1. ISSR analysis on genetic diversity in Phyllanthus urinaria

Objective To detect the genetic diversity of 30 populations of Phyllanthus urinaria from some provinces of China. Methods ISSR was used to analyze the genetic diversity of 30 populations of P. urinaria from Chongqing, Shaanxi, Henan, Yunnan, Guangxi, Guangdong, Zhejiang, Guizhou, Fujian, and Hainan Provinces of China. Results Twenty four primers selected from 60 ISSR primers were used for amplification and a total of 264 DNA bands were obtained, including 245 polymorphic bands and 19 common bands. At species level, the average percentage of polymorphic bands (PPB) was 92.80%. The range of the genetic similarity (GS) value was 0.579 5-0.916 7, with a higher genetic diversity. Cluster analysis showed that the P. urinaria population was roughly divided into three branches: inland branch, coastal branch, and transition branch which located in Yunnan Province; Individual populations presented obvious specificity. Conclusion The genetic diversity of germplasm resources in P. urinaria is higher.
2.Change and significance of serum HO-1/CO system level in patients with coronary heart disease complicating heart failure

Objective To compare the difference of serum heme oxygenase-1 (HO-1) and carbon monoxide(CO) levels between the patients with coronary heart disease(CHD) caused chronic heart failure(CHF) and CHD patients with normal cardiac function,and further to explore the protective mechanism of HO-1/CO system during the pathogenesis process of CHF.Methods Ninety-one patients with CHF were selected as the observation group and 72 CHD cases with normal cardiac function were taken as the control group.The concentration of HO-1 was determined by ELISA arid the Chalmer S method was used to detect serum CO concentration.The general clinical data of the two groups were recorded by the using the heart failure questionnaire.And the liver and kidney functions,blood lipids,NT-proBNP,BNP and cardiac echocardiography examination were performed.Results The serum HO-1 level in the observation group was (8.13±0.27)ng/mL,which was higher than (2.80±0.52)ng/mL in the control group;the CO level in the observation group was (0.35±0.06)mg/L,which was lower than(0.59±0.07)mg/L in the control group,the difference was statistically significant(P<0.01);the HO-1 level in the observation group was gradually increased with the increase of cardiac function grade (P<0.01);while the CO level was decreased with the increase of cardiac function grade (P<0.01).Conclusion The serum HO-11evel in the patients with CHF is highly expressed with the heart failure aggravation;endogenous CO is gradually decreased due to consumption after cardiac failure aggravation.
3. Analysis of genetic relationship and quality assessment on plants in Flemingia Roxb. ex Ait. et Ait. f.

Objective: To analyze the genetic relationship of the plants in Flemingia Roxb. ex Ait. et Ait. f. and make preliminary quality accessment, in order to provide the theoretical basis for screening the new medicinal resource. Methods: Inter-simple sequence repeats (ISSR) markers were used to analyze the genetic relationship of 14 species in Flemingia Roxb. ex Ait. et Ait. f.. Meanwhile, spectrophotometry was used to measure total flavones. Results: Thirty primers were screened out of 60 ISSR random primers and produced 367 bands totally. Among them 363 bands (98.91%) were polymorphic, four bands were owned by all; The genetic similarity (GS) was 0.5668-0.8338, showing abundant genetic diversity; F. wallichii and F. philippinensis have the closest genetic relationship, F. lineate and F. chappar have the largest genetic distance; The highest content of total flavones is in F. mengpengensis and secondary is in F. philippinensis, total content of flavones in F. wallichii is above average. Conclusion: The plants in Flemingia Roxb. ex Ait. et Ait. f. have the abundant genetic diversity. As medicinal plants, F. wallichii has the potential for replacing F. philippinensis.
4. ISSR analysis on genetic diversity in Amomi Frucius

Objective: To evaluate the genetic diversity of germplasm resources for Amomi Fructus from various habitats in different phenotypes. Methods: Inter-simple sequence repeats (ISSR) markers were used to analyze the genetic diversity and the genetic relationship among 21 samples Amomi Fructus collected from Yunnan, Hainan, Guangdong, and Fujian Provinces; At the same time, the phenotypic characters of 16 Amomum villosum samples were measured in height, stem diameter, blade number, and blade size, etc. Then cluster analysis on all populations of Amomi Fructus was carried out based on the above data. Results: Eleven primers selected from 60 ISSR-primers were used for amplification and a total of 54 DNA bands were obtained,including 22 polymorphic bands. At species level, the average percentage of polymorphic bands (PPB) was 40.74%, Nei's gerne diversity (H) was 0.116 1, Shannon's information index (I) was 0.184 2. There was a little difference of morphological characteristics. Conclusion: The genetic diversity of germplasm resources in Amomi Fructus is lower.
5.Laser scanning confocal microscopy-assisted obtain of limbal tissue for the ex vivo culture of human limbal epithelial stem cells and identification

Background Human limbal allograft transplantation or limbal autograft transplantation are the primary approaches to the severe corneal-blindness,but their application in clinic were limited because of the defects of donor material.With the development of tissue engineering technology,transplantation of in vitro cultured limbal epithelial stem cells is being an advanced management.Objective The aim of this work was to expand human limbal epithelial stem cells ex vivo under the guidance of confocal microscope and to lay the foundation for fabricating ex vivo cultured cell sheets.Methods Ten eyes of ten patients were examined with the Heidelberg Retina Tomography Ⅲ Rostock Cornea Module(HRT3-RCM)to elucidate the structure of the human corneoscleral limbus and to correlate limbal epithelial dimensions.According to the analysis of the images of limbal epithelia,the limbal tissues provided by Eye Bank of Henan Eye Institute were cut into suitable explants.Then,this study was conducted to expand limbal epithelial stem cells ex vivo on denuded amniotic membrane.The phenotypes of primary cultured cells were evaluated by morphology and immunofluorescent staining with antibodies for limbal epithelial stem cell markers (p63,cytokeratinl9)and differentiation markers(keratin 3,involucrin).This experimental procedure was approved by the Ethic Committee of Henan Provincial People's Hospital.The written informed consent was obtained from subjects before initiation of any examination.Results The palisade morphology of human limbus was imaged clearly on the laser scanning in vivo confocal microscopy and many hyperreflective cells were observed in palisade basal cells.The cell-island phenomenon was seen in the basement membrane under the laser scanning in vivo confocal microscopy.The oblique sections of limbus showed many papilla-like epithelial columns below the superficial limbal epithelia.Throughout the experiment duration,the epithelial cells grew well with the migration rates from limbal tissue (68.62± 16.94)% and the migration time(5.83 ±2.04)days,which depended on the tissue freshness.Compared with the second and forth batch of tissue,the migration rates of the third and sixth batch of tissues were significantly higher(P<0.05),and the migration time was evidently longer in the forth and sixth batch of tissue compared with the first,second,third and fifth batch(P<0.05).The positively expressing rates in the cultured corneal stem cells were 4.05% and 36.52% for p63,26.07% and 40.55% for CK19,57.88% and 40.81% for K3,64.66% and 59.19% for involucrin.Conclusion Human limbal epithelial stem cells can be successfully and purposefully obtained from the limbal tissue based on the guidance confocal miscroscope.The cultured corneal stem cells can grow well on the denuded amniotic membrane
6.Comparison of three neoadjuvant chemotherapy in treating cervical carcinoma

Objective To explore the application value of neoadjuvant chemotherapy (NACT) in treating cervical carcinoma. Methods Seventy-one cervical carcinoma patients who were divided into three groups:PF group (cisplatin and fluorouracil,23 cases) or CBP group (carboplatin,bleomycetin and cyclophosphamide, 24 cases) or TP group (paclitaxel and cisplatin, 24 cases); operation was made 14 - 21 days afterwards. The therapeutic effect, chemotherapy side-effect and the effect on operation and pathology for these three groups were analyzed and compared. Results The effective rate was 93.8%(30/32),88.6% (31/35),82.4%(28/34) in TP group,CBP group,PF group,respectively. However, the therapeutic effect had no statistics significance with age, preoperative tumor grade, pathologic type for all the three groups, the therapeutic effect had statistical significance with clinical stage. The rate which pathological examination showed no residual cancer was biggest in CBP group [CBP group was 28.6%(6/21), TP group was 4.3% (1/23), PF group was 11.8%(2/17), P < 0.05 ]. Conclusions Three NACT projects are safe and effective treatment for cervical cancer. However,each project has advantages. The NACT projects can be elected for different patient according to his illness and economy.
7.Outcomes of endoscopic submucosal dissection for colorectal large laterally spreading tumors

Objective To evaluate the efficacy and safety of endoscopic submucosal dissection for the treatment of colorectal large laterally spreading tumor. Methods ESD was applied to treat 150 cases of colorectal LST with diameter larger than 4 cm. The morphological features of LST, distribution, the clinicopathological data and the en-bloc resection rate, complete resection rate, complications were retrospectively evaluated. Results There were 87 patients with LST-granular lesions and 63 patients with LST-nongranular lesions. Colorectal LST mainly distributed in the rectum for 109 cases (72.7%), sigmoid colon for 13 cases (8.7%), descending colon for 5 cases (3.3%), transverse colon for 8 cases (5.3%), ascending colon for 13 cases (8.7%), cecum for 2 cases (1.3%). There were 23 patients with low-grade neoplasia, 104 patients with high-grade intraepithelial neoplasia, 7 with intramucosal carcinoma and 16 with submucosal carcinoma. The en-bloc resection rate and complete resection rate were 92.7% (139/150) and 89.3%(134/150). Adverse events were intra-operative bleeding in 12 patients (8.0%), postoperative bleeding in 2 patients (1.3%), perforation in 3 patients (2.0%), postoperative stenosis in 3 patients (2.0%). Conclusion Colorectal large LST-NG has higher potential for malignancy. ESD is a safe and effective method to provide en-bloc and complete resection of colorectal large LST.
8.The value of MRI in monitoring incomplete healing of the uterine incision after abdominal delivery

Objective To explore MRI manifestations of incomplete healing of the uterine incision after abdominal delivery.Methods Nine patients with clinical suspected incomplete healing of the uterine incision after abdominal delivery underwent cavitas pelvis MRI scans with 3.0T MRI.Results According to the characteristics of the MRI images,healing conditions of the uterine incisions were divides into 2 groups.GroupⅠwas showed that the uterine incision healed well,the uterine junctional zone and myometrium were continuous,and the incision scar was linear low signal intensity on T2 WI (2 cases,22%).GroupⅡwas showed that the uterine incisions healed incompletely,the uterine junctional zone and myometrium were discontinuous,and the myometrium edema was in some cases with high signal intensity on T2 WI (7 cases,78%).Conclusion MRI could directly displays incomplete healing of the uterine incision after cesarean section,provide the basis for clinical diagnosis and treatment.
9.Study on the Interaction of Gliotoxin with BSA

The interaction between Gliotoxin and bovine serum albumin (BSA) was studied by the fluo-rescence, Circular Dichroism (CD) and ultraviolet visible (UV-Vis) techniques. The fluorescent experiment showed that the intrinsic fluorescence of BSA was quenched by the binding of gliotoxin in a static quenching procedure, with an association constant of 7.2?103 L/mol and in hydropobic forces. And the CD spectrum revealed that gliotoxin effected the conformation of BSA by increased the mass of ?-helix.
10. ITS2 and rbcL sequence analysis on plants of Marsdenia R. Br. and its related species

Objective: To identify and preliminarily analyze the plants of Marsdenia R. Br. and its related species by ITS2 and rbcL sequences. Methods: The ITS2 and rbcL regions were amplified and sequenced directly. The sequence was manually calibrated using CodonCode Aligner, and splited and jointed by Contig software. The genetic distance analysis was carried out by MEGA 4.0 and the phylogenetic tree was constructed using the Neighbor-joining method. Results: The samples analyzed were well identified by ITS2 and rbcL sequences, and the authentication ability of ITS2 was superior to rbcL. The genetic relationship among the plants of Marsdenia R. Br., Dregea E. Mey., and Gymnema R. Br. was very close. Conclusion: ITS2 and rbcL sequences could distinguish Marsdenia R. Br. species very well, and the authentication ability of ITS2 is superior to rbcL. The preliminary studies suggest that the Dregea E. Mey. and Gymnema R. Br. should be classified into genus Marsdenia R. Br.
