inflammatory response and related cytokines such as interleukin-4 and -13, play a key role in atopic dermatitis (AD) . Th2 cells and type 2 cytokines are the core factors in the type 2 inflammatory response, which is mainly involved in the pathogenesis of AD through 3 pathways: aggravating skin barrier defects; activating and strengthening the itching response; pathologically reprograming regulatory T cells into Th2-like regulatory T cells to amplify the type 2 inflammatory response. This review explores the relationship between the type 2 inflammatory response and AD, aiming to help optimize treatment regimens and provide new treatment options for patients with AD.
7.Inhibitory effect of quercetin on recombinant human protein kinase CK2 holoenzyme and its kinetics analysis

Object To study the direct effect of quercetin on recombinant human protein kinase CK2 holoenzyme and its kinetics. Methods Recombinant human protein kinase CK2? and ? subunits were cloned and expressed by gene engineering, and were purified. The two subunits were mixed at the same molar ratio, thus reconstituting CK2 holoenzyme, which displayed the maximum bioactivity. The CK2 activity was assayed by detecting incorporation of 32 P of [? 32 P] ATP into the substrate in the various conditions. Results The recombinant human protein kinase CK2 was the second messenger (Ca 2+ , cAMP and cGMP) independent protein kinase, the characterization and function of the reconstituted holoenzyme were consistent with those of native CK2. It was found that quercetin strongly inhibited the holoenzyme activity of recombinant human protein kinase CK2 with an IC 50 of 522 nmol/L, which was much more effective than DRB and A3, known as CK2 special inhibitors. Kinetic studies of quercetin on recombinant human protein kinase CK2 showed: the inhibition was competitive with ATP and noncompetitive with casein. Conclusion Quercetin is a potent inhibitor of recombinant human protein kinase CK2. The inhibition may be another molecular mechanism of antitumor effect of quercetin. This study provides a simple and rapid screening method for the development of more effective inhibitors of recombinant human protein kinase CK2.
8.ENHANCEMENT OF RESISTANCE OF HUMAN BONE MARROW CELLS TO ANTICANCER AGENTS BY mdrl GENE TRANSFECTION

To investigate the possibility to enhance resistance of human bone marrow cells to anticancer agents by transfection of mdrl gene. We transferred mdrl gene into human bone marrow cells with plasmid pHaMDR1/A containing human mdrl cDNA by Lipofectin.Pgp,mdrl gene expression product was detected by both flow cytometry and immunohistochemistry.Also,Pgp function was tested by efflux study using rhodamine.The resistance of human bone marrow cells to anticancer agents was assayed by color forming unit culture after exposure to Vincristine,Daunomycin, and Vp16.The results showed that mdrl gene was successfully transferred into human bone marrow cells, and it expressed.Biological activity of Pgp was confirmed by efflux study using rhodamine.Transferred human bone marrow cells possessed resistance to anticancer agents.It suggested that transfection of mdrl gene can increase resistance of human bone marrow cells to anticancer agents.
9.Exploration of acupoints selection rule of coronary heart disease treated with acupuncture and moxibustion in recent 10 years.

With the key words such as acupuncture and moxibustion, coronary heart disease, etc., CNKI, WANFANG and VIP databases were retrieved to get Chinese literature of coronary heart disease treated with acupuncture and moxibustion in recent 10 years. Related 76 articles were collected and 72 acupoints were applied. The potential relation among acupoints of the top 20 was analyzed by hierarchical cluster analysis. It was discovered that the main acupoints were Neiguan (PC 6), Xinshu (BL 15), Danzhong (CV 17), Geshu (BL 17) and Zusanli (ST 36), etc., especially the specific acupoints. The back-shu points and five-shu points were emphasized. Pericardium meridian of hand-jueyin was the high frequency meridian. The four extremities were the main position.
