Aim To study the effect of As_2O_3(arsenic trioxide) on the expressions of telomeric repeat binding factor1,2 and telomeric stability of human gastric cancer MGC803 cells, and explore the mechanism of cell apoptosis. Method MGC803 cell growth inhibition was measured with MTT assay. Apoptosis was analyzed with flow cytometry. Influence on chromosome distal end was analyzed with chromosome end-end fusion analysis. Expressions of TRF1 and TRF2 were determined with Western blot analysis. Results MTT assay showed that As_2O_3 clearly inhibited the growth of MGC803 cells, depending on time and dosage. The apoptosis rates were significantly higher than those of the control group in a concentration and time-dependent manner. These changes were not found in the control group. After disposal with 5 ?mol?L -1 As_2O_3, chromosome fusion rate was obviously increased in 48 h. After 48 h of disposal with 5 ?mol?L -1 As_2O_3, the TRF1 of MGC803 cells was up-regulated while TRF2 was down-regulated. Conclusion As_2O_3 induced chromosome fusion and MGC803 cells apoptosis through up-regulating expressin of TRF1 and down-regulating expressin of TRF2.

Chinese Journal of Cancer 2001;20(1):72-75
Objective: The current study was designed to investigate the expression of glutathione s-transferase π ( GST-π ) and its clinical significance in human primary transitional cell carcinoma(TCCs) of bladder. Methods: A total of 107 human primary bladder TCCs of the previously untreated patients were analyzed for expression of GST-π and P-glycoprotein (Pgp) by SP immunohistochemistry (IHC). Results: The GST-π positive staining were divided into three groups: single cytoplastic staining, single nuclear staining, both cytoplastic and nuclear staining. GST-π were present in 72.9% generally, GST-π cytoplastic staining (single cytoplastic, both cytoplastic and nuclear staining) were found in 58.9% of the samples, 46.7% of them were positive nuclear staining (single nuclear, both cytoplastic and nuclear staining). GST-π general positive staining and cytoplastic positive staining were significantly correlated with grade, stage and recurrence after post-operative intravesical chemotherapy. Both of them were also strongly related to the expression of Pgp. While GST-π nuclear positive staining was related to stage but not with grade and recurrence. Conclusions: Overexpression of GST-π can be used as a predictive marker of post-operative intravesical chemotherapy in TCCs. Furthermore increased cytoplastic staining is a better predictor of drug resistance while increased nuclear staining might be associated with progression in TCCs.

Abstract: Objective To analyze the surveillance of Mycobacterium tuberculosis/HIV (TB/HIV) co-infection in Liaoning Province from 2016 to 2021, providing a scientific basis for formulating future prevention and control measures applicable to the province-wide co-infection. Methods A descriptive analysis was conducted on patient registration, two-way screening, and treatment information from the annual report of patients with TB/HIV co-infection in Liaoning Province from 2016 to 2021. Results From 2016 to 2021, 222 cases of TB/HIV co-infection were registered in Liaoning Province, accounting for 0.16% (222/136 709) of TB patients and 1.42% (222/15 584) of HIV/AIDS patients. In HIV/AIDS, the symptom screening rate was 96.01%, showing a downward trend (χ2trend=982.28, P<0.05). The TB detection rate was 0.47%, also showing a downward trend (χ2trend=36.56, P<0.05), while the chest radiograph or sputum detection rate was 94.61%, showing an overall upward trend (χ2trend=96.5, P<0.05). Among TB patients, 65 188 were tested for HIV antibodies, with a detection rate of 47.68%, which showed an overall upward trend (χ2trend=3 326.38, P<0.05). There were 39 HIV-positive cases detected, with the positive detection rate in TB patients decreasing from 0.12% to 0.07%. From 2016 to 2021, the majority of co-infected patients were male and ≥15 years old, with 496 cases (92.36%) of males and 41 cases (7.64%) of females. Among the 537 patients with co-infection, 47 cases (8.75%) received anti-tuberculosis therapy alone, 72 cases (13.41%) received anti-HIV therapy alone, and 408 cases (75.98%) received combined therapy, with an overall downward trend (χ2trend=9.54, P<0.05). Conclusions Focus should be placed on the male population and those aged ≥15 years, with future efforts aimed at enhancing two-way screening work among TB and HIV/AIDS patients and improving the rate of combined treatment.
