1.Study on the Quality Standard of Shilong Qingxue Effervescent Granules

Objective To establish the quality standard of Shilong Qingxue Effervescent Granules. Method Rhizoma curcuma, flos sophorae, cornus officinalis and gamdir vine in Shilong Qingxue Effervescent Granules were identified by TLC. The content of gastrodin in Shilong Qingxue Effervescent Granules was determined by HPLC. Results The TLC spots were highly clear without the interference of negative samples and were reproductive. A good linear for gastrodin was in the range of 14~70 ?g/mL (r=0.999 9). The average recovery was 97.33% (RSD=0.63%). Conclusion This method is simple, rapid and accurate, and can be used for the quality control of Shilong Qingxue Effervescent Granules.

5.Comparison and quality control between domestic and foreign hepatitis virus nucleic acid amplification technology reagents for quantitative and qualitative tests

Hepatitis virus NAT reagents are now widely used clinically. However, the qulity of domestic and foreign NAT reagents varies dramatically. The main reasons for these differences including the manufacture technique, test principle and assay procedure were discussed in this paper and current status of the quality control of the NAT reagents were also described. Finally, it was pointed out that strengthening public supervision and laboratory internal control are very important for the quality improvement of the domestic reagents.
6.Comparative observation of C5 nerve root palsy after posterior surgical treatment of multilevel cervical spondylotic myelopathy

Objective To compare the incidence of C5 nerve root palsy after laminoplasty and laminectomy with internal fixation for treating multilevel cervical spondylotic myelopathy (MCSM).Methods From January 2008 to August 2012,98 patients with MCSM were treated with laminoplasty (47 patients,group A) or laminectomy (51 patients,group B) with internal fixation.All the patients were followed up for 13-56(26.5 ± 7.9) months.In both groups,Cobb's method was applied to measure cervical lordotic angle,and Ishihara's method was conducted to measure cervical curvature index (CCI) before and after operation.The incidence of C5 nerveroot palsy was recorded and compared.Results The incidence of C5 nerve root palsy in group A was 2.1% (1/47),while 21.6 % (11/51) in group B (x2 =5.430,P < 0.05).The JOA scores in group A and group B before and after operation and improvement rate of JOA scores had no significant difference (P> 0.05).The cervical lordotic angle and CCI in group A and group B before and after operation had no significant difference (P > 0.05).The improvement rate of CCI between two groups had no significant difference (P > 0.05).All of 11 patients with C5 nerve root palsy were group B 1,and other 40 patients were group B2.The improvement rate of CCI in group B1 was significantly higher than that in group B2 [(38.7 ± 18.3)% vs.(22.1 ± 12.1)%](t =1.772,P< 0.05).Conclusions Compared with laminoplasty,laminectomy with internal fixation has a higher incidence of C5 nerve root palsy.The C5 nerve root palsy may be associated with postoperative increase of cervical lordosis angle.Moreover,tethering of the C5 root may he one of its important pathomechanisms.
7.Study on Stability of Qingrejiedu Granules

Objective: To evaluated the granules stabilities in four conditions by taking the change of Baricalin content and granules surface color as stability test markers. Methods: The content of baicalin was determined by dual wave length iso absortion method and the granules surface color was determined by the diffuse reflectrance spectroscopy. ① The granules were tested in a simulated summer environment (30 ?C , RH92.5%, 1month) and in a high intensity light condition (3000LX, 10dyas). ② The influence of humidity on the stability of the granules was investigated. ③ The expiry date of the granules was predicted by the mutivariate linear regression analysis and compared with the result of the classical constant temperature method. ④ The influences of temperature and humidity on the granules stability were etudied by orthogonal test. Results: ① There was hardly any change in baicalin content of samples in a simulated summer environment and in a high intensity light condition. ② The results showed that the decrease of baicalin content and charge of granules color were obvious when the relative humidity of environment was high and the granules color was more easily influenced by the humidity. ③ The expiry date was about 2.87 years. ④ The results indicated that both temperature and humidity had effects on the change of baicalin content. Conclusion: We tried to estimate the stability of the granules by two indexes. The results showed that Qingrejedu granules was stable in different conditions. It satisfied the stability requirements of solid dosage form.
8.Screening exonic genes of proteins in hepatocytes interacting with HBEBP2 by yeast two-hybrid technique

Objective To construct the eukaryotic expression vector of HBEBP2 gene,and screen the exonic genes of proteins in hepatocytes interacting with HBEBP2.Methods The DNA fragment of HBEBP2 was amplified by PCR.The eukaryotic expression vector pGBKT7-HBEBP2 was constructed successfully by yeast-two hybrid system 3 and then transformed into yeast cells AH109.The transformed AH109 were then mated with yeast cells Y187 containing hepatic cDNA library plasmid.The diploid yeast cells were plated on synthetic dropout nutrient medium(SD/-Trp/-Leu/-His/-Ade) containing X-?-gal for the first selection.Library plasmids pACT2-DNA were extracted and co-transformed into yeast cell AH109 together with pGBKT7-HBEBP2.Then the yeast cells were plated on synthetic dropout nutrient medium(SD/-Trp/-Leu /-His/-Ade) containing X-?-gal for the second screening to eliminate false positive clones.The real positive clones were sequenced and analyzed by bioinformatics.Results Six proteins binding to HBEBP2 were screened,including human sapiens lactate dehydrogenase D,human sapiens mitochondrion,human sapiens mannose,human sapiens aldehyde oxidase,human sapiens serpin peptidase inhibitor and other two unknown proteins.Conclusions A novel class of proteins in hepatocytes interacting with HBEBP2 has been obtained.It is presumed that HBEBP2 protein is correlated with glycosylation,lipid metabolism and cell proliferation,etc.
9.Study of the probing points of Doppler sonography in diagnosis of femoral vein reflux

ObjectiveTo discuss the influence on diagn osing of femoral vein reflux by using the different ultrasound Doppler probing points. MethodsUsing pulse wave(PW) and color Doppler flow imaging, 134 femoral veins in 67 cases of lower limb varicosity were detected at 3 different probing points to check up the deep venous reflux after Valsava maneuver. The ultrasonographic data were compared with the results of venous DSA and operation to analyze the influence on diagnosis. ResultsThe reflux data obtained from the upper point of superficial femoral vein was most correlative with the results of DSA and operation. ConclusionsThe PW is a satisfactory technique in diagnosing the femoral reflux, and the upper point of superficial femoral vein is a most reasonable probing point.

Objective: To investigate the effect of F.nucleatum(Fn) and P. gingivalis(Pg) on soluble intercellular adhesion molecule-1(sICAM-1) in peri-implant sulcular fluid(PISF).Methods: PCR was used to detect microorganism samples from PISF. According to detection of Fn and Pg, 51 titanium implants in 32 partly edentulous patients were divided into 3 groups. Clinical parameters included PD, mPLI and sICAM-1.
