Objective To explore the change of glycogen synthase kinase-3(GSK-3) activity in lymphocytes in patients with mild cognition impairment(MCI).Methods The phosphorylation of GSK-3? at Ser 9 site and total level of GSK-3? in peripheral blood were measured by Western-blot in normal control group,one-year MCI group,two-year MCI group and three-year MCI group. Results The phosphorylation of GSK-3? at Ser 9 site were lower in 1-year MCI group,2-year MCI group and 3-year MCI group, compared with normal control group(all P0.05).Conclusion The activity of GSK-3 in lymphocytes in MCI patients is raised, which implies a subsidiary value of MCI diagnosis.

Objective To investigate the changes of serum endotoxin and its receptor CD 14 gene expression in multiple organs in models of acute forebrain ischemia complicated with multiple organ dysfunction syndrome (MODS), and the pathogenesis of cerebrogenic multiple organ dysfunction syndrome (CMODS). Methods 54 Wistar rats were randomly divided into normal control group ( n=6), sham-operative group ( n=8) and forebrain ischemic group ( n=40). The rats in forebrain ischemic group were randomly divided into 5 subgroups: 12, 24, 36, 48, and 72 h (8 rats in each group). The contents of endotoxin in plasma were determined after models of acute forebrain infarction established. The area density and optical density of positive staining expressing CD 14mRNA in lung, liver, intestine and kidney were analyzed for the relative content of CD 14mRNA using in situ hybridization and CMIA medical image analysis system. Results Plasma endotoxin level was markedly high at 12 hours after acute forebrain ischemia, peaked at 24 hours and somewhat decreased at 72 hours. The CD 14mRNA expression in lung, liver, intestine, and kidney tissues increased after brain ischemia, reached the peak at 24~36 h, and decreased after 48 hours. The highest change of CD 14mRNA expression was found in lung ( P

Objective To understand the prevalence rate of mild cognitive impairment(MCI) of the aged people in the communities of Taiyuan, and to provide evidence for early diagnosis of Alzheimer's disease(AD). Methods Using a simple mental state examination(MMSE),2895 aged people of over 60 years from three communities of Wanbailin in Taiyuan were selected by cluster random sampling method. The MCI elderly patients screened were investigated further by using community health status questionnaire. Results The average MCI prevalent rate of the aged people was 7.6%;Age,marital status and education background significantly affected prevalent rate(P

Objective To study the function of hepatocyte growth factor/scatter factor(HGF/SF) in the (proliferation) of colorectal cancer cells. Methods The expression of c-met,the receptor of HGF,was(detected) in Caco-2 and Colo320 cell lines by Western blot. The activation of p42/p44MAPK and p38MAPK induced by HGF in these two cell lines was observed.Observation of the effect of the inhibitor of p42/p44MAPK(PD98059),p38MAPK(SB203580) on the inhibition of HGF-induced proliferation of Caco-2 and Colo320 cells were made by Using -TdR,MTT assay.Results(1)Both cell lines(expressed) the c-met.(2)HGF activated p42/p44MAPK and p38MAPK,and 20ng/ml HGF treated cells showed maximum activity in both to be within 10min.(p42/p44MAPK,2.28?0.01;p38MAPK,2.25?0.01).(3)HGF was found to significantly increase thymidine incorporation(P

Objective To survey the status of nosocomial infection after total knee arthroplasty, analyze the risk factors of nosocomial infection and possible prevention measures. Methods Datas were collected retrospectively on 80pmients (80 knee joints) who were treated by total knee arthrophsly, the patients were divided into two groups, group A with nosocomial infection and group B without nosocomial infection.Statistic patient's age, basic diseases situation, preoperative hemoglobin content, serum albumin, operation time, blood transfusions, indwelling urethral catheter time, antibiotic treatment time of the two groups. And study the location, pathogenic bacteria and outcomes of the nosocomial infection patients. Results 10 patients occured nosocomial infection, the infected site in turn is urinary tract in 5 cases, respiratory tract 4 cases, skin infections in 1 case, the incidence of nosocomial infection is 12.5%.In noscomial infection group, patient's age, blood transfusions, operation time and postoperative indwelling urinary canal time significantly higher than no nosocomial infection group, anemia, hypoalbuminemia have relevance of nosocomial infection, there is no difference between the two groups in basic diseases situation. Conclusions The nosocomial infection after total knee arthroplasty caused by multiple factors, patient's age, hypoalbuminemia, anemia, operation time and indwelling urethral catheter time is closely related with nosocomial infection

Journal of Chinese Physician 2012;14(6):746-749
ObjectiveThe present study aimed to determine whether or not dual paroxysm proliferator-activated receptor (PPAR) agonist,WY14643,improved the dysfunctioned vascular endothelium in hypertension by reducing endothelium-derived contracting factors ( EDCFs ),and to explore the molecular mechanism it was involved in.MethodsIsometric tension in isolated thoracic aortic rings of spontaneously hypertensive rats was recorded.Endothelium-dependent contractions evoked by acetylcholine in the presence of L NAME were reduced by fenofibrate.Cyclooxygenase 1 ( COX1 ) activities were determined by analyzing the peroxidase activity of cyclooxygenase colorimetrically by using ELISA kit.ResultsCompared to the control group,WY14643 significantly decreased the vasoconstriction in aorta of the SHR rats(P=0.014).PPARα antagonist MK866 enhanced the vascular contractility of SHR rats that were incubated with 10.0μmol/L WY14643( P=0.021 ).PPARΥ antagonist GW9662 did not significantly affect the vascular contractility of SHR rats that were incubated with 10.0 μmol/L WY14643( P=0.061 ).The levels of serum PGFlα(P=0.012),2α( P =0.019) and TXB2(P=0.023) in SHR rats incubated with 10.0 μmol/L WY14643 were significantly lower than the control group, respectively.Under the condition of the existence of vascular endothelium, the expression of COX-1 in SHR rats incubated with WY14643 was significantly lower than that in SHR rats incubated without WY14643 (P=0.017).ConclusionsThose data showed that WY14643 reduced the release of EDCFs,it suggests that WY14643 protects against vascular diseases through the PPAR activators in spontaneous hypertension.
