Objectives To evaluate the relationship between the expression of B7-1 in liver tissue and the effect of interferon- alpha(α- IFN) treatment in patients with chronic hepatitis B(CHB).Methods The expression of B7 -1in liver biopsy specimens from 68 CHB patients was studied with immunohistochemistry before α-IFN treatment.Results B7-1 was expressed in 45(66.2%) liver tissues among 68 patients with CHB,but none in 5 normal controls.The total response ratio of α- IFN in patients with B7-1 positive was 66.7%(30/45),which was significantly higherthan 39.1%(9/23)in the patients with B7-1 negative(x2 =7.20,P <0.01).B7-1 expression was closely corelat-ed with the histological activity grade(HAI) and serum alanine transaminase(ALT) level.Conclusions The level of B7-1 expression in liver tissue may be regarded as an effective parameter for predicting α-IFN response in patients with CHB.
4.Studies of three fields lymphadenectomy for patients with thoracic segments squamous cell carcinoma of esophagus

Objective To investigate the difference of complication incidence, death rate, quantity of lymphadeneetomy, lymphatic metastasis rate and long-term survival rate in thoracic squamons cell carcinoma of esophagus between three fields lymphadenectomy (3-FL) and traditional method. Methods Homoehronous 96 esophageal cancer patients were fractionated in two groups, 46 patients for 3-FL, the other 50 patients for traditional method. Results The average quantity of lymphadenectomy was 39.28 pieces per patient in 3-FL, and was significantly higher than 13.30 pieces per patient in traditional method (P <0.01). The rate of metastatic lymphatic nodes was 73.91%(34/46) in 3-FL patients, significantly higher than38.00%(19/50) in traditional method patients. For incidence of postoperative complications, recurrent laryngeal nerve damage and respiratory complication in 3-FL patients was significantly higher than patients in traditional method (P <0.05). The chest fluid obviously inereasod in 3-FL patients than in traditional method patients. There was significant contrast in 3-year survival rate between 3-FL patients and traditional method patients. Conclusion The there fields lymphadenectomy expand scope of lymphdenectomy effectually, accurate the staging of thoracic esophageal carcinoma. At the same time, a lot of evidence was found in raising survival rate for 3-FL. Disadvantage of 3-FL was severe surgical trauma, high incidence of complication, and a long recovery time after operation.
5.A revascularization model of chick embryo chorioallantoic membrane carrying hydroxyapatite

BACKGROUND: Using the rapid growth period of the chorioallantoic membrane from the 8th day to the 12th day, vascular growth at bone defect end and vascular implantation of a bone graft substitute into the human body can be simulated.OBJECTIVE: To load hydroxyapatite into a chick embryo chorioallantoic membrane model, and to establish an implanted revascularization model.METHODS: Twelve rosette eggs were incubated under the same suitable conditions for 8 days. After windowing, the eggs were randomly divided into two groups: experimental group was implanted with hydroxyapatite material to establish the model of chick embryo chorioallantoic membrane carrying hydroxyapatite; control group was implanted with filter paper material, sealed and placed into a constant temperature and humidity box to continue to hatch. At the 12th day of incubation, length of new vessels and length of new vessels per unit area in the two groups were observed, and the angiogenesis on the hydroxyapatite and filter paper was observed.RESULTS AND CONCLUSION: The mean length of new vessels and length of new vessels per unit area were 24.031 mm and 0.242 mm/mm2 in the experimental group, and 23.561 mm and 0.212 mm/mm2 in the control group, respectively. There were no significant differences between two groups. Additionally, vascular tissues were obviously observed on the hydroxyapatite samples in the experimental group. These experimental results show that the establishment of the model of chick embryo chorioallantoic membrane carrying hydroxyapatite provides a simple and accurate in vitro animal model for studies on ideal bone graft materials and angiogenesis. The hydroxyapatite has no effect on the angiogenesis on the chorioallantoic membrane.
6.The analysis of the allergens in 576 patients with allergic rhinitis in Qingyang of Gansu Province.

OBJECTIVE:
To investigate the allergens distribution of 576 allergic rhinitis patients in Qingyang, and to provide basic epidemiologic information for the prevention and treatment of allergic rhinitis.
METHOD:
Skin prick test was done to all the 576 patients with allergic rhinitis with 28 kinds of allergens.
RESULT:
Four hundred and eighty cases (83.3%) showed positive reaction to at least one allergen of 28 allergens. The most common allergens were Magwort (73.3%), Giant Ragweed (55.0%) Tree II (51.7%), Tree I (48.3%) Dermatophagoides farinae (43.3%) and Dermatophagoides pteronyssinus (36.7%). Moreover. the positivity decreased with age. There was no difference between male and female.
CONCLUSION
The study shows that Magwort. Giant ragweed and tree II are the most important allergens on Qingyang district.

7.Role of catecholamine hormone in heroin addicts.

OBJECTIVETo investigate the effects of catecholamine hormone on the blood and brain of heroin addicts.
METHODSRats were divided into three groups and treated with the glucose (control group), the heroin (im) (heroin group), and the combination of the intramuscular injection of reserpine and heroin (reserpine group). Changes in the levels of the dopamine (DA), cAMP, and cGMP were detected by the radioimmunoassay (RIA) method in the blood and brain tissue.
RESULTSNo significant withdrawal symptoms were observed in the reserpine group. Compared with the control and heroin groups, the blood cAMP levels were increased by 35.36% and 15.53% in the reserpine group, respectively; the cAMP levels in the midbrain ventral tegmental area (VTA), prefrontal cortex (PFC), and hippocampus (Hipp) were increased by 24.08% & 8.53%, 15.66% & 8.13%, and 21.95% & 8.40%, respectively. While compared to the control and heroin groups, the DA levels of the PFC, Hipp, striatum, and nucleus accumbens (NAc) were significantly reduced in the reserpine group, decreasing by 74.09% & 82.86%, 81.06% & 82.23%, 91.62% & 86.55% and 84.35% & 90.63%, respectively. The concentrations of cGMP of the brain tissues in the reserpine group were lower than those in the control group. In addition, the neural electrophysiological testing showed that the electroencephalogram (EEG), electrocardiogram (ECG), and muscle spindle discharge diagram of rats in both the reserpine and heroin groups were apparently changed.
CONCLUSIONCatecholamine hormone plays an important role in heroin addiction.

8.Construction of eukaryotic expression vector of E4F1 and interactions between E4F1 and p53

Objective To construct eukaryotic expression vector of wild type E 4F1 and the mutant deleting amino acid region 32-81, and to detect the interaction between wild type or mutant E 4F1 and p53 and to study the effect of E4F1 on the expression level of p21.Methods Wild type and mutant sequences of E 4F1 were amplified from the mammary library using standard PCR and recombinant PCR .The sequences were cloned into pXJ 40-MYC vector to generate the MYC-E4F1 and MYC-E4F1(Δ32-81) recombinant plasmids that were transfected into 293T cells and identified by Western blotting . FLAG-p53 and MYC-E4F1 or MYC-E4F1(Δ32-81) were co-transfected into 293T cells and immunoprecipitation assay was performed to detect the interaction of wild type or mutant E 4F1 with p53.Wild type and mutant E4F1 expressing vec-tors were co-transfected into osteosarcoma U2OS cells and the expression of p21was detected.Results Recombinant plas-mids of MYC-E4F1 and MYC-E4F1(Δ32-81) were successfully constructed.Both wild type and mutant E4F1 interacted with p53.Deletion of amino acid region 32-81 of E4F1 increased the interaction .The expression level of p21 was in-creased by wild-type E4F1, but not by mutant E4F1.Conclusion The eukaryotic expression vector of wild type E4F1 and its deletion mutant is successfully constructed .Both of them interact with p53.Deletion of amino acid region 32-81 of E4F1 increases the interaction .This study contributes to further studies on the regulation and mechanism of E 4F1 on p53.

10.Clinical observation of acupuncture plus Frenkel exercises for ataxia after cerebral stroke

Objective: To observe the clinical efficacy of mind-refreshing and balance-restoring needling method combined with Frenkel exercises in treating ataxia after cerebral stroke. Methods: The recruited 120 patients were randomized into an observation group and a control group, with 60 cases in each group. The control group was intervened by mind-refreshing and balance-restoring needling method, while the observation group was given additional lower-limb Frenkel exercises. Before and after treatment and at the follow-up, the ataxic lower-limb function was scored using Berg balance scale (BBS) and international cooperative ataxia rating scale (ICARS), and Barthel index (BI) was adopted to score the activities of daily living (ADL). Results: After treatment, the markedly effective rate was 70.2% and the total effective rate was 96.5% in the observation group, versus 39.7% and 87.9% in the control group, and the differences in the markedly effective rate and the total effective rate were statistically significant (P<0.01, P<0.05). The intra-group comparisons showed that the BBS, ICARS and BI scores after treatment and at the follow-up were significantly different from those before treatment in both groups (all P<0.01).There were significant differences in the BBS score between the two groups after treatment and at the follow-up (P<0.05, P<0.01); the between-group differences in the ICARS and BI scores were statistically insignificant after treatment (both P>0.05), while the between-group differences in the ICARS and BI scores were statistically significant at the follow-up (both P<0.05). The interaction effects between the scoring time of BBS and BI and the group factor were statistically significant (P<0.01, P<0.05). Conclusion: Mind-refreshing and balance-restoring needling can effectively improve the lower-limb ataxic symptoms and ADL after stroke; when combined with Fenkel exercises, this needling method can produce more significant efficacy.
