Objective:To explore the influence of chaperone tapasin on HLA E expression on cell surface.Methods:The infection method was used to transfer the recombinant retrovirus expression vector which was constructed with HLA E molecules on target cells was detected with FACS technique.Results:Exogenous HLA E expression on T2 cell surface was detected with 74.13% and no HLA E expression was detected on control cells and that was transfected with empty vector.Conclusion:It is possible that the expression of HLA E molecules on cell surface involves in TAP independent mechanism.
3. Inhibitory effects of periplocin from cortex periplocae on proliferation of human esophageal carcinoma TE-13 cells

Objective: To study the inhibitory effects of periplocin from cortex periplocae (CPP) on proliferation of human esophageal carcinoma TE-13 cells and the related mechanism for cell cycle arrest. Methods: The inhibitory effects of CPP on the growth of TE-13 cells were measured by MTT assay. The morphological changes of TE-13 cells were analyzed by Gimsa staining. Detection of cell apoptosis and cell cycles were performed by flow cytometry. Protein expressions of cyclin-dependent kinase CDK2 and CDK4 were analyzed by Western blotting assay. Results:CPP significantly inhibited the proliferation of TE-13 cells in a dose- and time-dependent manner (P < 0.01). After treatment with CPP for 48 h, the IC50 was 0.61 μg/mL. TE-13 cells showed morphologic changes of apoptosis after treatment with CPP 2 μg/mL for 48 h. The proportion of cells in G0/G1 phase significantly increased (P < 0.01) and the number of cells in S phase decreased (P < 0.01). The proportion of cells in G2/M phase had no significant difference before and after CPP treatment (P > 0.05). CDK4 expression was inhibites by CPP at different concentrations, but the expression of CDK2 had no marked changes (P > 0.05). Conclusion: CPP significantly inhibites the growth of TE-13 cells. The action mechanism may be related with induction of cell cycle blocking and apoptosis.
5.Development of a software for 3D virtual phantom design.

In this paper, we present a 3D virtual phantom design software, which was developed based on object-oriented programming methodology and dedicated to medical physics research. This software was named Magical Phan tom (MPhantom), which is composed of 3D visual builder module and virtual CT scanner. The users can conveniently construct any complex 3D phantom, and then export the phantom as DICOM 3.0 CT images. MPhantom is a user-friendly and powerful software for 3D phantom configuration, and has passed the real scene's application test. MPhantom will accelerate the Monte Carlo simulation for dose calculation in radiation therapy and X ray imaging reconstruction algorithm research.

6.CHANGES IN T CELL SUBSETS OF MICE AFTER ACUTE IRRADIATION INJURY

In order to evaluate the changes in T cell subsets after acute irradiation injury, and investigate the mechanism of deficiency of cellular immunity in acute irradiation syndrome, Balb/c mice were exposed to a single total body irradiation of 5Gy gamma rays from a 60 Co source. On days 10, 28 and 120, the amounts of CD4 + and CD8 + subsets in spleen cells were evaluated by flow cytometry (FCM), and expression of IL 4 gene in ConA stimulated spleen cells was evaluated by reverse transcription linked polymerase chain reaction, in which ?-actin gene was used as an internal control. It was found that after irradiation, the ratio of CD4 + /CD8 + became significantly higher than that of the controls, and then the ratio lowered gradually until day 120. However, it still did not return to the level of the controls. On day 10, expression of IL 4 gene at mRNA level increased significantly. The results indicated that CD8 + subset was more radiosensitive than CD4 + , and the pattern of cytokine seeretion shifted to that of Th2 soon after irradiation.
7.Applied distally based peroneus brevis muscle flaps for coverage of the soft tissue defects over the lower one-third of the leg, ankle and foot

Objective To investigate a simple, valuable new method for coverage of the soft tissue defect over the lower one-third of the leg, ankle and the foot. Methods The distally based peroneus brevis muscle flaps were applied for coverage of the soft tissue defects over the lower one-third of the leg, the ankle and the foot. The muscle flaps were covered with split-thickness skin grafts. Results The distally based peroneus brevis muscle flaps were applied for coverage of the soft tissue defects over the lower one-third of the leg, the ankle and the foot in 16 cases. The largest area of the soft tissue defect was 5 cm?7 cm. The smallest was 3 cm?4 cm. Primary healing occured in 14 cases undergoing muscle flap construction, second-stage healing occured in 2 cases, no total flap necrosis occured in any cases. Conclusion This technigue is a simple and complications are lesser. The successful rates are higher. This muscle flap is suitable to the mudium or small soft tissue defect especially.
8.Changes of expression of neuronal nitric oxide synthase after sleep deprivation and effects of Xingnaojing on them

Objective To investigate the changes of expression of neuronal nitric oxide synthase(nNOS) after sleep deprivation(SD) in rats,and to study the effects of Xingnaojing on them. Methods Rapid eye movement(REM) SD models(SD group,n=40) were established by the modified multiple platform methods.The rats in SD group were subjected to drug intervention group(n=20) and non-drug intervention group(n=20).Xingnaojing injection was administered to the drug intervention group.Rats in SD group were deprived of sleep for 1,3,5 and 7 d. The expression of nNOS in brain tissues was detected by immunohistochemistry.The rats without SD were severed as cage control group(n=10) and tank control group(n=10). Results The expression of positive cells of nNOS in brain tissues increased with the time of SD,and Xingnaojing decreased the number of positive cells of nNOS.The area density of positive reaction cells in SD group was significantly higher than those in cage control group and tank control group(P
9.The Effect of Sertoli Cells Feeders on Mouse Spermatogonial Stem Cells Cultured in vitro

Objective Spermatogonial stem cells(SSCs)dissociated from 2~5 days postpartum mice were cultured on Sertoli cells feeders,to study the effect of Sertoli cells feeders on culture of mouse spermatogonial stem cells.Methods Specific markers CD9 of mouse SSCs cultured serum-free StemPro-34 SFM culture medium were identified by immunohistochemical assay.Result During the first week of culture,on Sertoli cells feeders or STO feeders,the biologica1 behaviors of spermatogonial stem cells showed no obvious difference.After a week of culture,compared with control,there were more number of spermatogonial stem cells remained when they were cultured for 60 days.These cells were expressed CD9 positive.In conclusion Sertoli cells can be used as feeders not only to promote survival but also renewal of spermatogonial stem cells.
10.Expression of Secretory Aspartyl Proteinase in Human Vulvovaginal Candidiasis

Objective To investigate the differential expression of secretory aspartyl proteinase in patients with vaginal C.albicans infection and in asymptomatic candidal carriers.Methods Secretory aspartyl proteinase expression was determined with reverse transcription-PCR in vaginal specimens taken from 10 asymptomatic candidal carriers,14 patients with vulvovaginal candidiasis (VVC) and 10 patients with recurrent VVC (RVVC).Results SAP2 and SAP4 to SAP6 subfamily were detected in both candidal carriers and patients with vaginal candidiasis.SAP1 and SAP3 transcripts were not observed in 10 asymptomatic candidal carriers.All 9 SAP genes were differently expressed in VVC and RVVC patients.SAP1 and SAP3 transcripts were frequently amplified in some patients.Conclusion It is shown that C.albicans infection is associated with differential expression of individual SAP genes,which may be involved in the pathogenesis of vaginal candidiasis.
