Objective: To study the effect of tripterygium wilfordii polyglycosidium (TWP) on atopie dermatitis (AD) by detecting the expression of IL-2,IL-4,IFN-γ and IL-IO mRNA. Methods: RT-RCR was used to detect the mRNA of IL-2,IL-4, IFN-γ and IL-10 in peripheral blood mononuelear cells (PBMC)of AD cultured with different concentrations of TWP.ResuRs:(1)The expression of IL-2 was significantly different between the high concentration group and other groups(P<0.05 or P<0.01 ), and between the control group and the middle concentration group (P<0.01). (2)The expression of IL-4 was significantly different between the three different concentration groups (P<0.01 ). (3) The expression of IFN-γmRNA was significantly different between the high concentration group and the control, the low, the middle concentration groups (P<0.05 or P<0.01 ). (4)There was no significant difference in expression of IL-10 mRNA between the high concentration group and the middle concentration group (P>0.05), there was significant difference among the other groups (P<0.05 or P<0.01).Conclusion: TWP can suppress the expression of IL-2, IL-4, IFN-γand IL-10 mRNA, but the low concentration of TWP can suppress the high-level expression of IL-4 and IL-10mRNA in vitro.

Chinese Journal of Orthopaedic Trauma 2004;0(07):-
Osteoclasts are derived from pluripotent stem cells in bone marrow and spleen. They play a critical role in inflammation-induced bone loss and joint destruction because in the absence of them, bone de- struction does not occur even when inflammation exists.Synovioblasts in an inflamed joint can secrete numerous inflammatory factors, including tumor necrosis factor alpha(TNF-?)and interleukin-1(IL-1)which not only induce inflammatory reactions but also elevate osteoclast formation and function indirectly or directly through promoting RANKL expression. In this wdy the inflammatory reactions are associated with bone loss and destruction. In this article,we focus on the recent progress in study of TNF-?,IL-1 and osteoclast-target therapies in management of osteoclast-mediated inflammatory bone loss.TNF-?promotes differentiation of osteoclast precursor cells in the peripheral blood and spleen, which causes a marked increase in mature osteoclasts in a diseased joint. However, IL-I supports osteoblast survival and regulates the recombination of osteoclast cytoskeleton, which further stimulates bone resorption.Since osteoclast-target therapies may inhibit osteoclast formation and function, they are becoming more and more important for inflammation-induced bone loss and joint destruction.

With the development of medical education, the traditional cell biology teaching modes and methods need constant adjustment to adapt to the current teaching. In view of the present high-speed development of cell biology,we seriously picked some representative themes to carry out thematic teaching.Students were encouraged to read some references about the corresponding content and thought primarily before the class. After the lesson, the teacher guided students to discuss and find the answers to the questions they asked before.Participation in class discussion and homework completion accounted for 10% of the final assessment results. The thematic teaching helped to optimize classical teaching contents and frontier progress. This teaching mode not only stimulated learning interest but also fully exercised learning ability.

OBJECTIVETo investigate the semen quality and its influencing factors in preconception males in Nanjing area so as to provide some evidence for working out effective intervention measures.
METHODSTotally 687 men receiving preconceptional physical examination were enrolled in this study. A questionnaire survey was conducted among the subjects along with an analysis of their semen quality.
RESULTSThe median of sperm concentration was 63.3 x 10(6)/ml (95% CI [19.88-119] x 10(6)/ml). The median of grade a sperm was 33.03% (95% CI [19.38-55.05]%), that of grade a + b sperm was 52.08% (95% CI [39.53-69.37]%), and that of teratosperm was 91.75% (95% CI [69-100]%). The median concentration of seminal plasma PMN-elastase was 195.55 ng/ml (95% CI [76.16-3330.38] ng/ml) and that of seminal plasma zinc was 7.62 μmol/L (95% CI [1.5-23, 45] μmol/L). The positive rates of Ureaplasma urealyticum (UU), Chlamydia trachomatis (CT), and Gardnerella vaginalis (GV) were 42.4%, 0.3%, and 2.4%, respectively. The median of sperm DNA fragmentation index (DFI) of those whose wives had a history of adverse pregnancy was 20.25% (95% CI [2.15-68.25]%). Multivariate logistic regression analysis suggested that mental stress (OR 1.567, 95% CI [1.081-2.27]) and sedentariness (OR 1.772, 95% CI [1.211-2.592]) were independent risk factors for asthenospermia.
CONCLUSIONThe sperm quality of preconception males in Nanjing area is not encouraging, and it can be improved by changing undesirable lifestyle and reducing mental stress.

To investigate the effect of Tanreqing injection on immune activity of peripheral blood lymphocytes of patients with lung cancer. The peripheral blood lymphocytes of patients with lung cancer and healthy persons were separated by the density gradient centrifugation method for subsequent experiments, with those from healthy persons as the positive control. The effect of Tanreqing injection on stimulating the proliferation of lymphocytes with phytohemagglutinin (PHA) was determined by MTT method. The effect of Tanreqing injection on the lymphocyte secretions of IFN-γ and TNF-α and the subset ratio of lymphocytes cultured separately or with Tanreqing injection of different concentrations were examined by ELISA and flow cytometry (FCM) respectively. In addition, the LDH release assay was used to detect the cytotoxicity of cytotoxic T cells (CTL) and natural killer cells (NK). According to the findings, all of immunological indexes of lymphocytes from patients with lung cancer were weaker than that of healthy persons, but with the obvious increases in proliferation activity and IFN-γ and TNF-α secretions of lymphocytes co-cultured with Tanreqing Injection (P < 0.05). Among lymphocyte subsets co-cultured with Tanreqing Injection, CD3+, CD3+ CD4+ and CD3- CD16 + 56+ cell ratios notably increased, whereas CD4+ CD25+ Treg cell ratio obviously decreased (P < 0.05). In the meantime, Tanreqing injection can markedly promote the cytotoxicities of CTL and NK (P < 0.05). In conclusion, Tanreqing injection shows a significant effect in promoting the immune activity of lymphocytes from patients with lung cancer and their anti-tumor immunity.
