The platinum-based chemotherapy play an important role in alleviating symptoms of advanced ovarian cancer patients and prolongation of survival time ,The multidrug resistance(MDR),however,seriously affects the chemotherapy affect, the current MDR and its Mechanism studies show that,as a phase Ⅱ detoxification enzymes ghtathione S-transferase-π(GST-π) promote tumor cell resistance to chemotherapoutic drugs by affecting the biotransformation and metabolic processes of drugs,and closely related to MDR in ovarian cancer.
2.Effect of Different Concentrations of Dextrose Transfusion on Cerebral Glucose Metabolism in Pediatric Neurosurgical Operations

Objective: To investigate the effect of different concentrations of dextrose transfusion on cerebral glucose metabolism in pediatric neurosurgical operations. Methods: Sixty anesthetized children undergoing selective cerebral tumor resection were randomly divided into three groups: Ringer lactate solution group (A, n = 20), 2.5% dextrose lactated Ringer's solution group(B, n = 20), 1% dextrose lactated Ringer's solution group (C, n = 20). All the hemodynamic data were recorded during anesthesia. The values of blood glucose and lactic acid were collected before intubation(T_a), 2 hours after opening cerebral dura mater (T_b) and the end of operation (T_c). The catheters for microdialysis were punctured into normal brain tissue. The contents of glucose and lactic acid were determined in the collected intercellular fluid by biochemical analysis instruments at T_A and T_B respectively. Results: There were no differences in heart rate(HR), mean arterial pressure(MAP) and arterial oxygen saturation (SpO_2) among three groups(P > 0.05). There were no differences in blood glucose at T_a between three groups(P > 0.05). The values of blood glucose at T_b and T_c were B group>C group>A group(P < 0.05). The values of blood glucose were higher at T_b and T_c than that of T_a in three groups(P < 0.05). There were no differences in blood lactic acid among three groups(P > 0.05). There were no significant differences in values of glucose and lactic acid in the collected intercellular fluid among three groups(P > 0.05). Conclusion: Transfusion of Ringer lactate solution or low concentration dextrose solution will increase blood glucose in pediatric neurosurgical operations, but still in the normal range, which has a little influence on blood lactic acid, glucose and lactic acid in brain intercellular fluid. It is a safe choice during neurosurgical operation.

Conventional CT plays an important role in the diagnosis, staging, radiotherapy target delineation, and prognosis of NSCLC, but it still has many limitations.PET/CT has been widely used in the diagnosis and treamtent of lung cancer.In particular, PET/CT metabolic parameters have great guiding significance in the prognosis of NSCLC patients undergoing radiotherapy, and may be superior to many clinical indices.This review summarizes the advances in the application of PET/CT in the prognostic evaluation of NSCLC patients undergoing radiotherapy.

6.Study on the Effects of Arsenic Trioxide on the Encoding Protein Expressions of Metastatic Gene in Gastric Cancer Cells of Human Body

OBJECTIVE:To study the effects of arsenic trioxide(As 2 O 3 )on the encoding protein expressions of nm23and P53in gastric cancer cells and to study the anti-canecer mechanism of which in the tumor.METHODS:A control group and a test group were set up.The expressions of gene coding proteinum nm23and P53of the gastric cancer cells cultured in vitro by the action of As 2 O 3 were determined by immunohistochemistry method.RESULTS:The expression levels of nm23and P53in gastric cancer cells have been lowered compared with the control group.CONCLUSION:As 2 O 3 can exert its anti-tumor effect by decreasing the expression of gene coding proteinum of nm23and P53.
8.Effects of moderate oxygen concentration on A549 cells and expression of TGF-β1 mRNA and CTGF mRNA in A549 cells

Objective To evaluate the effects of moderate oxygen concentration (40% O2) on A549 cells and expression of transforming growth factor-β1 (TGF-β1) mRNA and connective tissue growth factor (CTGF) mRNA in A549 cells.Methods Cultured A549 cells were seeded in 6-well plates (1 mYhole) or in 10 cm diameter dishes (5 ml/dish) with the density of 1 × 1O5 cells/ml and randomly divided into 3 groups (n =12 each) using a random number table:control group (group C),40% oxygen concentration group (group 40% O2),and 95% oxygen concentration group (group 95 % O2).A549 cells were exposed to air,40 % O2 and 95 % O2 for 48 h in C,40% O2 and 95% O2 groups,respectively.The necrosis and expression of TGF-β1 mRNA and CTGF mRNA were detected at the end of culture.Results Compared with group C,the necrosis rate was significantly increased,while the expression of TGF-β1 mRNA and CTGF mRNA was up-regulated in 40% O2 and 95% O2 groups (P < 0.05).Compared with group 95 % O2,the necrosis rate was significantly decreased,while the expression of TGF-β1 mRNA was down-regulated (P < 0.05),and there was no significant difference in CTGF mRNA expression in group 40 % O2 (P > 0.05).Conclusion Exposure to moderate oxygen concentration for a certain time can induce damage to A549 cells,but the severity is lighter than that caused by high oxygen concentration,and up-regulation of TGF-β1 and CTGF gene expression may be involved in the mechanism.
9.Three-dimensionally printed porous beta-tricalcium phosphate scaffold loading poly(lactic-co-glycolic acid)/anti-tuberculosis drug control-release microspheres:a cytotoxic evaluation

BACKGROUND: So far there is a lack of reliable biomedical evidence about the effects of three-dimensional y (3D) printed porous β-tricalcium phosphate (β-TCP) scaffold loading poly(lactic-co-glycolic acid) (PLGA)/anti-tuberculosis drug control-release microspheres on the growth and proliferation of cel s, especial y osteoblasts. OBJECTIVE: To construct porous β-TCP scaffold loading PLGA/anti-tuberculosis drug control-release microspheres by 3D printing technology and to detect its cytotoxicity. METHODS: Twenty porous β-TCP scaffolds whose aperture was 400 μm were prepared by 3D printing technology. Ten of these scaffolds were randomly selected for loading PLGA/anti-tuberculosis drug control-release microspheres, and the others were without any drugs. Then the extracts from two groups were cultured with osteoblasts for 72 hours. Afterwards, cel morphology was observed by inverted phase contrast microscope, and the absorbance value was detected using cel counting kit-8 assay. Besides, the relative growth rate of osteoblasts was calculated to evaluate the cytotoxicity of the scaffold. RESULTS AND CONCLUSION: The drug-loaded scaffold exhibited with moderate size, regular structure and uniform pores. Within 72 hours of culture in the extracts from the drug-loaded scaffold, elongated or fusiform osteoblasts appeared, with less karyopycnosis. Moreover, the drug-loaded scaffold showed slight cytotoxicity, which was classified as grade 1. In conclusion, the 3D-pinted porous β-TCP scaffold loading PLGA/anti-tuberculosis drug control-release microspheres exhibits no obvious cytotoxicity.
10.Progress in Defining Treatment Endpoint of Crohn’s Disease

Crohn’s disease( CD)is a gastrointestinal progressive granulomatous disease with unknown etiology. It is characterized as active stage alternating with remission stage,long-term course with many complications,sometimes needing surgery and having the tendency of relapse in patient ’s whole life. Therefore,it is important to define an effective therapeutic endpoint to ameliorate the natural course of CD. This article reviewed the progress in defining treatment endpoint of CD.
