1.Analysis on blood test result of voluntary blood donors in Fuling District during the past seven years

Objective To get the information about the infection condition of blood in voluntary donors in Fuling District from 2001 to 2007, so as to take measures to improve blood quality and ensure blood safety.Methods Serum ALT was detected with Reitman-Frankel assay, and HBsAg, anti-HCV, anti-HIV and anti-TP were detected with ELISA.Results A total of 72 868 serum samples from blood donors were detected, in which 5 512 (7.56%) were positive. The positive rates of HBsAg, anti-HCV, anti-HIV and anti-TP were 3.09%, 0.48%, 0.07% and 1.27% respectively, and the abnormality rate of ALT was 2.73%. There was statistical difference of positive rate among various years (from 2001 to 2007).Conclusion The positive rate of blood in voluntary donors decreases year by year. HBsAg was the highest (1.16%), followed by ALT (2.73%), anti-TP (1.27%), anti-HCV and anti-HIV. It suggests that blood quality monitoring should be strengthed to assure blood safety.
2.Analysis of the reason for blood disqualification from different donation modes and the countermeasures

Objective To identify factors for blood disqualification,so as to take appropriate measures to prevent or reduce the blood disqualification.Methods According to the Operation Standards of Blood Station[No.448(2000)]published by the Ministry of Public Health,blood qualification and disqualification was judged.The blood samples were divided into three groups:group 1 collected from the mode of planning donation without reward;group 2 from the mode of voluntary blood donation without reward;and group 3 from the combination of the above.Results Totally 63 408 u were collected during the four years,of which 2 418 were disqualified and the disqualification rate was 3.81%.The disqualification rate of group 3(4.75%)was the highest,followed by that of group 2 (4.24%)and group 1(1.82%).There was significant difference among the samples collected from different months.And the descending order of blood type of disqualified blood was as follows:type O,A, B, and AB.The existence of anti-TP antibody and ALT elevation were the two main factors for blood disqualification,which disqualification rate was 1.73% and 0.93% respectively.Conclusion It suggests that propaganda of blood donation knowledge,selection of blood donors,ID and finger print identification for them,quality control of the blood station should be enhanced so as to prevent or reduce the blood disqualification.

6.Reactive oxygen species mediate neuroprotection induced by mitochondrial ATP-sensitive potassium channel opener in rat hippocampal slices during hypoxia

AIM: To examine whether reactive oxygen species (ROS) is involved in the neuroprotection by mitochondrial ATP- sensitive potassium channel (mitoKATp) in rat hippocampal slices during hypoxia. METHODS: The technique of elec-trophysiology was used, and the latency to hypoxic depolarization (HD) and the amplitude of population spike (PS) in the stratum pyramidale of the CA1 region were measured. RESULTS: Pretreatment of the slices with diazoxide (DIA, a mitoKATP opener, at concentration of 300 ?mol/L) , prolonged the latency to HD, delayed the onset of PS disappearance and improved the recovery of PS after reoxygenation. The effects induced by DIA were attenuated by 5 - hydroxydecanoic acid (a mitoKATP blocker, at concentration of 200 ?mol/L). Pretreatment with N - 2 - mercaptopropionyl glycine (MPG, a ROS scavenger, at concentration of 500 ?mol/L), also abrogated the effects induced by DIA, while treatment of MPG alone had no effect on PS and HD. CONCLUSION: ROS participates in neuroprotection offered by mitoKATp opener during hypoxia.
7.Effects of Ursolic Acid on Cholesterol Metabolism in Hepatic Cells

Objective To explore the effects and mechanism of ursolic acid ( UA) on cholesterol metabolism in human hepatocellular carcinoma HepG2 and mouse hepatocyte AML-12. Methods HepG2 and AML-12 cells were treated with different concentrations of UA (0,10,20,40 μmol·L-1)for 24 h, then the mRNA and protein expression of cholesterol 7alpha-hydroxylase (CYP7A1) and intracellular cholesterol level was detected by RT-PCR、Western blotting and enzymatic method, respectively. Results Compared with 0 μmol·L-1 UA, 20 μmol·L-1 and 40 μmol·L-1 UA significantly increased the expressions of CYP7A1 mRNA and protein(P<0. 05), and decreased intracellular cholesterol level in HepG2 and AML-12 cells (P<0. 05). Conclusion A certain concentration of UA can reduce the level of cholesterol in HepG2 and AML-12 cells. CYP7A1 may be involved in the regulation process.

10.The quality of life evaluation of patients with rectal cancer before and after Miles' operation

Objective To understand the influence of operation to quality of life by evaluating the QoL of the patients with rectal cancer, before and after the Miles' operation. Methods To select 57 patients received Miles operation. To evaluate the quality of life of the 57 sample patients that before Miles' operation and the first, the sixth, the twelfth month after the operation by using the QoL evaluation sheet. Results The QoL score of the rectal cancer patients after operation is obviously lower than that before operation [(89.24±10.62) vs (106.32±11.2.9, P<0.01]; the QoL scores the 12th month after operation is obviously higher than before operation [(114.05±8.20) vs (106.32±11.29), P<0.05]. Conclusion The recent QoL of the patients with rectal caucer after Miles' operation was descents obviously, but the long-term QoL of the patients after operation promotes obviously.
