Objective To elucidate the molecular basis on the differences of infectivity in infected cells between Sindbis virus(SINV:YN87448 virus)and Sindbis like virus(SINLV:XJ-160 virus).Methods Compare the E(glycoprotein)gene sequence and secondary structure of YN87448 virus and XJ-160 virus by bioinformatics analysis.Analyze the contribution of E gene to the biological differences between SINV and SINLV by constructing recombinant virus.Results By bioinformatics analysis,YN87448 virus and XJ-160 virus have the same genomic structure,which has 11 717 nt and 11 626 nt respectively.There are 82 amino acid differences between E gene of these two viruses,and showed scattered distribution.The main peak is basically the same for the hydrophobic of the E gene protein,but in some region existing small differences.The recombinant virus which exchanged the E gene of XJ-160 virus with YN87448 virus totally showed the biological character of YN87448 virus,either in the showing time of CPE,plaque forming time and plaque diameter,or in expression of functional proteins.Conclusion E gene plays a major role in the differences of infectivity in infected cells between SINV and SINLV,this result provide the molecular biological evidences for elucidating the biological differences between SINV and SINLV.
3. The plasma concentration of omeprazole and metabolites 5′- hydroxy omeprazole, omeprazole sulphone are determinated by HPLC

AIM: To set up a method which can determining the blood concentration of omeprazole and its metabolite 5′-omeprazole, omerpazole sulphone in order to study its cinical pharmacokinetics. METHODS: The blood concentration of omerpazole was determinated by HPLC. RESULTS: Calibrated standard curve of omeprazole in blood is Y = -0.004 499 + 0.001 909X (r = 0.9990), the recoveries of three concentrations 50, 500, 2 000 mg·L-1 are 90.36, 109.62, 108.91%, respectively; and the precisions are 9.86, 7.86, 15.52%, respectively. Calibrated standard curve of its metabolite 5′-OH omeprazole in plasma is Y = - 0. 003 659 + 0. 001 328X (r = 0.9970), the recoveries of three concentrations 20, 200, 1 000 mg·L-1 are 79.42%, 90.49 %, 93.04%, respectively; and the precisions are 8.95%, 4.52%, 9.73%, respectively. Calibrated standard curve of its another metabolite omeprazole sulphone in plasma is Y = 0.009 248 + 0.001 765X (r = 0. 999 2), the recoveries of three concentrations 20, 200, 1 000 mg·L-1 are 94.44%, 105.59%, 104.26%, respectively; and the precisions are 8.72, 8.58, 9.60%, respectively. After 20 mg omeprazole were administered by a voluteer via oral, Cmax of 5′-OH omeprazole, omeprazole, and omerpazole sulphone were 14.622 7, 408.433 2, 454.363 7 mg·L-1. CONCLUSION: The method is good enough to study pharmacokinetics of omeprazole.
4.Effects of bone marrow mesenchymal stem cells on apoptosis relative proteins of cerebral tissue in brain ischemic rat

Objective To observe the distribution of bone marrow mesenchymal stem cells(MSCs) and the effects on expression of apoptosis relative proteins Caspase 3 and Bcl-2 after intravenous transplanted into ischemic rat brains.Methods MSCs from SD rats were cultivated and proliferated in vitro and marked with CFSE.MSCs were then intravenously transplanted into middle cerebral artery occlusion(MCAO)models of SD rats.The rats were killed at different time points to observe the distribution of MSCs under fluorescence microscoDe as well as the effects on expression of apoptosis relative proteins Caspase 3 and Bcl-2 using immunohistochemical method.Results Density of Caspase 3 in immunohistochemically positive area in transplantion group were(2.81±0.35)%,(3.98±0.67)%,(5.58±0.92)%,(3.51±0.63)%,(1.64±0.29)%in 6,12,24,72 hours and in 7 days,respectively,and decreased significantly compared with those of control group[(3.92±0.44)%,(5.23±0.30)%,(6.89±0.57)%,(4.39±0.57)%,(2.29±0.21)%],the difference being significant(t=4.37,3.34,2.60,2.32,3.90,P<0.05 or<0.01).The density of Bcl-2 in immunohistochemically positive area in transplantation group were(4.70±0.16)%,(5.61±0.26)%,(3.00±0.28)%respectively in 6,12 hours and in 7 days,which had improved significantly compared with those of control group[(3.28±0.27)%,(4.54±0.59)%,(2.15±0.62)%],the difference being significant(t=8.32,3.25,2.54,P<0.05 or<0.01).Conclusions Bone marrow MSCs can exert protective effects on brain ischemia and reperfusion injury possibly by down-regulating Caspase 3 and up-regulating Bcl-2.

Objective: To observe the clinical efficacy of moxibustion therapy plus Liu's pediatric massage (tuina) for children with recurrent respiratory tract infections due to qi deficiency of spleen and lung. Methods: A total of 60 children who met the inclusion criteria were divided into an observation group and a control group according to the visiting sequence, with 30 cases in each group. Children in the observation group were treated with moxibustion therapy plus Liu's pediatric massage, and those in the control group were treated with Liu's pediatric massage alone. The incidence of respiratory tract infections and traditional Chinese medicine (TCM) symptoms score were observed and recorded in both groups before and after treatment. And the clinical efficacy was compared between the two groups. Results: The total effective rate of the observation group was 93.3%, and that of the control group was 83.3%. The difference between the two groups was statistically significant (P<0.05). After treatment, the TCM symptoms score and total times of infections in both groups were all statistically different from those before treatment (all P<0.05). The differences in TCM symptoms score and infection frequency before and after treatment in the observation group were statistically different from those in the control group (both P<0.05). Conclusion: Moxibustion therapy plus Liu's pediatric massage has a better effect in improving the clinical symptoms and reducing the frequency of respiratory tract infections for children with recurrent respiratory tract infections due to qi deficiency of spleen and lung than the pediatric massage alone.
7.The modified peroneal artery perforator-based propeller flap for the reconstruction of distal lower extremity defect

Objective To explore the feasibility and clinical effect of modified peroneal artery perforatorbased propeller flap which excludes small saphenous vein and sural nerve for coverage of the soft tissue defects of the ankle and foot.Methods From January,2009 to August,2013,20 patients with soft tissue defects of the foot or ankle underwent the procedures of reconstruction.After the proper perforators being identified with doppler sonography,the propeller flap was designed,which selected the location where terminal perforator vessels perforate deep fascia as the pivot point,the line from this point to the point where the second perforator near the wound around the line from the midpopliteal point to the midpoint of the achilles tendon and lateral malleolus perforated the deep facia as the axis.The flap was raised above the level of the deep fascia.The small extrinsic vessels around the sural nerve and small saphenous vein were contained in the flap,while the small saphenous vein and sural nerve were kept in the original position.The donor site was closed directly.The skin flaps measured from 7 cm × 4 cm to 21 cm × 7 cm.Results All 20 flaps survived completely without complications.During 10-28 months' followed-up (average 13 months),all flaps showed good texture matches and contour.All patients recovered walking and shoe wearing function.No one showed sensory disturbances at the lateral foot.Conclusion The modified peroneal artery perforator-based propeller flap excluding small saphenous vein and sural nerve still has stable blood supply,which is an idea way for covering skin and soft tissue defects in foot and ankle.
8.Expression of Ki-67 in moderate and high risk gastrointestinal stromal tumors with CD117 immunoreactive

Objective To study the expression and significance of Ki-67 in gastrointestinal stromal tumors(GIST) with CD117 immunoreactive. Methods The expression of Ki-67 was detected by SP method in 25 cases of high risk GISTS, 18 cases of moderate GISTS and 33 cases of extremely low and low risk GISTS,which were compared with the follow-up results. The relationship between Ki-67 index (LI) and risk degree was analyzed. Results Forty patients with moderate and high risk GIST were followed-up, including 26 alive,12 die of GIST and 2 die of other causes. Compared with patients of extremely low and low risk GIST, Ki-67 LI>5 % was correlated with moderate and high risk cases (P <0.01), meanwhile Ki-67 LI was positively correlated with tumor sizes of >5 cm and tumor mitotic cell count >5/50 HPF, but was not with locations(P >0.05). Conclusion Ki-67 LI>5 %, tumor size>5 cm and tumor mitotic cell count>5/50 HPF are risk indicator for GIST with CD117 immunoreactive.
9.Clinical application of special forms of perforator flaps in orthopedic trauma

Objective To investigate the clinical results,indications and precautions of a variety of special forms of the perforator flap for reconstructing limb soft-tissue defect.Methods From January 2008 to December 2013,480 cases suffering from limb soft-tissue defect were treated with special forms of the perforator flap.Types of special forms of the perforator flap included flow-through perforator flaps,micro-dissected perforator flaps,conjointed perforator flaps,polyfoliate perforator flaps and chimeric perforator flaps.Derivative types combined two or three technologies used in basic types.Results Venous crisis of the flap was shown in 10 cases.Through a second surgical exploration,5 cases were diagnosed with anastomotic thrombosis,2 with vascular torsion and 3 with hematoma.After appropriate treatments,flaps survived in 4 cases,but partial necrosis occurred in 6 cases.Twelve cases were found to have infection,which led to arterial crisis followed by total flap necrosis in one of them at postoperative day 5.Other flaps survived and wounds healed by first intention.Donor sites were repaired by skin flap transfer in 8 cases,skin grafting in 3 cases and direct suture in the rest cases.Conclusion Special forms of the perforator flap are optimal for repair of soft-tissue defect of extremities.

Objective To study the expression of Galectin-3,CK19 and Ki-67 in the papillary thyroid carcinoma and papillary thyroid hyperplasia and to find the differential diagnostic makers. Methods A total of 200 cases, including 100 with papillary thyroid carcinoma and 100 papillary thyroid hyperplasia by Immunohistochemistry. Results The positive rates for Galectin-3,CK19 and Ki-67 in the papillary thyroid carcinoma were 100 %, 97 % and 93 %, which were significantly higher than those in the papillary thyroid hyperplasia (13 %, 31 %, 1 %) (P
