Objective To investigate the effects of apoptosis and related gene p21WAF1 and Bcl-2 in the tumorigenesis of primary gallbladder neoplasms. Methods p21WAF1 and Bcl-2 were detected by immunohistochemistry and apoptotic cells were stained in situ by terminal deoxynucleotidyl transferase mediated-dUTP nick end labeling(TUNAL) in 46 cases of primary gallbladder neoplasms with different histological grades. 3.Comparative Analysis of Serological and Molecular Methods for the detection of Rice Grassy Stunt Virus

Methods of ELISA, nonradioactive molecular hybridiz ation and RT-PCR were applied in the detection of rice grassy stunt virus (RGSV ). The detection sensitivity of indirect ELISA using antiserum against fusion p rotein GST-NC was 1 mg of infected leaves or 84 ng of purified virus. The metho d of dot hybridization using NC, a DIG-labelled DNA probe was 50 μg diseased l e aves, or 6 ng purified preparations. The detection endpoint of RT-PCR was 10 μg diseased leaves, or 2 ng purified virus preparation. Comparisons of sensitivit y and maneuverability were made among these methods.
4.Langerhans cell histiocytosis of the thyroid

Langerhans cell histiocytosis is an uncommon disease, involvement of the thyroid by langerhans cell histiocytosis is rare. Two cases of Langerhans cell histiocytosis of the thyroid confirmed by pathology were reported in this paper. The main clinical feature was enlargement of the thyroids. One patient had hypothyroidism. Meanwhile, the lungs and pituitaries of the two patients were affected. Langerhans cell histiocytosis of the thyroid had no typical symptoms and specific laboratory examination, and might be clinically diagnosed as chronic lymphocytic thyroiditis, benign or malignant thyroid tumors. Thyroid fine needle aspiration cytology might be useful in confirming the diagnosis. The effective treatments include systemic chemotherapy, local radiation therapy and surgical excision. The chemotherapy is still the major technological approach. Most patients can relieve partially after receiving the systemic chemotherapy.
5.Selection of Different Occluders in Transcatheter Closure of Congenital Ventricular Septal Defect in Children

Objective To discuss the methods of selecting different shapes occluder and to evaluate the feasibility,safety and efficacy of them in transcatheter closures of congenital ventricular septal defect (VSD) in children.Methods Transcatheter closures were performed in 226 children with congenital VSD,age ranging from 2 to 14 years(mean 5.62 years) under the guidance of transthoracic echocardiography(TTE) and fluoroscopy.There were 14 patients with intracristal VSD,209 patients with perimembranous VSD and 3 patients with muscular VSD.Left ventriculography and transthoracic echocardiography were performed repeatedly after the procedure to assess the effect of occlusion.The echocardiography and electrocardiography were scheduled before discharge,1,6 and 12 months for the follow-up.Results The occluders were deployed successfully in 211 patients.The successful rate was 93.4%.Thin waist shape occluders,were deployed in 7 patients;equal side shape occluders,were deployed in 191 patients;eccentric shape occluders were deployed in 12 patients,and muscular defect occluders were deployed in 1 patient.There were no complications encountered during or after closure.Conclusions It is very important in transcatheter closure of congenital in children to select different shape occluder according to pathologic characteristics.In general,equal side shape occluder is suita-ble for a large number of defect and it is easy for deployment.In some conditions,the other shape occluder may be necessary.
6.Imaging and quantitative analysis of early caries using optical coherence tomography.

OBJECTIVETo explore the efficacy of optical coherence tomography (OCT) in imaging and quantitatively evaluating early enamel caries and provide experimental evidence for the clinical diagnosis of early caries.
METHODSThe smooth surface of bovine teeth showing early artificial caries and demineralization changes was scanned by an all fiber-based OCT for images and optical profiles, which were compared with images from a polarized light microscope (PLM) and mineral loss from an atomic absorption spectrometer. Data were analyzed by paired-sample t-test and Pearson correlation analysis using SPSS software.
RESULTSThe images of enamel with early caries and demineralization from OCT coincided with those from PLM. The integrated reflectivity loss in the early caries group was significantly lower than that in the demineralized group (P < 0.05), whereas no significant difference in demineralized depth was found between the two groups (P > 0.05). A linear correlation was observed between integrated reflectivity loss and mineral loss (P < 0.05).
CONCLUSIONOCT can be used to nondestructively and accurately image early caries and quantify lesion extent and degree, suggesting the extensive use of this approach in the clinical diagnosis of early caries.

7.Chief physician Lian Yu-lin's experience on treatment of cervical spondylosis by classification.

In clinical teaching practice of over 30 years, professor LIAN Yu-lin combines physiological, pathological and anatomic knowledge with TCM treatment based on syndrome differentiation, forming own unique theoretical system. This paper introduces LIAN's experience on treatment of cervical spondylosis in detail, and location of cervical vertebra three lines and determination of needling depth and reinforcing-reducing manipulation based on clinical manifestations of various types of cervical spondylosis, and combination of long needle with short needle, deep needling with superficially needling, stressing the main point and having a definite object. This is an effective pathway for treatment of cervical spondylosis.

9.Effect of meisoindigo on proliferation of leukemic cells regulated by bone marrow stromal cells

Objective To observe the effect of meisoindigo on co-culture leukemic cells and bone marrow stromal cells.Methods Bone marrow stromal cells were cultured using bone marrow mononuclear cells from patients with leukemia,and built the co-culture leukemic cells and bone marrow stromal cells.Trypan-blue-exclusion test detected the proliferation of leukemic cells.The expression levels of membrane CXCR4 on leukemic cells were assessed by flow cytometric analysis.Results Bone marrow stromal cell of leukemia could inhibit leukemic cells proliferation,leukemic cells could be promoted their growth when exposed to low concentration (5 μmol/L) of meisoindigo.Abnormal high expression of CXCR4 in leukemic cells,and meisoindigo could markedly inhibit the expression of CXCR4 in HL-60 and del leukemic cells.Expression of CXCR4 in leukemic cells can up-regulated by bone marrow stromal cells from patients with leukemia.Conclusion Meisoindigo can down regulate expression of CXCR4 in leukeimic cells to antileukemic cells.
10.Effects of APP17-mer peptide on oxidative damage and expression of MMP-1 mRNA in cultured human skin fibrobiasts irradiated with ultraviolet light

Objective To establish an ultraviolet-irradiation damage model in cultured fibroblasts derived from human skin and to explore the potential protective effects and mechanisms of amyloid precursor protein 17-met peptide (APP17-mer peptide) on the oxidative damage and collagen metabolism in cultured fibroblasts after ultraviolet irradiation. Methods Human dermal fibroblast cultures were established by outgrowth from foreskin biopsies of a healthy donor and were irradiated by a single exposure to ultraviolet rays and cultured in a series of concentrations of APP17-mer peptide (0, 20, 40, 80 μmol/L).The activity of fibroblasts was detected by the assay of MTT. The intracellular ROS level was measured with a confocal microscope. The expression of MMP-1 mRNA was analyzed real-time quantitatively following RT-PCR. Results Primary cultures of human skin fibroblasts were established from human foreskin in DMEM supplemented with 10 % fetal bovine serum. UV irradiation depressed cellular activity and increased intracellular level of ROS (P<0.05). 40μmol/L and 80μmol/L APP17-mer peptide increased the cellular activity in both UV irradiated fibroblasts and unirradiated fibroblasts (P<0.05), however,20 μmol/L did not show such protective effects (P>0. 05). 40μmol/L APP17-mer peptide could depress the level of ROS in irradiated libroblasts. A single exposure of fibroblasts to UV irradiation resulted in 1.78 foldup-regulation of MMP-1 mRNA compared with unirradiated sample, 40μmol/L and 80μmol/L APP17-mer peptide decreased the expression of MMP-1 mRNA (P<0.05 and P<0.01, respectively).Conclusion APP17-mer peptide can enhance cellular activity under UV-induced oxidative stress and in-hibit collagen degradation in fibroblasts irradiated with ultraviolet rays. Inhibition of ROS production may be involved in the protective mechanism of APP17 peptide.
