2.Preventive Effect of Tangshenkang Mixture on Experimental Diabetic Nephropathy

Objective To evaluate the effect of Tangshenkang Mixture (TM) on preventing diabetic nephropathy (DN). Methods SD rat model of diabetes was established by intraperitoneal injection of streptozotocin (STZ). Contents of blood sugar,urine sugar and urine albumin in rats were measured. The pathological changes of unilateral kidney were also observed under microscope by HE and PAS staining. Glomerulus dimensions,capillary vessel dimensions and PAS-positive matter area were measured with Pathological Picture Analyzing System. 3.Amplification of Full-length mRNA 5′Ends Based on Template-switching Effects

"CapFinder" technology,which can be used to clone the full length of 5′ UTR sequence of mRNA,was described.This technology used the terminal transferase activity of certain MMLV RT variants that added 3-5 residues(predominantly dC) to the 3′end of the first-strand cDNA exhibited when MMLV RT reached the 5′cap structure of mRNA.In the reverse reaction system containing GGG oligo,the terminal transferase activity was harnessed by the GGG oligo whose terminal stretch of dG residues can anneal to the dC-rich cDNA tail and serve as an extended template for RT.After RT switch templates from the mRNA template to the GGG oligo,a complete cDNA copy of the original RNA was synthesized with the additional GGG oligo sequences at the end.5′UTR of mRNA can be amplified with GGG oligo as forward primer and a gene-specific reverse primer.5′UTR of Bt toxin receptor E-Cadherin gene in midgut of cotton bollworm was cloned.
4.Exploration on thyroid hormones changes of different gestation periods

Objective To investigate the state of thyroid hormones and thyrotropin level in different gestation periods. Methods FT3, FT4, TSH and hCG were measured in 623 patients by ACS-180 automatic chemiluninescence analyzer. The urinary iodine was random measured by the use of iodine catalytic reaction mechanism of Arsenic-Cerium, rapid urinary iodine was detected with cold digestion method of quantitative detection. Result Serum FT3,FT4 ,and hCG levels in the first and second trimester were higher than in the third trimester ( F =53. 19,78.00,58.77, P <0. 05) ,and TSH levels were lower than in the third trimester ( F =5.68, P <0.05). The incidence of thyrotropin level disorder was 7. 54%, there were no significant differences in the gestation periods ( χ2 = 3.92, P > 0. 05 ). The incidence of subclinical hypothyroidism,subclinical hyperthyroidism, hypothyroidism and hyperthyroidism were 3.53%, 2. 57%, 1.12% and 0. 32%, respectively. The incidence of subclinical hypothyroidism in the first and third trimester was high, while the incidence of subclinical hyperthyroidism in the second trimester was high. Conclusions Thyrotropin level in the third trimester was lower than that of the first and second trimester. HCG changes were not the main reason that caused the thyrotropin level disorder during the pregnancy.
5.Distribution of T lymphocyte subsets in tumor microenvironment in papillary thyroid carcinoma

Objective To analyze the distribution of T lymphocyte subsets in tumor microenvironment of papillary thyroid carcinoma (PTC). Methods The levels of CD3+ CD4+ CD8- ,CD3+ CD4- CD8+ ,CD3+ CD4+ CD8+ ,CD3+ CD4- CD8- T cell and CD4+/CD8+ ra- tio in tumor microenvironment of 35 cases PTC, 25 cases benign thyroid lesion and 15 healthy eases were detected by flow cytometry. Results Local tumor tissue of 35 cases PTC and 25 cases benign thyroid lesion, and blood of 15 healthy cases were collected. The ratio of CD4+/ CD8+ in blood, tumor microenvironment of benign thyroid lesions and PTCs decreased significantly ( P<0.01). The levels of CD3+ CD4+ CD8+ cell in tumor microenvironment of PTC and benign thyroid lesion were higher than that in peripheral blood( P<0.01). But the differ- ence of the level of CD3+ CD4+ CD8+ cell between PTC and benign thyroid lesion was not significant ( P>0.05). The levels of CD3+ CD4- CD8- cell in tumor microenvironment of benign thyroid lesion were higher than those in PTC and peripheral blood(P<0.01). The difference of the level of CD3+ CD4- CD8- cell between PTC and peripheral blood was not significant (P>0.05 ). The differences of CD4+/CD8+ ratio and the level of each of T lymphocyte subset such as CD3+ CD4+ CD8-, CD3+ CD4- CD8+, CD3+ CD4+ CD8+, CD3+ CD4- CD8- cell between 15 PTC cases with lymph node metastasis and 20 PTC cases without metastasis were not significant (P>0.05). Conclusions CD8+ T cell is the main immune cell in tumor microenvironment in PTC. The local cell-mediated immunity of PTC is in the status of obviously suppressed. But the obvious increase of CD3+ CD4+ CD8+ and CD3+ CD4- CD8- T cells in tumor microenvironment in thyroid benign lesion may be beneficial for creating the location for immune responding against tumor. The distribution of T lymphocyte sub- sets in tumor microenvironment in PTC is not associated with lymph node metastasis.
6.Key points in verifying novelty assessment of medical literature

8.Application of big data in medical care and their privacy protection

Big data can provide effective data support for clinical research, biomedical technology progress, and individualized treatment of patients.However, medical ethics and data privacy must be considered due to the poor relation between patients and medical workers .The advantages and disadvantages of big data in medical practice were thus analyzed with certain strategies proposed for their development and privacy protection , and for the balance between the benefits and risks of technological innovations .
9.Analysis of the Local Immune Response in Papillary Thyroid Carcinoma

10.Study on structure-activity relationship of flavonoids' multidrug resistance-associated protein inhibitory activity.

To study the quantitative structure-activity relationship (QSAR) between the stuctures of 29 flavonoids and the inhibitory activity of their multidrug resistance-associated protein (MRP) 1 and 2 by using the comparative molecular similarity index analysis (CoMSIA). By studying the impact of the combination of different molecular force fields, researchers obtained the molecular force fields that played an important role in inhibiting the activity of MRP1 and MRP2, built the optimized QSAR model, and discussed the structural modification method for flavonoids' multidrug resistance-associated protein inhibitor. The results of the study could not only provide the guidance for new drug R&D, but also help partially discuss the synergy mechanism between MRP1 and MRP2 receptors and traditional Chinese medicines containing flavonoids.
