Lin-Lang LIANG ; Guo-Liang LIU ;
Chinese Journal of Endocrinology and Metabolism 1986;0(03):-
The clearance of proteins differing in charge and/or size (IgG4, total IgG and albumin) was evaluated in 95 diabetic patients. Urinary IgG4 values increased in group mi-croalbuminuria. The increased urinary IgG4 excretion correlated significantly with the degree of albuminuria and diabetic retinopathy. Urinary total IgG levels increased obviously only in group with massive macroalbuminuria. The IgG4/total IgG ratio was highest in group mi-croalbuminuria, and its change had a special pattern. Urinary IgG4 and IgG4/total IgG ratio may be sensitive parameters in assessing early protein charge-selectivity impairment and in detecting incipient diabetic nephropathy.
4.Analysis of patients' awareness rate and intent for dual referral
Chinese Journal of Hospital Administration 2010;26(4):293-295
Objective To learn the awareness rate of patients for dual referral and analyze factors affecting their intent, in order to provide references for healthcare decision making. Method Data were collected with questionnaires, and analyzed with logistic regress model. Results (1) Factors for patients' awareness rate: Patients' attitude toward first treatment in community) accessibility to community healthcare centers in their vicinity. (2) Factors affecting patients' intention for dual referral: Patients' attitude toward dual referral feasibility; patients' attitude toward rehabilitation back to community if the community joins hands with a hospital; whether patients would choose the community for treatment if the community offers benefits. Conclusion Greater efforts are expected to encourage the people to embrace the practices of dual referral and first treatment in community, and to support community healthcare centers, in addition to expanding the cooperation between such centers and hospitals.
5.Inhibitory effect of tyrphostin AG114 on recombinant human protein kinase CK2 holoenzyme
Chinese Journal of Pharmacology and Toxicology 2002;16(1):8-14
AIM To study the direct effect of tyrphostin AG114 on recombinant human protein kinase CK2 holoenzyme and its kinetics. METHODS Recombinant human protein kinase CK2 α and β subunits were cloned and expressed by genetic engineering, and purified to homogeneity. The two subunits were mixed at equal molar ratio and reconstituted CK2 holoenzyme, which exerted the maximum biological activity. The CK2 activity was assayed by detecting incorporation of 32P of [γ-32P]ATP or [γ-32P]GTP into the substrate in various conditions. RESULTS The recombinant human CK2 was a second messenger (Ca2+, cAMP and cGMP) independent protein kinase, the characterization and function of the reconstituted holoenzyme were consistent with those of native CK2. AG114 strongly inhibited the holoenzyme activity of recombinant human protein kinase CK2 with an IC50 of 20.8 μmol·L-1, which lay between IC50 of 5,6-dichloro-1-β-D-ribofuranosyl-benzimidazole(DRB) and N-(2-aminoethyl)-5-chloronaphthalene-1-sulfonamide(A3), known as CK2 special inhibitors. Kinetic studies of AG114 inhibition on recombinant human CK2 showed that the inhibition was mixed competitive with GTP and noncompetitive with casein. CONCLUSION AG114 not only is an effective inhibitor of protein tyrosine kinases, but also is a novel potent inhibitor of protein kinase CK2. The recombinant human protein kinase CK2 might be used as a molecular target for simpler screening method and development of more effective inhibitors of CK2.
6.The effect of AD injection on immune functions in tumor-bearing mice
Chinese Journal of Immunology 2010;26(3):224-227
Objective:In order to study the antitumor effect of AD injection and its immunological mechanisms. Methods:Liver cancer H22-bearing mice were treated with AD injection and 7 days later to observe the effect of AD injection on the inhibitory rate of tumor, indexes of thymus and spleen, lymphocytes transformation function, phagocytosis of the peritoneal macrophage, activity of TNF,the level of IL-2 and IFN-α in serum. Results:AD injection could inhibit the growth of tumor in bearing mice significantly and promote the transformation function of lymphocytes and enhance the peritoneal macrophage phagocytosis and the activities of TNF.AD injection could improve the level of IL-2 of spleen cells and IFN-α in serum. Conclusion:These results suggested that AD injection could obviously inhibit the growth of tumor and enhance the immune functions in bearing mice.
7.Portable ECG Holter's Design Based on Dual-port RAM
Chinese Medical Equipment Journal 1993;0(06):-
Objective To transmit ECG signals through a portable dual-mcu ECG holter based dual-port RAM.Methods Dual-port RAM had two independent address, data and control buses, which allowed the two processors accessed it's resource independently at different time. The master processor buffered the acquired data on its RAM until the data accumulated to 200 Bytes. Then the 200 Bytes would be transferred into the dual-port RAM at one time. The slave processor was informed to fetch the data when it reached the flash's one whole page. Results Problems in such aspects were conquered as the port contention of dual-port RAM,limiting sampling frequency, data sharing between the master and slave processors as well as acquiring signals by the cache. Conclusion The portable dual-mcu ECG holter based dual-port RAM can be applied to transmission of ECG signals.
8.Establishment of Serum Protein Pattern Model for Screening Gastric Cancer by Surface-enhanced Laser Desorption/ionization Time-of-flight Mass Spectrometry.
Journal of Medical Research 2006;0(01):-
Objective To detect the serum proteomic patterns using SELDI-TOF-MS ProteinChip array technology in gastric cancer, screen biomarker candidates, build diagnostic models and evaluate its clinical significance in early gastric cancer. Methods SELDI-TOF-MS ProteinChip was used to detect the serum proteomic patterns of 40 patients with gastric cancer and 30 healthy people. The diagnostic models were developed and validated by discriminant analysis. Results The model composed by 3 protein peaks 8592m/z、13725m/z and 8678m/z could do the best in the diagnosis of gastric cancer. The specificity and sensitivity of it were 86.5%(26/30)and 90%(36/40)respectively.Conclusions This method show great potential for the early detection, staging before operation and screening novel and better biomarkers to early gastric cancer.

Chinese Pharmacological Bulletin 1987;0(03):-
Aim To study the effect of sodium quercetin-7-sulfate (SQMS) on activity of recombinant human phosphoinositide 3-kinase (PI3-K) p110? catalytic subunit. Methods Recombinant human PI3-K p110? catalytic subunit was expressed by gene engineering.PI3-K activity was assayed by incubating recombinant PI3-K p110? with phosphatidylinositol-4,5-bisphosphate and [?-~ ~32 P]ATP; the ~ ~32 P-radiolabeled lipids were extracted with chloroform and methanol, and assessed by scintillation counter. Results SQMS showed inhibition on the recombinant p110? catalytic subunit with IC_~50 13.14 ?mol?L~-1 .Conclusions SQMS is an inhibitor of PI3-K. The recombinant PI3-K p110? catalytic subunit might be used as a molecular target for simpler target screening more effective inhibitors of PI3-K.
