2.Progress in the mechanism of plasma N-terminal pro-brain natriuretic peptide in children with Kawasaki disease

Kawasaki disease ( KD) is an acute febrile systemic vasculitic syndrome. Since there is no specific diagnostic test for KD, the diagnosis is mainly based on the clinical menifetations according to the criterias and excluding of other diseases. Recently the value of plasma brain natriuretic peptide ( BNP) in the diagnosis of KD has been paid more attention gradually. The plasma NT-proBNP level is significantly elevated in KD patients in the acute phase. It is of some specificity and sensitivity, and it may due to the local myocardial inflammation, cytokines and diastolic function of left ventricular. This review summarizes the mechanisms by which the elevated levels of plasma NT-proBNP occur in the acute phase of KD.

4.Amplification of Full-length mRNA 5′Ends Based on Template-switching Effects

"CapFinder" technology,which can be used to clone the full length of 5′ UTR sequence of mRNA,was described.This technology used the terminal transferase activity of certain MMLV RT variants that added 3-5 residues(predominantly dC) to the 3′end of the first-strand cDNA exhibited when MMLV RT reached the 5′cap structure of mRNA.In the reverse reaction system containing GGG oligo,the terminal transferase activity was harnessed by the GGG oligo whose terminal stretch of dG residues can anneal to the dC-rich cDNA tail and serve as an extended template for RT.After RT switch templates from the mRNA template to the GGG oligo,a complete cDNA copy of the original RNA was synthesized with the additional GGG oligo sequences at the end.5′UTR of mRNA can be amplified with GGG oligo as forward primer and a gene-specific reverse primer.5′UTR of Bt toxin receptor E-Cadherin gene in midgut of cotton bollworm was cloned.
5.Optimization of Purification and Isolation of Punicalagin from Pomegranate Peel

Objective: To establish the methods for extraction and isolation of punicalagin from pomegranate peel, and further study the purification and quantification of punicalagin. Methods: Using an ultrasonic-assisted extraction method, punicalagin in pomegranate peel was extracted at room temperature by 50% ethanol with 20-fold volume of raw material. The content of punicalagin in the crude extract was determined by HPLC. To optimize the purification process of punicalagin, static adsorption and desorption experi-ments were employed to study five kinds of macroporous adsorbent resins (D101, A8-8, NKA-9, HPD-100 and HPD-500) for the one with the highest purification efficacy of punicalagin. In addition, the technical parameters of the macroporous adsorbent resin were opti-mized to obtain punicalagin with higher purity. Punicalagin was further separated and purified by using a reverse phase MCI GEL CHP20P column. Results:HPD500 resin showed the best ability to absorb and separate punicalagin in among five kinds of macro-porous adsorbent resins. The best technical parameters were as follows:the mass concentration of sample solution was 15 mg·ml-1 , the loading amount was 2BV, the pH was 2 and the eluting solvent was 8BV of 30% ethanol. With the best process as described a-bove, the content of punicalagin extracted from pomegranate peel increased from 10. 3% to 30. 7%. The obtained punicalagin could be further purified to 61. 3% from 30% in ethanol eluate by the reverse phase MCI GEL CHP20P column. Conclusion:HPD500 resin is the most effective in the purification of punicalagin from pomegranate peel, and the content of punicalagin can be dramatically increased after the purification by a reverse phase MCI GEL CHP20P column. The optimized process shows good reproducibility and stability.
6.Vitrification technology in whole embryo freezing

Objective To investigate the clinical outcome of patients who underwent whole embryo vitrification freezing and thawed frozen embryo transplantation, and to compare it with the patients at the same period in the fresh cycle to explore the value of vitrification technology in the whole embryo freezing combined with recovery transplantation. Methods The whole embryo freezing group included 40 untransplanted cases of in-vitro fertilization (IVF), while another 300 patients in the fresh cycle with in vitro fertilization and embryo transfer (IVF-ET) at the same period served as the fresh cycle group. The average number of transferred embryos, high-quality embryos, endometrial thickness, estradiol (E2) level, and clinical pregnancy rate were compared.Results The average number of transferred embryos, high-quality embryos, difference in endometrial thickness were not significant between the 2 groups (P>0.05); the E2 level of the fresh cylcle group was significantly higher than that of the whole embryo freezing group (P<0.05), and the clinical pregnancy rate in the fresh cylcle group was significantly lower than that of the whole embryo freezing group (P<0.05). Conclusion Vitrification technology is a simple, reliable, and efficient embryo freezing technology. For patients who meet the requirements, whole embryo vitrfication freezing and thawed frozen embryo transplantation is feasible. It is worth for further clinical research.
7.Effect of L-arginine Therapy on Serum Nitric Oxide and Endothelin in Acute Hypoxic Rat

PurposeTo study the changes of serum nitric oxide(NO)and endothelin(ET)levels by L-Arg therapy in acute hypoxic model. MethodsNADPH diaphorase histochemical staining method, Griess biochemical assay and radioimmune assay were applied to investigate the changes of nitric oxide synthase (NOS) ,NO, cGMP and ET in normal, hypoxic and L-Arg treated hypoxic rats. Resultsduring acute hypoxia, while serum ET rose up significantly, both the levels of serum NO and cGMP and the activity of NOS were significantly lower. After the application of L-Arg therapy, the activity of NOS did not change significantly, but the levels of serum NO and cGMP rose up significantly, and the level of serum ET was to the opposite direction. ConclusionsNO and ET may modulate hypoxic pulmonary hypertension, and acute hypoxia can result in acute hypoxic pulmonary hypertension. L-Arg can reverse the acute hypoxic hypertension.
8.Complement mediated killing of human glomerular mesangial cell by Fcα/μ receptor

Objective To study whether Fcα/μ receptor(Fcα/μR)can mediate complement killing of human glomerular mesangial cells.Methods Fcα/μR cDNA contained plasmid,pcDNA3.1-Fcα/μR was transfected into a human glomerular mesangial cell(NHMC).Fcα/μR expression was detected by Western blot and laser scanning eonfocal microscopy.Binding of IgM-immune complexes(IgM-IC)to the Fcα/μR on cell membrane was detected by flowcytometry and laser scanning confocal microscopy.Killing of cells by complement was shown by Trypan blue exclusion assay.Results NHMC cells transfected with Fcα/μR could bind IgM and IgM-IC.After treatment with complement,added IgM-IC,the death rate of pcDNA3.1-Fcα/μR transfected cell was significant higher than the control groups of wild type cell,pcDNA3.1 transfected cell and the pcDNA3.1-Fcα/μR transfected cell without IgM-IC.Conclusion IgM-IC can bind to the Fcα/μR expressed NHMC cells and mediate complement killing of the cells.
9.Evaluation verification of Mindary BC5390 hematology analyzer

Objective To evaluate the performance verification of Mindary BC5390 hematology analyzer .Methods According to requirement of CLSI documents ,the precision ,accuracy ,linearity ,carryover were evaluated .Results The background counts all met the designed requirements of manufacturer .Precision ,accuracy ,linearity of the WBC ,RBC ,PLT ,Hb ,HCT ,MCV were good and the contamination carrying rates were low .Except for basophils ,the classification of leukocyte was well correlated with the artificial microscopy classification(r2 >0 .95) .Conclusion The BC5390 is a ideal automated hematology analyzer which has high precision and accuracy ,wide linearity ,low contamination carrying rate ,and it can be used for the blood samples analysis .
10.Analysis of In Vitro Antifungal Susceptibility of Vaginal Isolates of Candida spp.

Objectives To analyse the antifungal susceptibility of Candida spp. isolated from the patients with recurrent vulvovaginal candidiasis (RVVC), vulvovaginal candidiasis (VVC) and asymptomatic carriers and to study the correlation between different Candida strains and antifungal susceptibility. Method According to the NCCLS-M27-A scheme, the antifungal susceptibility of Candida spp. isolated from the above different groups was measured. Results Almost all the MICs of C. glabrata and C. krusei to 8 antifungal agents were higher than those of C. albicans. The average MIC of C. albicans isolated from RVVC patients was higher than that from asymptomatic carriers. The resistant strains were mainly isolated from the RVVC group. No resistant strains against itraconazole, fluconazole, ketoconazole, econazole and nystatin was found in asymptomatic carriers. Conclusions These results indicate that more attention has to be paid to the low susceptibility of non-Candida albicans in the treatment of vulvovaginal candidiasis, and the resistant strains may result from long-term or irregular antifungal treatment.
