2.Prostaglandin E1 Combined with the Hepatocyte Growth-promoting Factor in the Treatment of Patients with Chronic Hepatitis

Objective:To observe the efficacy and safety of PGE1 combined with PHGF in the treatment of patients with moderate or severe chronic hepatitis.Methods:110 patients with chronic hepatitis were randomly divided into two groups:60 in test group and 50 in control group.The patients of test group received the PGE1 combined with the PHGF for 30 days and the control group only received the most in using PGE1 for 30 days.Results:At the end of treatment,the total effective rates were 93.3% in the test group and 80% in control group.No severe side effect was observed in both groups.Conclusion:The PGE1 in conjunction with the PHGF is an effective and safe therapeutic intervention for patients with moderate or severe chronic hepatitis.
3.Study on Effect of Lamivudine in the Treatment of 36 Patients with CHB and HBV Carrier

Objective:To Study the efficacy and satety of lamivudine in the treatment of CHB and HBV carrier. Methods:All patients were divided into three groups. 21 patients with CHB were classified CHB test group(namely groupⅠ), 16 HBV carries were classified ASC test group(namely Group Ⅱ), 40 patients with CHB were CHB controlled group(namely controlled group). Both group Ⅰ and group Ⅱ received lamivudine, group Ⅰ was added protecting liver therapy. The controlled group only received protecting liver therapy. The treatment courses of three groups were one year. Results:The effective rates of group Ⅰ and group Ⅱ were higher than the controlled group in turning into negative HBeAg and HBV-DNA, both the negative-turning rate of liver functions and hepatic serology target declining fibrosis in group Ⅰ was more significant than the controlled group without severe side effect. Conclusion:Lamivudin is efficient and safe medicine in the treatment of CHB and HBV carrier.
4.Inhibition of clotrimazole on Paneth cell defensins

Aim To investigate the Clotrimazoles (CLT) inhibitive effect on the secretion of ?-defensins of the Paneth cells.Methods The ?-defensin mRNA expression is determined by semi-quantitative RT-PCR. The secretion of defensins is determined by dot-blot and ELISA. Results CLT has no influence on the defensin mRNA expression. All of 125 nmol?L -1, 250 nmol?L -1 and 500 nmol?L -1 CLT can inhibit the secretion of ?-defensins. Conclusion CLT could inhibit the secretion of defensins, which suggests that CLT maybe influence the function of small intestine

Objective Micronucleus test(MNT) was used to evaluate the single and combined genotoxicity of dichlorvos(DDVP),dimethoate(DM) and malathion(Mal) in HepG2 cell line,and 2?2 factorial design was adopted to elucidate the combined genotoxic effects.Methods In cytotoxicity test,HepG2 cells were exposed to the three OPPs(DDCP,DM,Mal) for 4 h respectively,and the doses at which cell viability above 80% were selected for the MNT,DDVP:3.125-25 ?g/ml,DM:25-200 ?g/ml,Mal:50400 ?g/ml,the micronucleated cell(MNC) rates and the replicative index(RI) were calculated.The combined genotoxicity of them was investigated with their doses as follows:low dose(DDVP:3.125 ?g/ml,DM:25 ?g/ml,Mal:50 ?g/ml);high dose(DDVP:12.5 ?g/ml,DM:100 ?g/ml,Mal:200 ?g/ml).Results In MNT,after treatment of HepG2 cells with DDVP,DM or Mal alone for 4 h,the MNC rates were increased in a dose-response manner(DDVP:r=0.955,P
7.Effect of endoscopic dilatation on children with benign esophageal stricture

Objective To study the curative effect of endoscopic dilatation in children with benign esophageal stricture. Methods 98 patients with benign esophageal stenosis from June 2013 to June 2016 were selected as the subjects of group A, group B and group C, 11 patients in group A were reflux esophageal stenosis, 43 children in group B were congenital esophageal atresia and in group C were 44 patients with chemical burn esophageal stricture. Then compare the three groups of children before the expansion of stenosis before the situation, the number of expansion and complications occurred. Results There was no significant difference in the average diameter of stenosis among the three groups (P > 0.05). The average length of the children in group C was higher than that in group A and group B, and group B was higher than group A, the difference was statistically significant (P > 0.05). The average number of expansion in group C was higher than that in group A and group B, and group B was higher than that in group A and group B higher than group A, the difference was statistically significant (P < 0.05). The incidence of complications in group C was higher than that in group A and B. The difference was statistically significant (P < 0.05). There was no significant difference in the incidence of complications between group A and group B (P > 0.05). Conclusion Endoscopic dilatation has a good clinical effect on children with benign esophageal stricture, but children with chemical burn esophageal stricture need to be expanded more often, while the complications are higher and the treatment is more difficult.
8.Mechanism and Evaluation Methods of Allergy Caused by Traditional Chinese Medicine Injection

Objective:To summarize the allergic mechanism caused by traditional Chinese medicine injection (TCMI) to provide some references for perfecting Good Laboratory Practice(GLP) of TCMI.Method:The related iteratures in data bases at home and abroad were reviewed,and the present experimental research methods about allergies were referred to to have a view of future studies.Result:The allergic mechanism of TCMI was mostly antigen-antibody reaction, part of which was anaphylactoid reaction.The method of the evaluation of allergy caused by TCMI only was animal experiments, but there were still some allergies caused by TCMI after the evaluation with this method.The present experimental research methods indicated that the detection of mediators of inflammation and FCM(Flow Cytometry) could be used to evaluate the allergies caused by TCMI.Conclusion:More attention should be paid to allergies caused by TCMI for its complicated mechanism and frequent occurrences in clinic.It may be an effective way to evaluate the allergies caused by TCMI with several methods including in vivo and in vitro.
9.Ile796Val polymorphism in the human SCAP gene and the correlation with serum lipid levels

Objective To study the Ile796Val polymorphism in human SCAP gene in Hubei area, and analyze its correlation with coronary heart disease (CHP) and hypertension and the relationship between polymorphism and lipid metabolism. Methods Using PCR RFLP, we detected genotypes of Ile796Val polymorphism in human SCAP gene. Results The allele A frequencies of Ile796Val in human SCAP gene for controls, CHD patients and the hypertension patients were 0.32, 0.45 and 0.42 respectively. The allele G frequencies were 0 68, 0.55 and 0.58 respectively. There were significant differences in frequencies of genotype and alleles between controls and hypertension group. And there was significant difference in the level of TC, LDL C and ApoB. In CHD group, there was significant difference in the TC level between different genotypes. In hypertension patients, although a difference was noted in genotype, there was no significant difference in allele frequencies and lipid level exceps a significant difference in the levels of TC, LDL C and ApoB in hypertension patients. Conclusion Ile796Val polymorphism in human SCAP gene may be a great agent to cause disorder in the lipid level of blood and lipid metabolism of tissue. It is of great significance in disorder in lipid metabolism of inter cellular and genetic investigation of hyperlipidemia.
10.Construction,transfection and activity identification of eukaryotic expression vector of human PRKCB1 containing enhanced green fluorescence protein gene

Objective To construct the eukaryotic expression vector of human PRKCB1 containing enhanced green fluorescence protein gene and transfer into human umbilical vein endothelial cells(HUVECs),then identify activity of expression protein.Methods The pReceiver-M29-PRKCB1 eukaryotic expression plasmid was constructed by frame amplified from pMD18-T-PRKCB1 plasmid.Then the recombinant plasmids were identified by enzyme analysis and DNA sequencing.According to optimized conditions,the eukaryotic expression plasmids were transfered into HUVECs and observed under fluorescence microscope.After that,transfection efficiency was calculated under random vision.The plasma membrane/cytosol ratio of fluorescence was calculated under confocal microscope.The translocation was identified.Results The gene sequence was completely consistent with that reported in GenBank.The enhanced green fluorescence protein was observed in HUVECs after 48 hours.Transfection efficiency was 18.6%?1.6%.The translocation was observed.Conclusion The eukaryotic expression plasmid is successfullyconstructed and transfered into HUVECs,the translocation was identified.It is a potential tool for screening HUVECs stably expressing human protein kinase C ?2 and isolating protein complex.
