2.Experimental study of c-erbB2 on the fertilization in mouse.

AIM AND METHODSThe distribution of ErbB2 in mouse testis, epididymidis, ovaries, oocyte-cumulus cells-complexes in oviducts and sperms was investigated immunohistochemically. To study the effect of c-erbB2 on mouse fertilization in vitro, various concentrations of c-erbB2 antisense oligonucleotides (c-erbB2 ASODNs) were incubated with sperms and oocyte-cumulus cells-complexes during fertilization in vitro. To explore possible mechanisms involved in fertilization, the relationship between c-erbB2 ASODNs and GABA, or dbcAMP, or verapamil during fertilization in vitro was also observed.
RESULTSErbB2 oncoprotein was observed in epithelial cells in epididymis, sperms and cumulus cells. C-erbB2 ASODNs inhibited the rate of fertilization in vitro in a dose-dependent way. The fertilization rate of the control group, low (5 micromol/L), medium (10 micromol/L), high (20 micromol/L) concentration c-erbB2 ASODNs group, and nonsense at oligonucleotides group (20 micromol/L) was 38.3%, 19.6%, 10.7%, 5.0%, and 33.8% respectively. Integral optical density immunostaining of ErbB2 in sperms was notably reduced. Medium and high concentration of c-erbB2 ASODNs notably inhibited cumulus cells adhering to inner wall of Petri dish. Treated alone with GABA or dbcAMP, the rate of fertilization was increased. Both GABA and dbcAMP partially inversed the ASODNs inhibition effect on fertilization rate, but neither of them showed significant effect on sperm integral optical density of ErbB2 immunostaining. In contrast, verapamil inhibited fertilization rate. Co-treated with c-erbB2 ASODN, verapamil showed synergic inhibiting effect on fertilization with c-erbB2 ASODN. Verapamil also inhibited the expression of c-erbB2 in sperms.
CONCLUSIONIt is suggested that c-erbB2 is closely correlated with fertilization. Ca2+ may inhibit fertilization in vitro through regulation the expression of c-erbB2 gene in sperm cells, while both of GABA and dbcAMP may affect the process of fertilization through the way other than c-erbB2 expression in sperm cells.

3.Clinical study on the treatment of postoperative recurrence of lumbar disc herniation with intervertebral fusion

Objective To explore clinical effect of different intervertebral fusion devices(cage)in treating postoperative recurrent lumbar disc herniation(LDH).Methods One hundred and forty-two LDH patients with recurrence after simple in-tervertebral disc nucleus pulpoideectomy from January 2019 to January 2021 were retrospectively analyzed.All patients were treated with combined underchannel fixation and interbody fusion and divided into a single anatomical group,two-anatomical group and a single banana group according to types and numbers of implanted cage.There were 51 patients in a single anatom-ical group,included 29 males and 22 females,aged from 39 to 65 years old with an average of(53.74±5.68)years old;body mass index(BMI)ranged from 18.62 to 28.13 kg·m-2 with an average of(22.08±2.15)kg·m-2;the interval between operation and recurrence ranged from 0.5 to 4.0 years with an average of(2.7±0.8)years;5 patients with L3,4,35 patients with L4,5 and 11 patients with L5S1;a single anatomical cage was implanted.There were 46 patients in two-anatomical group,included 25 males and 21 females,aged from 37 to 66 years old with an average of(54.52±6.02)years old;BMI ranged from 18.25 to 28.44 kg· m-2 with an average of(21.74±1.83)kg·m-2;the interval between operation and recurrence ranged from 0.5 to 5.0 years with an average of(2.7±0.9)years;4 patients with L3,4,32 patients with L4,5 and 10 patients with L5S1;two-anatomical cages were im-planted.There were 45 patients in a single banana group,included 22 males and 23 females,aged from 38 to 65 years old with an average of(54.49±6.45)years old;BMI ranged from 18.85 to 28.20 kg·m-2with an average of(21.63±1.59)kg·m-2;the interval between operation and recurrence ranged from 0.5 to 5.0 years with an average of(2.6±1.0)years;3 patients with L3,4,36 patients with L4,5 and 16 patients with L5S1;a single banana cage was implanted.Operation time,intraoperative blood loss,incision length,postoperative incision drainage volume,hospital stay and complications among 3 groups were observed and compared.The height of intervertebral space before and after operation,curvature of lordosis and the postoperative interverte-bral fusion were compared.Visual analogue scale(VAS)and Oswestry disability index(ODI)were used to evaluate degree of lumbar pain and lumbar function before operation,1 and 6 months after operation,respectively.Results All patients among 3 groups were followed up at least 6 months,and no cases were fell out.There were no significant difference in operation time,in-traoperative blood loss,incision length,postoperative incision drainage volume and hospital stay among 3 groups(P>0.05).At 6 months after operation,the height of intervertebral space in two-anatomical group and a single group were[(11.08±1.78)mm,(10.95±1.62)mm],curvature of lordosis were[(12.05±1.86)°,(11.63±1.57)°],which were higher than those in a sin-gle dissection group(10.14±1.54)mm,(10.92±1.45)°,and the difference were statistically significant(P<0.05).The inter-body fusion rate between two-anatomical and a banana group(95.65%,95.56%)were higher than that in a single anatomical group(78.43%)at 6 months after operation(P<0.05).VAS and ODI of lumbar among 3 groups were decreased at 1 and 6 months after operation(P<0.05).There was no significant difference in complications among 3 groups(P>0.05).Conclusion The three fusion devices could achieve significant results in treating postoperative recurrence of LDH,but the implantation of two-anatomical cage and a single banana cage are more helpful to maintain the height of intervertebral space and lordosis cur-vature of patients with postoperative recurrence of LDH,and obtain good intervertebral fusion results.
4.Association of Claudin-1,Ki-67,and CD34 expressions with astrocytic tumor cell proliferation and invasion

Objective To investigate the correlation ofClaudin-1, Ki-67 and CD34 expressionsin the tumor tissue to the invasiveness, grading and proliferation of astrocytic tumors. Methods In 50cases of astrocytic tumor, the expressions of Claudin-1, IO-67 and CD34 were detectedimmunohistochemically in the tumor tissue, normal brain tissues and in the junctional area between thetumor and normal tissues. The expressions of Claudin-1, Ki-67, and CD34 were analyzed in relation tothe clinicopathoiogical characteristics of the patients. Results The expression rate of Claudin-1 was70.59% in the astrocytic tumors of histological grades Ⅰ and Ⅱ, significantly higher than that in grade Ⅲand Ⅳ tumors (9.09%, χ2=20.206, P=0.000). In grade Ⅲ and Ⅳ tumors, the positivity rates of Ki-67 inthe normal tissues, junctional area and the tumor tissue were all significantly higher those in grade Ⅲ andⅣ tumors (t=9.144, 5.958, and 6.297; P=0.000, 0.000, and 0.000, respectively). Grade Ⅲ and Ⅳ tumorsdisplayed significantly greater mierovessel density (MVD) than grade Ⅰ and Ⅱ tumors in the tumor tissue(t=3.101, P=0.003) and the junctional area (t=4.139, P=0.000). In all the tumors, significantly higherKi-67 expression was seen in the tumor tissue (12.74±6.93) than in the junctional area (7.42±3.93) andthe normal tissue (3.22±1.57) (F=51.726, P=0.000), and the MVD was also greater in the tumor tissue(27.48±8.26) than in the junctional area (10.72±3.93) and normal tissue (6.48±1.45) (F=215.538, P=0.000).Conclusion Expressions of Claudin-1, Ki-67, and CD34 are closely correlated to the proliferation andinvasion of astrocytic tumors, and these cytokines may serve as important indicators for evaluating themalignancy of astrocytic tumors..
5.Tramadol inhibits c-fos expression in spinal cord dorsal horn and serum IL-6 levels induced by plantar incision in rats.

OBJECTIVETo investigate effect of tramadol on c-fos expression in spinal cord dorsal horn and serum IL-6 levels induced by plantar incision in rats.
METHODSThe Brennan pain model was induced by incision on the planter surface of left hind paw in rats. Forty-eight rats were randomly divided into six groups: Sham group (Group C), control group (Group I,pretreatment with saline 5 ml), three tramadol pretreatment groups (Group T1, T10 and T20,pretreated with 1 mg/kg, 10 mg/kg and 20 mg/kg tramadol, respectively) and one tramadol treatment group (Group PT10, treated with tramadol 10 mg/kg immediately after operation). Pain behavior was assessed by withdrawal threshold to von Frey filament stimulation intensity, response latency of the hind paw to radiant thermal and a cumulative pain score 2 h after incision. Fos-positive neurons in spinal cord were identified by the immunohistochemical technique. Serum IL-6 levels were measured by enzyme-linked immunosorbent assay (ELISA).
RESULTSWithdrawIal threshold to von Frey filament stimulation intensity and response latency of the hind paw to radiant thermal in Group I were significantly lower than those in Group C (P<0.01). Cumulative pain score in Group I was significantly higher than that in Group C (P<0.01). In Groups of T10 and T20, withdrawal threshold to von Frey filament stimulation intensity and response latency of the hind paw to radiant thermal were significantly higher than those in Group I (P<0.01), cumulative pain score was significantly lower than that in Group I in a dose-dependent manner (P<0.01), and were also those in Group PT10. The greatest density of Fos-positive neurons was located in lamine I-II in Group I. Serum IL-6 levels were significantly elevated in Group I. Pretreatment with tramadol showed a dose-depended inhibitory effect on c-fos expression and serum IL-6 production,but not in Group T1. Administration of tramadol postoperatively also suppressed the c-fos expression and serum IL-6 production as showed in PT10 but were weaker than those in Group T10.
CONCLUSIONPretreatment with tramadol can produce dose-dependent inhibitory effect on c-fos expression in spinal cord dorsal horn and then suppress the inflammatory response to the trauma.

6.Correlation of the contents of trace elements in male body fluids with sperm quality.

OBJECTIVETo explore the contents of trace elements in the blood and seminal plasma of men and their correlation with semen parameters.
METHODSWe detected and analyzed the contents of calcium, magnesium, copper, zinc, iron and lead in the blood and semen of 113 men with the BH-5100 5-channel atomic absorption spectrometer, YY-1001 blood lead instrument and Weili color sperm automatic analyzer to investigate the correlation of the contents of trace elements with quality.
RESULTSThe contents of lead, copper, zinc and iron in the blood were significantly correlated with those in the seminal plasma, but not the contents of calcium and magnesium. The contents of lead, copper, zinc, calcium, magnesium and iron in the blood were significantly different from those in the semen. In the seminal plasma, the content of calcium showed a significant positive correlation with sperm motility, linear motile sperm motility (LMSM), straight line velocity (VSL), curvilinear velocity (VCL), mean angle of deviation (MAD), active sperm density (ASD), average path velocity (VAP) and lateral head amplitude (ALH), while that of lead was negatively correlated with sperm motility, LMSM, ASD, motile sperm density (MSD) and linear motile sperm density (LMSD), and so was that of magnesium with sperm motility, LMSM and LMSD, and those of calcium, magnesium and iron with seminal pH. In the blood, the content of zinc was negatively correlated with sperm motility and LMSM, and so was that of magnesium with sperm motility, LMSM and VAP, while that of lead was positively correlated with ALH.
CONCLUSIONThe contents of trace elements in different body fluids are differently correlated with sperm quality.

7.Studies on the coumarins in the root of Zanthoxylum dimorphophyllum.

OBJECTIVETo study the chemical constituents of the dried roots of Zanthoxylum dimorphophyllumr. spinifolium and to find out the active constituents of the plant.
METHODModern chromatography was used to purify chemical constituents, and their structures were identified by various spectral methods.
RESULTFour compounds were isolated and identified as isopimpinellin (I), xanthoxyletin (II), 6-(2', 3'-dihydroxy-3'-methyl-butyl)-7-hydroxy-2H-1-benzopyran-2-one (III), 6-(2'-O-beta-D-glucopyranosyloxy, 3'-dihydroxy-3'-methybutyl)-7-hydroxy-2H-1-benzopyran-2-one (IV).
CONCLUSIONAll of the above compounds were isolated from the above mentioned plant for the first time.

RESULTFive compounds were isolated and identified as 6-(2', 3'-dihydroxy-3'-methyl-butyl)-7-hydroxy-5-methoxy-2H-1-benzopyran-2-one (I), 6-(2',3'-dihydroxy-3'-methyl-butyl)-7-methoxy-8-(3'-methyl-but-2'-enyl)-2H-1-benzopyran-2-one (II),6-(2',3'-dihydroxy-3'-methyl-butyl)-7-hydroxy-2H-1-benzopyran-2-one (III), 6-(2', 3'-oxiranyl-3'-methyl-butyl)-7-methoxy-8- (3-methyl-but-2-enyl)-2H-1-benzopyran-2-one (IV), 7-methoxy-8-(3'-methyl-but-2'-enyl)-2H-1-benzopyran-2-one (V).
CONCLUSIONThese compounds were isolated from the plant for the first time.

9.Expression of c-fos in various functional state of rat ovaries.

AIM AND METHODSThe method of labeled streptavidin biotin was used to study the expression of c-fos in various functional state of rat ovaries and its relationship with the levels of serum estradiol and progesterone.
RESULTS(1) In the mature rat, c-fos expression was found higher in interstitial gland and stroma of proestrous ovaries and lower in estrous ovaries, and was found higher in luteal cells and stroma of pregnant ovaries and lower in diestrous ovaries. There was a positive correlation between the area and optical density of c-fos expression and the levels of serum E2 and P. (2) In the immature rats, c-fos expression was not found in the ovaries containing preantral follicles from DES-treated rats,and was found both in interstitial gland and stroma of the ovaries containing preovulatory follicles from PMSG-treated rats ,and the expression was higher in day 4 luteinized ovaries and lower in day 9 luteinized ovaries from PMSG with hCG treated rats. There also was a positive correlation between the area and optical density of c-fos expression and the levels of serum E2 and P.
CONCLUSIONThe results suggested in rat ovaries and might play an important role in follicles development, ovulation, luteum formation and regression.

10.Intrathecal injection of ketamine and clonidine for chronic neuropathic pain model in rats.

OBJECTIVE:
To observe the effect of intrathecal injection of ketamine and clonidine for chronic constriction injury (CCI) in rats.
METHODS:
Thirty-two SD male rats weighing 220-280 g were anesthetized with intraperitoneal chloral hydrate 300 mg/kg. A catheter was implanted in the subarachnoid space at the lumbal region and CCI rat models were made successfully. On the 4th day after the surgery, the rats were randomly divided into 4 group: a control group,injecting 0.9%NS 20 microL intrathecally; a ketamine group, injecting ketamine 1 mg/kg(20 microL) intrathecally; a clonidine group (CL), injecting clonidine 20 microg/kg (20 microL) intrathecally; a combined ketamine and clonidine group, injecting ketamine 0.5mg/kg and clonidine 10 g/kg (20 microL) intrathecally, once a day for 1 week. BME-410A Plantar Analgesia Tester was used to measured pain threshold before the administration and 30 min after the administration. The rats were killed after the test was finished. And then we detected the nitric oxide synthase (NOS) activity and the NO production in the spinal cord.
RESULTS:
The combined injection of ketamine (0.5mg/kg)and clonidine(10 g/kg) produced significantly more potent analgesia than the injection of ketamine (1 mg/ kg) or clonidine (20 microg/ kg)alone. The NOS activity and the production of NO in the combined injection group were significantly lower than those of the single injection group (P<0.05). The weight of rats post-administration increased obviously in the 4 groups (P<0.05).
CONCLUSION
The combined injection of ketamine and clonidine can produce synergistic ab-irritation without obvious side effects.
