1.Clinical Study on Method of Soothing the Liver and Tonifying the Kidney for Treatment of 30 Cases of Female Depression

Objective:To observe changes of sexual hormone level,score for Hamilton depression scale(HAMD) and clinical score for TCM symptoms after treatment of female depression with the principle of soothing the liver and relieving depression,and tonifying the kidney and relieving mental stress.Methods:Sixty cases of female patients of depression were randomly assigned to a treatment group(n=30) and a control group(n=30) according to age and condition of patients.Scores for HAMD and TCM symptoms before and after treatment were recorded and compared in both groups.Serum estradiol(E_2),progestin(P) and prolactin(PRL) levels were determined in the patients of depression and healthy persons.Results:Hormone levels were correlated with HAMD scores and 4 factors of HAMD,and after treatment with the method for soothing the liver and tonifying the kidney,serum E_2 increased (P
2.Effects of maternal hypercholanemia on fetal myocardium in rats

Objective To explore the effects of maternal hypercholanemia on the myocardium changes in rat fetus. Methods Thirty clean SD female rats were equally randomized to three groups after mating successfully.From the 13th to 20th day of gestation,group A and B were injected injected with sodium chloride(NS) as control.Total bile acid(TBA) and cardiac troponin I(cTnI) were measured in the maternal and fetal serum on the 21st day when all rats were killed.Fetal cardiac muscle cells were also collected for examination with light microscope and electronic microscope.Results (1)TBA in maternal and fetal serum were(22.32±8.12)μmol/L and(28.84±8.06) μmol/L,respectively in group A,(9.77±3.56)/μmol/L and(9.34±3.54) μmol/L in group B,and (3.60±1.78) μmol/L and(3.95±1.19) μmol/L in group C.Significant differences were found among groups(P<0.01).(2)Fetal death rates were significantly different among the three groups (P<0.05),with 30.11%,16.85%,and 7.05%,respectively.(3)Fetal cTnl were also found significant difference among groups(P<0.01),with(19.98±7.75)ng/ml,(11.41±3.64)ng/ml and(4.38±1.19)ng/ml,respectively.(4)The integrated scores of fetal necrosis area were significantly different in three groups(P<0.05),with 1.92±0.43,1.36±0.37 and 0.44±0.12,respectively.(5)Under electronic microscope,the number density of mitochondria in group A was lower than that in group C(P<0.05)while the average volume of mitochondria was larger in group A (P<0.05).The average volume of mitoehondria in group B was larger than that in group C(P<0.05) while no difference was found with regard to the number density between the two groups.The number density and average volume of myofibril in group A were lower than those in group C(P<0.05).The number density of myofibril in group B was higher than that in group C(P<0.05) while no difference was found with the average volume.(6)Positive correlations were found in maternal TBA,fetal TBA,fetal cTnI and the integrate of fetal necrosis area when comparing every two of the above factors. Conclusions Fetal myocardium is impaired obviously in hypercholanemia rats.The serum level of TBA and cTnI in fetal rats are positively correlated with each other.

Physical and psychological stresses inhibit or harm reproductive endocrine especially female one at many levels of hypothalamo-pituitary-gonadal (HPG) axis, and result in subhealth conditions or clinic diseases in reproductive endocrine. The hypothalamic corticotropin-releasing factor (CRF) and adrenal glucocorticoids (GCS) overreleases and other neuroendocrine alterations induced by stress are implicated in the mechanisms responsible for the disturbance of reproductive endocrine. The declines in activities of synzyms for reproductive hormones cause sexual hormones reduces and disfunction in reproductive endocrine system.
4.Determination of Ginsenoside Rg_1 in Guyuling Capsule by HPLC

Objective To establish a HPLC method for the content determination of Ginsenoside Rg1 in Guyuling Capsule. Methods HPLC was performed on a C18 column ,the mobile phase consisting of acetonitrile-0.5 %H3PO4 (23 ∶77 )and detection wavelength at 205 nm. Results The linearity of Ginsenoside Rg1 was good in the range of 1.05 ?g ~7 ?g,r=0.999 6.The average recovery was 98.80 %,and RSD was 1.57 %.Conclusion The method is simple,feasible,and reproducible ,and can be used for the quality control of Guyuling Capsule.
5.PREPARATION OF ~(35)S-LABEJJED CPS_1 cDNA PROBE FOR IN SITU HYBRIDIZATION

The carbamyl phosphate synthetase I (CPS,) cDNA probe labelled with alpha ~(35)S-dATP was obtained by using modified nick translation technigue. The specific radioactivity of the obtained probe reached 0.9-1.5?10~8cpm/ug DNA. This probe could be satisfactorily used in RNA-DNA in situ hybrioization for detecting the gene expression of CPS_1 in liver tissue sections. The advantages of using ~(35)S instead of ~(32)P and ~3H for labelling DNA probe were discussed.
6.Baseline characteristics and visual outcomes after two years follow-up of exudative age-related macular ;degeneration patients treated with ranibizumb

Objective To observe the baseline characteristics and visual outcomes after two years follow-up of exudative age-related macular degeneration (AMD) patients treated with ranibizumb. Methods Forty-four eyes of 44 patients with exudative AMD were enrolled into this retrospective study, 19 were men and 25 were women. The mean age was 78 years (range 64–92 years). All patients were underwent best corrected visual acuity (BCVA, Early Treatment of Diabetic Retinopathy Study), fundus color photography, fundus fluorescein angiography (FFA), indocyanine green angiography (ICGA) and optical coherence tomography (OCT). The mean BCVA was (50.36±14.43) letters, the mean central foveal thickness (CFT) was (291.95± 82.19)μm, and the fluorescence leakage area of choroidal neovascularization (CNV) was (7.61±5.84) mm2. All patients received three initial intravitreous injection of ranibizumb (IVR) and were retreated with monthly IVR when needed. The mean follow up time was 25.6 months (range 24–29 months). On 1, 2, 3, 6, 12, 18 and 24 months after treatment, BCVA and OCT were repeated. On 3, 6, 12, 18 and 24 months after treatment, FFA and ICGA were repeated. The change of BCVA, CFT and fluorescence leakage area of CNV were observed. The association of baseline characteristics and two year visual outcomes were analyzed. Results On 1, 2, 3, 6, 12, 18 and 24 months after treatment, the BCVA were improved significantly (t=?1.89,?3.51,?4.61,?4.04,?5.77,?4.69;P<0.05), the CFT were decreased significantly (t=1.51, 2.30, 3.40, 3.28, 3.54, 3.88, 3.73;P<0.05). On 3, 6, 12, 18 and 24 months after treatment, the fluorescence leakage area of CNV were reduced significantly (t=2.12, 2.90, 3.51, 4.12, 4.06;P<0.05). The lower baseline BCVA, the more improved after treatment. The BCVA improvement degree has a negative relationship with baseline BCVA and fluorescence leakage area of CNV (r=0.505,?0.550;P<0.05), but no correlation with baseline CFT (r=0.210, P>0.05). Conclusion Two year visual outcomes of exudative AMD patients treated with ranibizumb is negative correlated with baseline BCVA and fluorescence leakage area of CNV, but not correlated with baseline CFT.
7.Apoptosis of human laryngeal carcinoma Hep-2 cell induced by matrine

Objective To investigate the mechanism of grow-inhibition and apoptosis induced by marine on Hep-2 line. Methods The growth inhibiting rate of matrine on Hep-2 cells was detected by MTT assay. Fluorescence microscope, DNA gel electrophoresis and flow cytometry (FCM) were applied to determine the presence of apoptosis and cell cycle. Results Matrine had cytotoxic effect to Hep-2 cells in a time- and dose-dependent manner. The FCM analysis showed that the number of Hep-2 cells of S phase and the apoptosis rate increased, while the number of G2/M phase decreased. There were morphological changes including chromatin condensation, nuclear fragmentation and apeptotic bodies formed. DNA gel electrophoresis analysis showed typical DNA ladder of apoptosis. Conclusion Matrine inhibits cell prolif-eration by blocking Hep-2 cell cycle to S phase, and exerts its anti-carcinoma function by inducing apoptosis.
8.Study of angiotensin-Ⅱactivating NF-κB in human pulmonary microvascular endothelial cells through classical pathway

Objective To investigate the activation of nuclear factor-κB(NF-κB),which was stimulated by angiotensin-Ⅱ(AngⅡ)through classical pathway in human pulmonary microvascular endothehal cells(HPMEC).Method The experiment was divided into two groups:in Ang Ⅱ group,HPMECS were incubated with 10-6mol/L AngⅡ for 0,0.5,1,2 and 4 hours,respectively;in losartan group,HPMEC was pretreated with 10-6mol/L losartan(inhibitor of AngⅡ type 1 receptor)for 1 hour,and then stimulated with 10-6mol/L AngⅡ for 2 hours,and the nucleax protein and the cell plasma protein were prepared by lysis and centrifugation.Electrophoretic mobility shift assay(EMSA)was used to detect the NF-κB DNA binding activity.The inhibitor of κBa(IκBα)was detected by Western blotting.The data were expressed as(x±s)and analyzed with one way analysis of variance.A P value less than 0.05 indicated significant difference.Results Compared with the activity of NF-κB at 0 h (100.0±25.1)after AngⅡstimalation,the activity increased significantly at 0.5 hour(144.5±16.1,P<0.05),and reached peak value at 2 hours(270.1±27.2,P<0.05).The concentration of IκBα at 0 hours was 44.4%±2.1%,decreased markedly at 0.5 hours(38.9%±3.6%,P<0.05),and to the lowest level at 2hours(32.6%±2.3%,P<0.05).The activity of NF-κB(115.4±10.7)and the concentration of IκBα(43.6%±3.7%)in losartan group had ilo significant difference with AngⅡ group at 0 h(P>0.05).The activity of NF-κB and the concentration of IκBα in losartan group had significant difference with AngⅡ group at 2hours.Conclusions NF-κB can be activated through classical pathway,which stimulated by AngⅡ in HPMEC.
10.Content Determination of Cyproheptadine Hydrochloride in Fuyan Cream by Simultaneous Equations

OBJECTIVE:To establish a method for content determination of cyproheptadine hydrochloride in fuyan cream.METHODS:Ultraviolet spectophotometry was applied to determine the contents of cyprohepatadine hydrochloride as well as its adjuvant as two coexisting components without isolation and extraction,the wavelengths were286nm and258nm res_ pectively,linear regression and simultaneous equations were used to analyze the results.RESULTS:Cyproheptadine hydrochlor_ ide and its adjuvant showed good linear relationship in the range of2.52~25.2and2.5022~25.022?g/ml respectively,the average recovery of cyproheptadine hydrochloride was98.61%(RSD=1.08%).CONCLUSION:The present method is simple,convenient,reproducible and reliable,which is suitable for rapid determination for fuyan cream.
