Objective To investigate the mechanism of renal interstitial fibrosis after unilateral ureteral obstruction and renal protctive effect of angiotensin coverting enzyme inhibitor (ACEI),benazepril on rat kidney.Methods Twenty-four rats were randomly assigned to shame operation group (normal group),operation group, benazepril(10mg?kg -1 ?d -1 ) treatment group after unilateral ureteral obstruction. Renal tissues were examined by light and electronmicroscopy at the 2nd week after operation. Immunohistochemistry was applied to measure the expression of collagen Ⅲ,transforming growth factor ? 1 (TGF-? 1 ) and nuclear factor-kB(NF-kB) in the renal interstitium.Results In comparison with shame operation group, the expression of collagen Ⅲ , TGF-? 1 and NF-kB in the interstitium of operation group markedly increased (P

BACKGROUND:Fluoride treatment of osteoporosis has been controversial.Literatures addressing the effect of fluoride on bone bio-mechanical parameters of femur in young rats are few.OBJECTIVE:To study the effects of fluoride on bone biomechanical parameters of femur in young rats.METHODS:Ninety 2-month-old SPF Sprague Dawley rats,half male and female,were randomly divided into 9 groups:control group(young,adult and long-time)and drug-administered group(young high-fluoride,young low-fluoride,adult high-fluoride,adut low-fluoride,long-term high-fluoride and long-term low-fluoride).Rats in the control group were orally administered with physiological saline,while in the drug-administered group were given orally with different dose fluoride at the corresponding times.After experiment,rats were sacrificed under anaesthesia.Three-point bending test was performed at the left femur.The effects of fluoride on maximum load and rigidity of femur were measured.RESULTS AND CONCLUSION:Compared with young control group,the maximum load and the rigidity of femur in the young high-fluoride group were decreased by 13.18%and 13.61%,respectively(P<0.05),which had no dramatically difference in the young low-fluoride group.Compared with long-term high-fluoride group,the maximum load and the rigidity offemur in the young high-fluoride were decreased by 17.22%and 17.17%(P<0.05),which were obvious increased in the long.term low-fluoride grou by 18.33%and 19.15%,respectively(P<0.05).The maximum load and the rigidity of femur were strengthened in the adult high-fluoride and adult low-fluoride groups(P<0.05).The results suggested that young rats are more sensitive to high-dose fluoride,which can reduce bone quality in rats.The negative effects on bone quailty of rats were gradually displayed as the prolongation of the period of fluoride.
3.EFFECTS OF LOW CALCIUM DIET ON DISTAL AND PROXIMAL TIBIAL METAPHYSIS IN MALE RATS

Objective:To investigate the effect of a low calcium diet on the distal and proximal tibial metaphysis in male rats using bone histomorphometrical techniques. Methods:Forty 3-month-old male Sprague-Dawley(SD) rats, with a mean weight of 280?22g, were randomized into five groups. Group one and group two were fed a normal diet (Ca 1.0%) and a very low calcium diet (VLCD ,Ca 0.1%) respectively for one month,and the rest three groups were fed a normal diet (Ca 1.0%), a very low calcium diet (VLCD, Ca 0.1%) and a low calcium diet (LCD , Ca 0.3%) respectively for three months. All animals received double bone fluorochrome labeling prior to sacrifice. At the end of experiment, the left tibiae were harvested for bone histomorphometrical evaluations. 4.Effects of l-glutamine monofluorophosphate and alendronate sodium on trabecular bone in ovariectomized rats

AIM To evaluate whether l glutamine monofluorophosphate(MFP) together with Alendronate sodium may further increase bone mass in ovariectomized rats. METHODS Forty two 3 month old Sprague Dawley female rats were randomized into six groups:group 1 rats were sham operated(Sham),group 2 rats were ovariectomized controls(Ovx),and groups 3~6 were ovariectomized and received either 1 mg?kg -1 ?d -1 of alendronate sodium ,270 mg?kg -1 ?d -1 of calcium gluconate,5 6 mg?kg -1 ?d -1 of MFP or combination of 5 6 mg?kg -1 ?d -1 of MFP and 1 mg?kg -1 ?d -1 of alendronate sodium for 3 months. All animals received double bone fluorochrome labeling prior to sacrifice. At the end of experiment,the left tibiae were havested for histomorphometrical evaluations. RESULTS Alendronate sodium increased trabecular bone volume significantly with suppressed bone resorption and bone formation. Administrated calcium gluconate had no influence on the bone mass. MFP also could not restore cancellous bone of ovariectomized rats. Administration of both MFP and alendronate sodium increased bone mass significantly but did not increase bone mass compared with alendronate sodium. CONCLUSIONS The results indicated administration of both alendronate sodium and MFP could not further increase bone mass of ovariectomized rats.
5.Effect of clinical nursing pathway on the mechanical ventilation effect of ARDS patients

Objective To investigate the effect of clinical nursing pathway on mechanical ventilation effect of ARDS patients. Methods 59 ARDS patients with mechanical ventilation from June 2008 to December 2010 were randomly divided into the observation group (30 cases)and the control group (29 cases). The control group used the traditional care model, conventional mechanical ventilation monitoring,treatment and care according to routine measures; the observation group used clinical nursing pathway. The mechanical ventilation time, complication rate of mechanical ventilation, and the gratification level of patients in the two groups of patients were observed. Results The mechanical ventilation time in the observation group was less than the control group, and incidence of complications was lower, and satisfaction degree of patients and their families was better, the difference was statistically significant. Conclusions The clinical nursing pathway can shorten the time of mechanical ventilation, lower incidence of complications of mechanical ventilation, and it improves satisfaction degree of patients and their families.
6.Protective effects and mechanism of insulin on impairment induced by glutamine in PC12 cells

BACKGROUND: More and more researches prove that cell apoptosis could be induced by glutamine, also there are more researches on studying the indirect and direct nervous-protective effects of insulin, but the nervous-protective effects of insulin on impairment induced by glutamine, as well as its mechanism still need further investigation.OBJECTIVE: To investigate the nervous-protective effects of insulin on impairment induced by glutamine in PC12 cells, and to explore its molecular mechanism.DESIGN: A prospective controlled study based on cells.SETTING: Department of Neurology, Zhejiang Hospital; Department of Neurology of Sun Yat-wen University Hospital.MATERIALS: The study was carried out at the Laboratory of the Third Affiliated Hospital and the Experimental Animal Center of Sun Yat-sen University from March 2002 to March 2003. PC12 cells were purchased from the same animal center.METHODS: Traumatic models were made in PC 12 cells by treated with 0.5 mmol/L glutamine for 20 minutes, and the insulin of different concentration were used for protection, after 24 hours, protective effects of insulin were assessed with MTT method, Hoechst33258 fluorescence staining, DNA agar gelatin electrophoresis, meanwhile the expression of PKB/Akt protein were also detected./Akt protein in experimental group.RESULTS: The A value of50 mU/L, 100 mU/L, 200 mU/L, 400 mU/L insulin groups were 0. 214 ±0. 062, 0. 234 ±0. 067, 0. 260 ±0. 076 and 0. 265 ± 0. 069, respectively, but the value of single glutamine group was 0. 201 ± 0. 079, statistical analysis indicated that compared with single glutamine group, there were no significant difference in 50 mU/L, 100 mU/L insulin groups( P > 0.05), but 200 mU/L, 400 mU/L insulin groups were found statistically different from single glutamine group(t=-2.398,-2. 716, P < 0.05); "DNA Ladder" could not be observed in 400 mU/L insulin group by electrophoresis;It was proved that Insulin could enhance the expression of PKB/Akt protein.CONCLUSION: Insulin has nervous-protective effects on impairment induced by glutamine in PC12 cells, furthermore it also has property of anti-apoptosis, and its protective mechanism might be associated with enhancement of the expression of PKB/Akt protein.
7.SonoVue and ultrasound mediated pEGFP-N1 transfection to mouse cornea in vivo study

Objective To investigate the efficiency and safety of transfection green fluorescent protein plasmid to mouse cornea mediated by SonoVue and ultrasound.Methods Saline,plasmid+saline,plasmid+SonoVue and liposome+plasmid were injected respectively to mouse eye anterior chamber.Then the mouse eye of plasmid+saline and plasmid+SonoVue injection group were exposed to pulse wave ultrasound under 50 Hz pulse reDetition frequence,2 W/cm2 intensity and 10 minutes duration time.Fluorescence stereomicroscope was used to observe the expression of EGFP in the eye at the 1 st day,3rd day,7th day,14th and 21 st day after injection.Two mice were taken randomly from each group and were sacrificed at the 3rd day after injection.Their eyes were enucleated and made into frozen coronal sections.And fluorescence microscopy was performed to observe the type and distribution of EGFP positive cell.Tissue damage was observed in pathological section.Results EGFP was exDressed over the ocular surface in SonoVue and ultrasound group,and it obviously higher than only ultrasound exDosure group and liposome group.The expression of EGFP was not detect ed over the ocular surface in other groups.Pathological sections were not found any difference in each group.Conclusions SonoVue and ultrasound can successfully and safely transfer gene to ocular in vivo study.
8.Clinical Study on Early Acupuncture for Acute Ischemic Stroke

Objective: To observe the clinical efficacy and safety of early acupuncture in the treatment of acute ischemic stroke. Methods: In clinical randomized controlled trial, 90 subjects were divided into drug group and acupuncture group, 45 cases in each group. The neurological deficit score, Bathel index and some blood laboratory indexes were detected before and after treatment in both groups. Results: In acute stage, there were striking differences in the neurological deficit score and activities of daily living (ADL) between before and after treatments in acupuncture group (P<0.01), and between acupuncture group and drug group (P<0.01); but there was no difference in blood lipid and theology between two groups. Conclusion: Acupuncture is rather safe in the early treatment of acute ischemic stroke, and improvement of disability and the patients' quality of life.
9.Factors influence elderly patients with hip fracture sitting early away from bed after operation and countermeasures

Objective To explore the factors influencing the sitting early away from bed after operation in elderly patients having hip fractures.Method Forty-six elderly patients with hip fractures who were taking postoperative functional exercises of sitting away from bed and 32 nurses were investigated on the influential factors by a questionnaire.Results More than 52.1%of the patients believed the factors influencing their sitting early away from bed including lack of family support,worry about insufficient physical strength and fear of pains.65.6%of the nurses thought the factor was their inadequate assessment.Conclusion Such measures as enhancing family support,nutritional support,relief of pains from sitting from bed and improving nurse’s ability in rehabilitative training instruction are important for the patients to sit early away from bed after operation on hip fractures.
10.Influence of evidence-based nursing on the compliance of sitting at early stage on post-operative elders with hip fracture

Objective To investigate the compliance of evidence-based nursing on post-operative elders with hip fracture sitting at early stage. Methods A total of 108 post-operative elders with hip fracture were divided into the observation group and the control group with 54 cases in each group with random digit table. The patients in the control group sit from bed based on their state of vital signs and mental status on the 2nd or 3rd postoperative day. And those in the observation group employ evidence-based nursing on the basis of the control group. The degree of anxiety, postoperative fatigue and compliance of patients were compared. Results Two weeks after the operation, the degree of anxiety scored 40.96±5.13, postoperative fatigue scored 1.89±0.65 in the observation group, which were lower than those of the control group, 56.40±4.28, 4.55±0.71, the difference was significant (t=288.41, 412.35, both P<0.01). The rate of compliance in the observation group was 94.44% (51/54), and the rate of the control group was 77.77%(42/54). The rate of compliance in the observation group was higher than that of the control group(χ2=8.57, P<0.05). Conclusions Using evidence-based nursing on post-operative elders with hip fracture sitting at early stage is able to improve the compliance of patients. It is good for reducing the postoperative complications, improving activities of daily life and mental state, promoting rehabilitation in patients as well.
