Objective To investigate the hemodynamics and structural effect of rat endothelial progenitor cells (EPC) transplant on pulmonary artery hypertension (PAH) induced by monocrotaline(MCT) in rats.Methods EPCs were identified and marked.Twenty-one days after injection of EPCs,the pulmonary hemodynamic parame- ters,average pulmonary artery pressure (mPAP),right heart index were determined.The vascular endothelial cells and pulmonary vascular structural changes were verified by fluoresccuse microscope.Results Compared with the model,EPCs treatment(n=10) decreased mPAP significantly (mPAP,EPCs:25.9?0.7 mmHg vs model group:29.3?2.2 mmHg,P

目的探讨对老年2型糖尿病患者行空气净化前后动脉血氧分压(PaO2)和二氧化碳分压(PaCO2)的变化。方法55例老年2型糖尿病患者分为观察组(29例)和对照组(26例)。观察组患者所处疗养室每日24h进行空气净化。分别于净化前、净化后第2、3、4周抽取动脉血检测PaO2 、PaCO2 、PH值和氧饱和度。 结果空气净化4周后,观察组PaO2、PaCO2 、氧饱和度有明显改善(P<0.01),PH值无显著性变化。结论空气净化能改善老年2型糖尿病患者的PaO2 、PaCO2。
3.Sodium ferulate protects against daunorubicin-induced cardiotoxicity in juvenile rats.

OBJECTIVETo investigate the protect effects of sodium ferulate (SF) on the daunormbicin(DNR-induced cardiotoxicity in juvenile rats.
METHODSForty male juvenile SD rats were randomly divided into control group (Control), daunorubicin group (DNR), sodium ferudate treatment group (DNR + SF), sodium ferudate group (SF) (n = 10) . Juvenile rats were intraperitoneally treated with DNR (2.5 mg/kg every week for a cumulative dose of 10 mg/kg) preparation immature myocardial injury model in presence with SF (60 mg/kg) oral treat- ment for 25 days. The left ventricular pressure and its response to isoproterenol were measured using left ventricular catheter. Rat myocardium myocardial pathology specimens and ultrastructure changes were also observed. The expression of cardiac Troponin I (cTNI) was detected by Western blot and RT-PCR. Results: SF treatment could inhibit the decreasing of heart rates induced by DNR damage (P < 0.05); it could increase the left ventrivular end diastolic pressure(LVEDP), heart rate, the maximal left ventrivular systolic speed(LVP + dp/dtmax) and the maximal left ventrivular diastolic speed (LVP-dp/dtmax) responding to isoproterenol stimulation(P < 0.01); SF also could improve the myocardial ultrastructure injuries and inhibit the decreasing of cTNI expression caused by DNR damages (P < 0.05).
CONCLUSIONSF treatment could alleviate the decreasing of cardiac reservation induced by DNR damages in juvenile rats, which might be related to its reversing the effects on the cardiac systolic and diastolic function injuries and its inhibiting effects on the decreasing of cTNI expression caused by DNR. The mechanism of SF preventing daunorubicin-induced cardiotoxicity in juvenile rats is relevant to inhabited cardiac Troponin I expression.

4.Relationship between epigenetics of sperm and embryogenesis.

Epigenetics comprises the modifications made in gene expressions without changing the DNA sequence itself. Significant epigenetic changes take place during spermatogenesis and fertilization and exert direct influences on embryogenesis. This article provides an overview of the latest researches on epigenetics of male germ cells and a brief discussion on the correlation of sperm with embryogenesis in four aspects: DNA methylation, histone modification, regulation of non-coding RNAs, and genomic imprinting.

5.Prenatal nicotine exposure aggravated abnormal adrenal function and its gender differences in adult offspring rats with high-fat diet

Aim To explore the alterations of blood corticosterone (CORT) level and adrenal steroidogenic function,as well as its sex specificity in intrauterine growth retardation (IUGR) rats induced by prenatal nicotine exposure (PNE) with high-fat diet (HFD) after birth,and to make clear its mechanism through insulin-like growth factor-1 (IGF-1) signaling pathway.Methods IUGR model was established by PNE (2.0mL · kg-1 · d-1),and the offspring rats were administered with HFD until postnatal week (PW) 24 after weaning.Blood CORT concentration,adrenal steroidogenesis enzymes,expressions of IGF-1 signaling pathway and 11β-HSDs/CR system were tested.Results Compared with HFD control group,the CORT concentration in male offspring of PNE group represented a decrease trend,while an increase trend in female;the expressions of adrenal steroidogenesis enzymes (such as StAR,3β-HSD and P450cll) in male offspring decreased,while increased in female offspring (such as SF-1 and P450c21);the expressions of IGF-1 signalling pathway (IGF-1 and IGF-1R) in male offspring increased,and they significantly increased in female offspring;the expression levels of 11 β-HSD2 and GR decreased,but 11β-HSD1/11β-HSD2 ratio was enhanced in male PNE group,while in female PNE group,the corresponding gene expressions increased.Conclusions PNE could induce abnormal alterations of adrenal steroidogenic function,and exhibit apparent gender differences.The potential mechanism is related to low adrenal steroidogenesis function programming induced by nicotine and catch-up growth mediated by IGF-1 after birth.
6.Effect of human follicular fluid from endometriosis patients treated with quyu jiedu granule on mouse embryonic development.

OBJECTIVETo observe the effect of human follicular fluid (HFF) from endometriosis (EM) patients treated with Quyu Jiedu Granule (QJG) on mouse embryonic development.
METHODSCultured 2-cell mouse embryos were divided into three groups. To the medium of Group A (70 embryos), HFF from endometriosis patients treated with QJG was added; to that of Group B (60 embryos), HFF from endometriosis patients untreated with QJG, and to Group C (59 embryos), HFF from patients with fallopian tube obstruction was added. The percentages of embryos developed to 8-cell stage, morula stage and blastula stage were counted, and the early stage number and rate of high-quality embryo were measured.
RESULTSAmong the 70 embryos in Group A, 53 (75.71%) developed to 8-cell stage, 48 (68.57%) to morula stage and 45 (64.28%) to blastula stage; while in the 60 embryos of Group B, the corresponding number (percentage) were 34 (56.67%), 29 (48.33%), 21 (35.00%), respectively, showing significant differences between groups (P < 0.05). High-quality of embryo was 61 (87.14%) in Group A and 44 (73.3%) in Group B, the difference between groups also showed statistical significance (P < 0.05).
CONCLUSIONHFF from endometriosis patients is toxic to 2-cell mouse embryos, but after treated by QJG, it could elevate the quality of cultured mouse embryo in the early stage.

7.Determination of six C-Glycoside flavones and antitumor activity of water-soluble total flavonoids from Isodon lophanthoides var. gerardianus.

This research established an HPLC method for determination of six C-Glycoside flavones of warer-soluble total flavonoids from Isodon lophanthoides var. gerardianus (Benth.) H. Hara, and studied the antitumor activity of the warer-soluble total flavonoids. The HPLC system consisted of Kromasil 100-5 C18 (4.6 mm x 250 mm, 5 microm) column and a solution system of methanol, acetonitrile and 0.5% formic acid gradient elution at a flow rate of 0. 8 mL x min(-1) and the wavelength of detector was at 334 nm. The column temperature was 25 degrees C. The antitumor activity of water-soluble flavonoids was assayed using HepG2 cell as the tested cell. The linear ranges of vicenin II, vicenin III, isoschaftoside, schaftoside, vitexin, 6, 8-di-C-a-L-arabinosylapigenin were 0.25-2.53, 0.12-1.20, 0.37-3.69, 0.16-1.63, 0.19-1.92, 0.14-1.42 microg, respectively. The average recoveries (n = 6) were 99.6% (RSD 0.87%), 100.2% (RSD 2.0%), 99.6% (RSD 1.8%), 97.9% (RSD 1.5%), 98.8% (RSD 1.2%), 98.6% (RSD 1.2%), respectively. After exposure in 24, 48, 72 h, the total flavonoids showed inhibitory effect on the proliferation of HepG2 cells with IC50 as the evaluation index, the IC50 values of 1.89, 1.71, 1.51 g x L(-1), respectively. The method is quick, simple and accurate with good re- producibility, and can be used for determination of vicenin II, vicenin III, isoschaftoside, schaftoside, vitexin, 6, 8-di-C-a-L-arabino- sylapigenin in the warer-soluble total flavonoids from L lophanthoides var. gerardianus. The warer-soluble total flavonoids from L lophanthoides have inhibitory effect on the proliferation of HepG2 cells.

9.In vitro experimental study of rat cardiomyocyte injury with targeting of perfluorocarbon lipid particles.

The present study was to investigate in vitro the rat cardiomyocyte injury with targeting of home-made perfluorocarbon lipid particles with avidin-biotin interaction. Neonatal rat cardiomyocytes were cultured in vitro and divided into two groups: TNF-alpha activated group and non-activated group. Those in the TNF-alpha activated group were exposed to 200 ng/ml TNF-alpha solution for 6 hours and then cardiomyocytes in both groups were pretargeted with biotinylated ICAM-1 monoclonal antibodies, and were exposed to streptavidin, and then to homemade green fluorescently-labeled biotinylated perfluorocarbon lipid particles. Cardiomyocytes nucleus stained with Hoechst. The results were detected with fluorescence microscope. As a result, in TNF-alpha activated group, around blue fluorescent cardiomyocytes nucleus, a great amount of green fluorescent particles were found, while there were few green fluorescent particles in non-TNF activated group. It has been shown that ICAM-1 is expressed in the surface of cardiomyocytes when they are stimulated by TNF-alpha. Perfluorocarbon lipid particles associated with ICAM-1 monoclonal antibodies can be targeted to injured cardiomyocytes by avidin-biotin interaction.

To carry out the secretive expression of human laminin alpha4 chain LG4-5 module (hLNalpha4LG4-5), recombinant expression plasmid pPICZalphaA-LG45 was constructed by inserting of hLNalpha4LG4-5 cDNA into yeast expression vector pPICZaA. The hLNalpha4LG4-5 protein was expressed in GS115 Pichia yeast strain after induced by methanol, and purified target protein can obviously promote the expansion and adhesion of 293 cells.
