2.Determination of Lasiokaurin in the Stems and Leaves of Rabdosia nervosa by HPLC

Objective To develop a HPLC method for the determination of Lasiokaurin in the stems and leaves of Rabdosia nervosa.Methods HPLC was performed on a Kromsil C18 reversed-phase column(250mm?4.2mm,5?m).All of the reference substances and samples were separated with the mobile phase of methanol :water(55:45) under isocratic elution for 30min,flow rate was 1.0ml/min,the detection wavelength was 234nm,and the column temperature was 30℃.Results The content of Lasiokaurin was 0.005mg/g in the stems and 0.372mg/g in the leaves,the content in the stems were almost 1/75 times as much as that in the leaves.The average recovery of Lasiokaurin was 96.21%.Conclusion This developed method,which is simple and with good precision,high sensitivity and selectivity,can be used for quality evaluation of Lasiokaurin in Rabdosia nervosa.
3.Relationship between levels of plasma coagulation factors and acute myocardial infarction in low age period

Objective To explore the relationship between the levels of plasma coagulation factors (F) and acute myocardial infarction (AMI) in low age period (<60 years old) and their diagnostic value in diagnosing AMI in low age period.Methods One hundred and sixty inpatients with low age AMI in the cardiology department of the Heze Municipal Hospital were selected as the case group,and contemporaneous 160 cases of low age non-AMI served as the control group.F Ⅱ,FⅦ,FⅧ,fibrinogen (Fg) and von willebrand (vWF) were measured with enzyme-linked immunoabsorbent anti-sandwich assay.The relationship between coagulation factors and low age AMI was analyzed with univariate and multivariate analysis,and their value for diagnosing low age AMI was evaluated with diagnostic test and receiver operating characteristics (ROC) curve.Results The univariate analysis showed that FⅡ,FⅦ,FⅧ and Fg levels had significantly statistical difference between the case group and control group(P<0.05),and the vWF level had no statistically difference(P>0.05).The multivariate analysis indicated that the FⅡ level≥ 14.27 μg/L and FⅦ level ≥22.99 μg/L were the independent risk factors for low age AMI.The value of FⅡ for diagnosing low age AMI was lower,and the optimal cut off value of Fg for diagnosing low age AMI was 22.99 μg/L,its area under ROC curve was 0.709 with a moderate diagnostic value,and the sensitivity (91.88%) and negative predictive value (86.02%) were higher,the false negative rate (13.98%) was lower,and the accuracy (70.94%) was moderate.Conclusion The FⅡ level ≥14.27 μg/L and Fg level ≥22.99 μg/L are the independent risk factors for low age AMI,and detecting the Fg level could have hint significance in diagnosing low age AMI.
4.Study of protection of muscone against local cerebral ischemia

Objective: To investigate the effect of muscone on infarction volume and neuronal glutamate transporter EAAC1 mRNA expression in rats. Methods: Rats were divided randomly, focal ischemic models of middle cerebral artery occlusion (MCAO) were achieved by inserting nylon surgical thread through the internal carotid artery(ICA) into the anterior cerebral artery(ACA). The rats were sacrificed, the brain was stained by TTC and the area of infarcton was measured. The expression of EAACl mRNA in the hippocampal regions was assessed by RT PCR. Results: Muscone could remarkably reduce tbe infarction volume and expression of EAAC1 mRNA. Conclusion: Muscone plays a significant role in against local cerebral ischemia, the mechanism may be related to reducing EEACl mRNA expression.
5.Determination of Oridonin in Rabdosia nervosa by HPLC

Objective To develop a HPLC method for the determination of oridonin in the leaves and stems of Rabdosia nervosa.Methods All of reference substances and samples were separated with the mobile phase of methanol:water (45 ∶55) under isocratic elution for 20 min on a Kromasil C18 reversed-phase column(250 mm?4.0 mm,5 ?m),the flow rate was 1.0 mL?min-1,the detection wavelength was 235 nm,and the column temperature was 30 ℃.Results The content of oridonin was 0.047 mg/g in stems and 0.791 mg/g in leaves,the content in stems being about one seventeenth of that in leaves.The average recovery of oridonin was 98.33 %.Conclusion This method is simple,sensitive,accurate and suitable for quantitative determination of oridonin in Rabdosia nervosa.
6.MRI of demyelinating pseudotumor of the central nervous system

Objective To describe the MR manifestations of demyelinating pseudotumor of the central nervous system (CNS), and to discuss the pathologic features and MR diagnostic value and limit in this disease entity. Methods Seven pathologically proved and one clinically proved cases of demyelinating pseudotumor of CNS were studied with MR imaging, and the MR imaging features were retrospectively analyzed. Results MR imaging demonstrated localized mass without adjacent multiple accompanying lesions in all 8 cases. On T 1WI, the lesions showed homogenous low signal in 5 cases, inhomogenous low signal in 2 cases, and mixed high and low signal in 1 case. On T 2WI, the lesions presented as homogenous high signal in 5 cases and inhomogenous high signal in 3 cases. Of the 7 cases with Gd DTPA administration, marked enhancement was seen in each case. The enhancement pattern of vertical distribution to the lateral ventricle was demonstrated in left frontoparietal lobe in one case, and predominant dorsal white matter enhancement of the cervical spinal cord was revealed in another case. Follow up MR imaging showed no lesion recurrence, and gradual shrinkage of the lesion after steroid therapy was demonstrated in one case. Conclusion It is a difficult task to make the correct diagnosis of CNS demyelinating pseudotumor based on the clinical information and imaging findings, and this disease entity is often misdiagnosed as tumor by MR imaging. Thorough analysis of the clinical history and careful observation of MR manifestations (especially contrast enhanced MR findings) would be helpful in diagnosing the demyelinating pseudotumor and, in such circumstances, providing test steroid therapy, thus avoiding the devastating injury caused by surgery or radiation therapy.

8.Reformation and Practice of the Experimental Course of Fermentation Engineering

The traditional fermentation engineering experiment requires a reform on the experimental con-tents and teaching pattern. According to the production process of industrial enzymes, we set up a two-week comprehensive experiment. The students designed and prepared the experiment by themselves. Moreover, the pattern of self-management was used in the process and the experiment scores included the self-assess-ment and objective assessment. It was proved that the new teaching pattern increased the study interesting of students, inspired their initiative and trained their spirits of team cooperation. The teaching effect was im-proved markedly and good ideas are also put forward to solve the possible problem.
9.Establishment of Loop-mediated Isothermal Amplification (LAMP) Method for Campylobacter jejuni Detection

A rapid LAMP detection method with primers designed on genus-specific region identified in the gyrA gene was established in this assay. All four Campylobacter jejuni from different sources were detected positive and fourteen non Campylobacter bacteria were negative, which shows excellent specificity of the primers. Compared with plate count and PCR method, the LAMP method and the PCR method had equal sensitivity, which were three orders of magnitude higher than plate count. In this assay, we also found out that the treatment of DNase could reduce the dead bacteria DNA effectively. The LAMP detection on chicken indicated relatively good result on detection of Campylobacter jejuni combining with treatment of DNase.
10.Surface marker changes in human umbilical cord-derived mesenchymal stem cells after cryopreservation and resuscitation

BACKGROUND: Mesenchymal stem cells are the stem cells that possess the capability for self-renewal and multi-directional differentiation. Umbilical cord is the tissue outside the embryos and would be fallen off after parturition. In addition, it has wide source and no ethical restriction, so it is promising to be the first choice for mesenchymal stem cells. OBJECTIVE: To detect the surface markers CD29, CD44, CD49e, CD73, CD90, CD34, CD45, and CD271 of human umbilical cord-derived mesenchymal stem cells (hUCMSCs) prior to and after cryopreservation and resuscitation. METHODS: After isolation and culture, morphology of the primary, P4 and P8 hUCMSCs was observed prior to cryopreservation and after resuscitation. Surface markers CD29, CD44, CD49e, CD73, CD90, CD34, CD45, and CD271 of primary, P4, and P8 hUCMSCs were detected through the use of flow cytometry prior to cryopreservation and after resuscitation RESULTS AND CONCLUSION: hUCMSCs prior to cryopreservation and hUCMSCs of different passages after resuscitation present the same phenotype, i.e., positive for CD29, CD44, CD49e, CD73, and CD90, and negative for CD34, CD45, CD271. These findings suggest that primary hUCMSCs do not present changes in surface markers after cryopreservation and resuscitation.
