1.PTEN/PI3K signal pathway involved in myocardial hypertrophy mediated overloaded pressure in rats

Aim To invesitgate the effect of PTEN(phosphatase and tensin homologue deleted on chromo-some ten) /PI3K(phosphatidylinositol 3 kinase) signaltransduction pathway in hypertrophied myocardium me-diated by overloaded pressure in rats,and the effect ofangiotensionⅡ(AngⅡ) receptors(AT1,AT2) antag-onists on it.Methods Overloaded pressure hypertro-phied myocardiumratmodel was established by abdom-inal arota constriction.Thirty two male wistar rats weredivided randomly into four groups,namely sham-opera-ted group,banding group,valsartan group(bandinggroup and valsartan administation),and PD123319group(banding group and PD123319 administration).AngⅡconcentration in plasma and left ventricular myo-cardium were measured by radioimmunoassays.Cardiacindex was measured and HE staining was performedwith left ventricular myocardium,immunoprecitipationwas used to assay the protein expression and phospho-rylation of PTEN、PI3K and Akt(Protein kinase B),protein expression of?-skeletal-actin in myocardial tis-sues.Results AngⅡ concentration in serum and leftventricular myocardium tissue in banding group washigher than that in sham-operated group,valsartangroup andPD123319 group(P0.05).Conclusions PTEN/PI3Ksignal pathway is involved in myocardium hyper-trophy in overloaded pressure rat model.The up-regu-lations of PI3K/Akt and down regulation of PTEN aremediated via AT1 and can be reduced by AT1 receptorantagonists.
2.Application of light invasive treatment of endometrial in husband sperm intrauterine insemination patients

Objective To discuss the effect of light invasive treatment of endometrial in husband sperm intrauterine insemination outcomes.Methods A total of 248 patients receiving husband sperm intrauterine insemination were divided into two groups by random digits table method.The patients in light invasive treatment of endometrial group (138 cases with 246 cycles) were performed with light invasive treatment of endometrial,and the patients in control group (110 cases with 201 cycles) were not perfomed with light invasive treatment of endometrial.Human chorionic gonadotropin day of endometrial thickness,intrauterine insemination tube placed successfully or not,whether fertilization tube bloody and clinical pregnancy rates and so on were compared between two groups.Results There was no significant difference in human chorionic gonadotropin day of endometrial thickness between two groups (P > 0.05).The success rate of intrauterine insemination tube placement in light invasive treatment of endometrial group was 78.5% (193/246),which was higher than that in control group (65.2%,131/201),and there was significant difference (P < 0.05).The intrauterine insemination tube bloody rate in light invasive treatment of endometrial group was 13.4% (33/246),which was lower than that in control group (34.8%,70/201),and there was significant difference (P < 0.05).The pregnancy rate in light invasive treatment of endometrial group was 31.9% (44/138),which was higher than that in control group (18.2%,20/110),and there was significant difference (P < 0.05).There were no significant differences in ectopic pregnancy rate and abortion rate between two groups (P > 0.05).Conclusion Through light invasive treatment of endometrial the husband sperm intrauterine insemination outcomes can be improved.
3.Candida parapsilosis-c aused candidiasis characterized by verrucous nodules: two case reports

Objective To report two cases of Candida parapsilosis-caused candidiasis characterized by verrucous nodules and masses,and to assess their clinical features,diagnosis and treatment.Methods A medical history including medication history and therapeutic response was carefully collected from two male patients.Physical examination was carried out with a focus on skin lesions.Diagnosis was made according to medical history as well as physical,mycological and histopathological examination findings.Antifungal agents were given at a high enough dose in time.After lesions improved,the doses of antifungal agents were tapered gradually,and drugs were withdrawn until patients completely healed.Compound preparations containing both antifungal agents and glucocorticoids were also topically applied in the early period of treatment.Results Both patients were diagnosed as Candida parapsilosis-caused candidiasis characterized by verrucous nodules and masses,and cured after 100-day supporting treatment and antifungal treatment with oral itraconazole,etc.Conclusions Candida parapsilosis-caused candidiasis should be managed with combination therapy mainly including antifungal agents.To achieve a satisfying efficacy,antifungal agents should be given early at a high enough dose for a long enough period.
4.The Effect of Tripterygium Wilfordii Polyglycosidium on Expression of Cytokines in Patients with Atopic Dermatitis

Objective: To study the effect of tripterygium wilfordii polyglycosidium (TWP) on atopie dermatitis (AD) by detecting the expression of IL-2,IL-4,IFN-γ and IL-IO mRNA. Methods: RT-RCR was used to detect the mRNA of IL-2,IL-4, IFN-γ and IL-10 in peripheral blood mononuelear cells (PBMC)of AD cultured with different concentrations of TWP.ResuRs:(1)The expression of IL-2 was significantly different between the high concentration group and other groups(P<0.05 or P<0.01 ), and between the control group and the middle concentration group (P<0.01). (2)The expression of IL-4 was significantly different between the three different concentration groups (P<0.01 ). (3) The expression of IFN-γmRNA was significantly different between the high concentration group and the control, the low, the middle concentration groups (P<0.05 or P<0.01 ). (4)There was no significant difference in expression of IL-10 mRNA between the high concentration group and the middle concentration group (P>0.05), there was significant difference among the other groups (P<0.05 or P<0.01).Conclusion: TWP can suppress the expression of IL-2, IL-4, IFN-γand IL-10 mRNA, but the low concentration of TWP can suppress the high-level expression of IL-4 and IL-10mRNA in vitro.

From the hospital information system (HIS) of 20 national grade III-A general hospitals, 2 960 cases of herpes zoster as the research object, analyzes the relations between the general information, syndrome of traditional Chinese medicine (TCM), western medicine combined diseases, the relationship between the solar term and the incidence of herpes zoster, and the combined use of Chinese and western medicine. Among the patients with 46-65 year old has the highest percentage of diseased; admission to general outpatient clinic is the most; the most common medical payment is medicare; combined disease such as hypertension, diabetes and coronary heart disease is more common; early treatment effect of herpes zoster is better than the sequelae; summer and autumn solar term patients is hospitalized more, TCM syndrome is damp heat of liver fire; about drugs, western medicine is the most commonly used vitamin B1 and mecobalamin, traditional Chinese medicine is the most frequently used Danhong injection, combination therapy with promoting blood circulation drugs and neurotrophic drugs. Thus, herpes zoster, more common in elderly patients, with no obvious relationship between solar term, should be early diagnosis and early treatment, often with combination of Chinese traditional and western medicine treatment.

7.Electron-beam irradiation therapy for keloids: retrospective study of 578 cases

Objective To analyze the outcomes of radiotherapy for keloids by high energy electron beams and the factors influencing the treatment outcome.Methods From Jan 1998 to Jun 2012,846lesions in 578 patients received radiotherapy.The median age is 29 years old (range 5-80 years old).There are 841 lesions with postoperative radiotherapy and 39 lesions with skin-grafting.656 lesions treated within 1 day after operations.The max diameter of 348 keloids are > 5 cm.We used 6 MeV and 7 MeV electron-beam radiation therapy.The total dose ranging from 16-18 Gy/2f (interval 1 week).Treatment fields including entire keloid scars,and any suture/puncture holes with a 1 cm-margin around the lesion were used.The skin grafting patients need radiotherapy after the flap survived (about 10-15 days after the operation).The median follow-up period was 36 months (range 8-185 months).Results There are 736 (87.0%) of 846 lesions with radiotherapy effective.Other 89 (10.5%) lesions relapse in 4-33 months (median 12 months).21 lesions were of no avail.The univarate analysis shows that keloids length,keloids location,skin-grafting,interval between operations and irradiations are the influencing factors of outcomes (P =0.007,0.000,0.000,0.001).The multivariate analysis shows that keloids location and skin-grafting remained statistically significant differences (P =0.001,0.001).Most of the recurrence cases are large and bent scars.Conclusions High-energy electron-beam radiotherapy for keloids can receive good outcomes.Treatment fields flat is very important for electron-beam radiotherapy.
8.Baricitinib inhibits type Ⅰ IFN-signaling during SARS-CoV-2 infection in vitro

Abstract: Objective To explore the antiviral effect of baricitinib in the SARS-CoV-2 infection and influence on cytokine levels. Methods Calu-3 cells were infected with SARS-CoV-2 at MOI of 0.1, and the levels of inflammatory cytokines (IL-6, IL-8, TNF-α and IL-1β), interferon β (IFN-β) and interferon-stimulated gene, IFIT2 in the infected cells were analyzed by qRT-PCR methods. At the same time, Calu-3 cells were infected with SARS-CoV-2 (MOI=0.1) after being treated with baricitinib for 2 hours. Cells were collected at 0, 24, 36, and 48 hours, and analyzed for the mRNA of the above genes in the drug-treated and untreated groups. Results The mRNA levels of IL-6, TNF-a, IL-1β, IFN-β and IFIT2 in Calu-3 infected by SARS-CoV-2 cells were increased significantly. These cytokines were increased by nearly 100-fold post-infection 48 h compared with the control (P<0.000 1), and continued to increase with the infection time (P<0.001 or P<0.000 1). The increase of IL-8 mRNA level was not as significant as IL-6, TNF-α, IL-8, IL-1β, but it also showed a 2-4 folds increase. Baricitinib does not affect the level of viral RNA in Calu-3 cells after SARS-CoV-2 infection (P>0.05). However, baricitinib can significantly inhibit the up-regulation of IL-6 and TNF-α levels induced by SARS-CoV-2 infection (5.25-fold and 3.90-fold down-regulation, respectively, P<0.01), and has little effect on the levels of IL-8 and IL-1β . In addition, the drug could also significantly down-regulate the increase in IFN-β and IFIT2 levels caused by viral infection (10.51-fold and 90.78-fold down-regulation, respectively, P<0.000 1). Conclusions Baricitinib inhibits the release of inflammatory cytokines to some extent, but it drastically down-regulates the expression of interferons and interferon-stimulated genes (ISGs), and has limited antiviral effect on SARS-CoV-2. Considering that interferon signal pathways play important roles on viral infection, caution should be exercised when using baricitinib to treat COVID-19 patients.
9.Interfractional dosimetric study of target volume and organs at risk following intracavitary brachytherapy for cervical cancer

Objective To examine the interfractional dosimetric variations among inverse three-dimensional (3D) plan, forward 3D plan, and two-dimensional (2D) plan of intracavitary brachytherapy for cervical cancer, and to discuss the risk of implementing the interval plan on different implantation applicators at short time intervals.Methods Twenty-five groups of CT-guided intracavitary brachytherapy (two consecutive radiations at ≤4 d apart) plans from 11 cervical cancer patients who received radical radiation therapy in our hospital were reviewed and compared.The dwelling location and time of the first intracavitary brachytherapy plan (Plan-1) were simulated on the CT image of the second intracavitary brachytherapy to form Plan-1-S.The target coverage indices and D 2 cc of organs at risk (OARs) of Plan-1-S and Plan 2(actual plan of the second intracavitary brachytherapy) under the three planning modes were recorded and compared using the paired t-test, Wilcoxon signed rank test, and ANOVA.Results The D90, D100, and V100 of high-risk CTV were significantly lower in Plan-1-S created under the inverse mode in the actual plan (-9.11±13.46%,-13.16±18.79%, and-7.80±13.34%, P=0.002, 0.002, and 0.005, respectively).D90, D100, and V100 of the interval plan had the greatest reduction under the inverse mode (76%, 80%, and 76%, respectively).The maximum reductions in D90, D100, and V100 were 332.14 cGy (2D), 244.12 cGy (forward), and 41.76%(inverse).OAR overdose occurred most frequently under the forward mode;the rates of D90, D100, and V100 reductions accompanied by one OAR overdose were 29.41%, 37.50%, and 25.00%, and the rates of D90, D100, and V100 reductions by two OAR overdoses were 5.88%, 12.50%, and 6.25%,respectively.Overdose occurred most frequently in the small intestine (36%).Comparison of the three planning modes showed that the inverse plan had a greater reduction in each target coverage index than the 2D plan.Conclusions The simulated interval plan can significantly reduce target coverage and increase the risk of OAR overdose regardless of the planning mode and the short time intervals, and is therefore not recommended for clinical application.
10.Observation of chimeric hepatitis C virus In infected Huh7.5 cell through transmission electron microscopy

Objective To observe the morphological characteristics of HCV particles and intracel-lular ultrastructure changes in Huh7. 5 cells which was infected with chimeric HCV via transmission electron microscopy. Methods Plasmid J6/JFH encoding the full length HCV chimeric genome was transcribed to HCV RNA in vitro and the RNA was transfected into Huh7.5 cells by electroporation. Quantitative real-time PCR(qRT-PCR) was used to assay HCV copies of the supernatant of transfected cells. Indirect immunofluo-rescence was used to detect the expression of HCV proteins. The cell culture superoatant were used to infect narve Huh7.5 cells, transmission electron microscopy was used to observe morphological characteristics of vi-rus particles and intracellular ultrastructure changes in infected Huh7. 5 cells. Results qRT-PCR showed high level virus copies in supernatant of transfected cells collected in different times, indirect immuno-fluo-rescencc proved high expression of HCV NS5A proteins in the transfected cells. Large numbers of enveloped or unenveloped virus-like particles (VLPs) were observed in infected Huh7. 5 cells via transmission electronmicroscopy. We also found hyperplasia of some membrane-enclosed organelles in the cytoplasm. Several fea-tures characterizing flavivirus infected cells and a cytoplasmic inclusion of unknown origin were observed. Conclusion The chimeric HCV from in vitro cell culture system is proved to be intact virus particles which can efficiently infect Huh7.5 cells.
