RESULTS:Microscopy showed P. notoginseng had obvious features;P. lactiflora and Sinomenii Caulis in preparation were well-separated with clear spots. The linear range of paeoniflorin was 0.014-1.412 μg/μl(r=0.999 2);RSDs of precision, stability and reproducibility tests were lower than 2%;recovery was 95.73%-99.48%(RSD=1.54%,n=6). CONCLUSIONS:The method is simple, accurate and reproducible, and can be used for the quality control of Jinwu jiangu capsule.
7.Combination of TMPRSS2 promoter regulation of CD-5-FC gene system and phytochemical drugs effect prostate cancer 22RV1 in vitro
Chinese Journal of Urology 2017;38(3):216-220
Objective To explore the effects of TMPRSS2 promoted suicide gene CD-5-FC combined with phytochemicals on the proliferation and apoptosis of prostate cancer (PCa) specific cell-line 22RV1.Methods From March 2016 to October 2016 TMPRSS2-VISA-CD/UPRT'vector was used to deliver pro-drug 5-FC (5-Fluorocytosine) in to PCa specific cell-line 22RV1.Transfection effect was verified by Western-blotting.5-FC,5-FU (5-Fluorouracil) and four phytochemicals epigallocatechin gallate (ECGC),Genistein,Daidzein, and Equol were selected in this study.MTS assay was performed to select dosages, which can kill 10%-40% of 22RV1 cells, from their pre-set drug concentrations, respectively.According to their select concentrations, either 5-FC plasmid or 5-FU in combination with one of four phytochemicals were added in 22RV1 cells in orders, and then cultured them together.Besides, these four phytochemicals were added in 22RV1 cells individually regarding their selecting concentrations and cultured at the same time.Cell viability was detected by MTS assay on 24hrs,48hrs and 72hrs.Comparing the cell killing rates between the combination groups and each single drug groups on cell-line 22RV1.Results TMPRSS2-VISA-CD/UPRT'plasmid with 5-FC was transfected on PCa specific cell-line 22RV1 successfully. The most obvious transfection occurred on 48hrs.The selected concentration of the pro-drug 5-FC were 83.32 μmol/L and 833.20 μmol/L,5-FU were 76.87 mol/L and 768.70 μmol/L.The concentrations of four phytochemical agents were 2.00 μmol/L,10.00 μmol/L and 20.00 μmol/L.In term of the cell killing numbers, results showed that the combination treatment of 83.32 μmol/L 5-FC/ 76.87μmol/L 5-FU and 10.00 μmol/L/20.00 μmol/L four phytochemicals compared with 10.00 μmol/L/20.00 μmol/L single treatment of four phytochemicals, respectively, there was no significant difference (P >0.05),while the combination treatment of 833.20 μmol/L 5-FC/768.70 μ mol/L 5-FU and 2.00 μmol/L/10.00 μmol/L four phytochemicals compared with 2.00 μ mol/L/10.00 μmol/L single treatment of four phytochemicals, respectively, there was a significant difference (P < 0.05).In addition, this study proved that 83.32 μmol/L 5-FC and 76.87 μmol/L 5-FU,833.20 μmol/L 5-FC and 768.70 μmol/L 5-FU reached the same cell killing effect, and there was no statistical difference (P > 0.05).Thus the suicide gene transfection was successful, and its function was the same as the effect of cytotoxic drugs. Conclusions This study has proved that this plasmid with 5-FC suicide gene system can be successfully and efficiently transfected into PCa cells 22RV1.As the pro-drug of 5-FU,5-FC got similar treatment effect with 5-FU,and inhibited cell proliferation. There was no synergistic reaction of enhancing cell apoptosis after combined phytochemical drugs EGCG,Genistein,Daidzen,Equol with suicide gene therapy on 22RV1 cell-line.

Objective To assess voice parametric characteristics of normal children in different age?sex?and phonation conditions. Methods Voice data of 240 children were collected and analyzed with Dr. Speech software. Acoustic signal data were measured in three phonation conditions: the lowest modal voice, natural voice, the highest modal voice. Results Perturbations and normalized noise energy from the highest value to the lowest one by one were that of the lowest modal voice, natural voice, the highest modal voice. The sequences of standard deviation of fundamental frequency were that of the highest modal voice, the lowest modal voice, natural voice. Conclusion The voice quality of normal children had distinctive characteristics in different phonation conditions.
9.Sites of action of propofol and isoflurane on somatosensory pathway
Chinese Journal of Anesthesiology 1994;0(03):-
Objective To determine the sites of action of propofol and isoflurane on somatosensory pathway using median nerve somatosensory evoked potential(MnSSEP) .Methods Twenty-six ASA I - II patients aged 20-50 yrs were randomly divided into two groups: I propofol group( n = 13) and H isoflurane group( n = 13) .In propofol group patients received propofol infusion at a rate of 10 mg?kg-1?h-1 .Oxygen was administered via mask and respiration was assisted or controlled to maintain SpO2 at 96%-100% and PETCO2 at 35-45 mm Hg. The propofol infusion was continued until the patient failed to respond to verbal command. Six minutes later if the patient was still breathing spontaneously, the rate of propofol infusion was increased to 20-40 mg?kg-1?h-1 . In isoflurane group anesthesia was induced with propofol 2 mg ? kg-1 and intubation was facilitated by succinylcholine and anesthesia was maintained with isoflurane inhalation and intermittent iv boluses of vecuronium. The end-tidal isoflurane concentration was maintained at 0.5,1.0 and 1.5 MAC. Each concentration was maintained for 15 minutes. MAP, HR, SpO2,PCTCO2, T0(naso-pharyngeal) and BIS were continuously monitored. MnSSEP( Viking IV D type) was measured and recorded before induction of anesthesia(baseline) and in propofol group when patients became unconscious and apneic; in isoflurane group when end-tidal isoflurane reached 0.5,1.0 and 1.5 MAC.Results In group I there was no change in both latencies and amplitudes of N9 and N13'.With increasing infusion rate, propofol gradually prolonged the latencies and decreased the amplitudes of N60, P45, N35, N20 and P25 waves. In isoflurane group there was no change in the latencies and amplitudes of N9. There was no change in the latencies of N13', but the amplitudes decreased at 1.0 and 1.5 MAC. With increasing concentration, isoflurane gradually prolonged the latencies and decreased the amplitudes of P45, N60, N35, N20 and P25 waves. At 1.5 MAC the inter-peak latencies between N13'-N20 and N13'-P25 were prolonged. Conclusions The sites of action of different infusion rates of propofol and different concentrations of isoflurane are different on somatosensory pathway. The higher the doses, more widespread are the sites of action.
