2.Proliferation and Apoptosis in the Developing Mouse Cochlear Sensory Epithelium

Objective To observe the principles of cell proliferation and apoptosis in the developing mouse cochlear sensory epithelium, and investigate correlation with differentiation of hair cells.Methods The developing inner ears of C57BL/6 embryonic mice,aged from eighth embryonic day (E8) to just natal mice (P0), were observed by transmission electron microscope.Results Cochlear primordium arose at E10. After E14, sensory epithelia began to differentiate. Corti primordium arose at E16, and shapes of supporting and hair cells tend to mature. But the organ of Corti was still poorly developed at P0. Meanwhile Cell karyokinesis of the cochlea arose at E11 ,then gradually increased. But it began to decrease from E14. Cell apoptosis arose at E13.It reached its peak from E14 to E15,then it began to decrease gradually.Conclusion Cell proliferation and apoptosis were inevitable in the developing mouse cochlea.Cell proliferation, cell apoptosis and differentiation of hair cells consequently arose and overlapped each other. Homeostasis between cell proliferation and apoptosis has an important role in the developing mouse cochlear sensory epithelium.
3.Quantitative analysis of the 5-lipoxygenase mRNA in the rat peritoneal macrophages cultured in vitro by capillary electrophoresis

Objective To quantify the mRNA of 5-lipoxygenase (5-LO) in the rat peritoneal macrophages cultured in vitro by capillary electrophoresis (CE). Methods The rat peritoneal macrophages were isolated and cultured for indicated time. The mRNA of 5-LO was detected by RT-PCR, and the products of RT-PCR were quantified by CE. Results The DNA fragments in the 100 bp DNA marker and the products of the RT-PCR were separated successfully by CE with the sieving buffer containing 1.8% hydroxypropylmethylcellulose (HPMC). It proved that there were no significant changes on the expressions of 5-LO mRNA when the cells were cultured for 72 h quantified by CE. The mRNA of 5-LO significantly decreased by almost 80% by CE with the cells cultured for 120 h in vitro.Conclusions The products of RT-PCR could be separated and quantified by CE directly.The 5-LO mRNA could express normally in the rat peritoneal macrophages for 72 h in vitro.
4.The change and clinical significance of serum tumor necrosis factor-?、interleukin-6 and interleukin-8 after cutting balloon angioplasty and stent implantation

Objective To evaluate the effect of cutting balloon angioplasty and stent implantation on serum TNF ?, IL 6 and IL 8, and investigate the relationship between the concentrations of serum TNF ?, IL 6 and IL 8 after operation and the incidences of late cardiac events after intervention Methods Eighty patients underwent PTCA with normal balloon or cutting balloon Values of serum TNF ?, IL 6 and IL 8 before and after intervention were measured using enzyme linked immuno sorbent assay Results The concentrations of serum TNF ?, IL 6 and IL 8 were increased significantly in the restenosis group, especially in the POBA group There was no obvious difference in no restenosis group Conclusion The lower restenosis rate in cutting balloon group was related to its lower injury and inflammation of blood vessels The concentrations of serum TNF ?, IL 6 and IL 8 6 hours after intervention may be the predictable sign of restenosis after intervention
5.THE CHANGES OF FN AND LN IN PULMONARY ARTERY OF HYPOXIC RATS

Objective In order to investigate the mechanism of the remodeling of pulmonary vascular in hypoxic pulmonary hypertension,normobaric hypoxic rat model (FIO 2=10%)was used to observe the changes of fibronectin(FN)and leminin(LN)within the pulmonary artery by morphological methods. Methods The normobaric hypoxic chamber was used to establish the hypoxic rat model. The lungs were taken off and stained with immunohistochemical method.Some specimens were examined by electromicroscopy. Results Immunohistochemical analysis revealed progressive increase in the deposition of FN and LN,which was most prominent in hypoxia 7d group.The content of FN was higher than LN.Electromicroscope revealed that the SMC of tunia media had transitted from a contractile to a synthetic phenotype. Conclusion Long term hypoxic exposure resulted in increasing of FN and LN in the wall of pulmonary artery.These increase not only raise the tension and make thickening of the vessels themselves,but also provide an environment for the transition of SMC from a contractile to a synthetic phenotype.FN and LN played an important role in the remodeling of pulmonary vascular directly and indirectly. AIM: To investigate the pentanucleotide repeat(PNR) polymorphism of apolipoprotein(a) in patients with coronary heart disease(CHD) in Hubei area, and evaluate the association of polymorphism of apo(a) PNR with the level of serum lipid. METHODS: Objects examined were composed of two groups: 88 patients with CHD and 153 healthy controls. Lp(a), TC,TG, HDL-C, LDL-C, ApoAⅠand ApoB of two groups were tested. Meanwhile,the PNR in the 5' control region of the Apo(a) was detected by using polymerase chain reaction (PCR) and high voltage polyacrylamid gels electropherosis. 7.The application of nalmefene in sufentanil and propofol anesthesia for abortion

Objective To investigate the effect of nalmefene on sufentanil and propofol anesthesia for abortion and its impact on BIS. Methods One hundred and twenty patients undergoing abortion patients were randomly divided into group A, B, C, and D (n = 30 each). Patients in group A and B received 0.2 μg/kg or 0.3 μg/kg sufentanil, respectively, followed with 1.5 mg/kg propofol for induction of anesthesia post-pretreatment with 0.2 μg/kg nalmefene. Patients in group C and D received induction of anesthesia as patients in group A and B. According to the BIS and fluctuation of hemodynamic , the amount of propofol was adjusted. If necessary, additional single intravenous injection of 0.5 mg/kg propofol. The mean arterial pressure (MAP), heart rate (HR), pulse oxygen saturation (SpO2) and respiratory rate (RR) in patient before injection (T1), the eyelash reflex (T2), dilatation (T3), curettage (T4) and surgery awake (T5) were detected. The additional amount of propofol , operation time , recovery time of surgery , the steward score of orientation recovery after 1min of surgery , body movement reaction , cough , respiratory depression , postoperative visual analog digital score (VAS) 15 min later were also recorded in each group. Results Compared with group A, propofol could reduce the intraoperative body movement reaction rate , with lower postoperative VAS in group B and group D (P <0.05, respectively), with no significant difference between group C and group A (P > 0.05). The rapid recovery, surgery within 1 min orientation recovery were higher in group B, C, D compared with group A (P <0.05). However, orientation recovery score in group D was higher than that in group B (P < 0.05); The respiratory depression and choking were higher in group A and B than those in group C , D (P < 0.05, respectively). Conclusion The doses of 0.2 μg/kg nalmefene can effectively antagonize the respiratory depression , delay recovery and other adverse reactions in painless which induced by sufentanil , and the dose of nalmefene in this study failed to enhance the effect of analgesic and change the BIS values.
8.Anti-tumor immunity and mechanism of microwave ablation

Microwave ablation,as a minimally invasive therapy,causes focal hyperthermic injury to ablated tumors.It also induces anti-tumor immunity which could contribute to another mechanism of tumor destruction.Combination with immunotherapy could further enhance the tumor-specific immunity,lead to stronger inhibitory effects on both primary and metastatic lesions and decrease recurrence and metastasis.Thus,combination of microwave ablation and immunotherapy is a promising approach in the treatment of solid tumors,especially for advanced cancers.This article reviews recent advances in the mechanism and application of immune effect induced by microwave ablation.
9.Determination of Weight Average Molecular Weight and Molecular Weight Distribution of Ferric Carboxy-maltose by HPGPC

OBJECTIVE:To establish a method for the determination of weight average molecular weight(Mw)and molecular weight distribution (D) of ferric carboxymaltose. METHODS:HPGPC method was adopted to detect the Mw and D of 3 batches of Ferric carboxymaltose injection (imported) and its raw material (self-made). The determination was performed on TSK G4000 PWXL column with 0.1% sodium azide solution with the flow rate of 0.5 ml/min. The detector was refractive index detector;the column temperature was set at 35 ℃,and sample size was 20 μl. The results were calculated with GPC software. RESULTS:RSDs of precision,stability and reproducibility tests were all lower than 3.0%(n=6);Mw and D of 3 imported samples were 157 667 and 1.30;those of self-made samples were 162 000 and 1.42. CONCLUSIONS:The method has high precision,good stability,repeat-ability and durability. It can be used for the determination of Mw and D of ferric carboxymaltose.
10.Clinical analysis of laryngeal neuroendocrine carcinoma

Objective:To investigate the clinical characteristics and therapy of laryngeal neuroendocrine carcinoma(LNEC).Method:Ten cases of LNEC treated in our department were retrospectively analyzed.Result:Eight out of 10 cases were were atypical carcinoid:1 case treated as supraglottic horizontal partial laryngectomy died 22 months after operation; 1 case was lost;the other 6 cases were followed up for more than 2 years. Two cases were small cell type,one received supra-cricoid partial laryngectomy-cricohyoidopexy (SCPL-CHP) ,neck dissection,and radiotherapy,but died after 14 months;the other was receiving radiotherapy.Conclusion:LNEC is a rare malignant tumor and the diagnosis relies on clinical characteristics, histopathological and immunohistochemistry examination. The different types of LNEC should give different treatment protocols.
