1.Correlations between illness uncertainty and social support of family members of elder patients with colostomy

Objective To investigate the illness uncertainty state of the family members of elder patients with colostomy,explore their correlation with social support.Methods A total of 138 family members of elder patients with colostomy in two hospitals from September 2013 to November 2014 were recruited in this study.The General Information Questionnaire,Mishel Uncertainty in Illness Scale-Family Member Form (MUIS-FM) and Social Support Rating Scale (SSRS) were used to investigate the family members of elder patients with colostomy and analyse the correlation between illness uncertainty of family members and its influencing factors and social support.Results The average total score of illness uncertainty was (72.17±9.44) scores.There was significantly different of the family members of elder patients with colostomy in total uncertainty on degree of education,the relationship with patient,stoma nature and economic conditions (P<0.05).The average total score of social support was (37.14±5.25) scores.The level of illness uncertainty was negatively associated with social support (r=-0.526,P<0.01).Conclusions The illness uncertainty was mostly in higher levels in the family members of elder patients with colostomy and was negatively correlated with social support.Clinical practice should improve social support of family members to relieve illness uncertainty,so as to provide better services of family and promote physical and psychological rehabilitation of patients.
2.Study to treatment effect of clinical useful method about diabetie neurogenic blader

Objective The study about comparison of the used treatments,to identify the best clinical treat-ment of diabetic neurogenie bladder.Methods 120 in-patients or out-patients with type 2 diabetes ale randomly di-vided into four groups.On the basis ofthe combined treatment,3 groups of A,B and C are given vitamin B12,B and C group are added respectively cisapride or neostigmine,D group are given methycobal.Results Before and after con-trast,the effects of four groups ale significant,but the effects of B,C and D group ale early and very significant;inter-group differences in contrast,between B,C,D group and A group is significant;between B,C and A group is signifi-cant;between B and C group is no significant.Conclusion The effect of neostigmine or cisapride and vitamin jointly can signifiCalltly reduce tlle DNB patients'residual urine in bladder and urinary retention.the effect is better than a simple application of vitamin or mecobalamin;For DNB milds,as appropriate,can use the treatment of mecobalamin.
3.Cost-Effectiveness Analysis of Three Drugs for Patients with Hypertension

OBJECTIVE:To compare the curative effects and pharmacoeconomical efficacy of three drugs in the treatment of patients with hypertension.METHODS:A total of 90 patients with hypertension were randomly divided into three groups:30(Group A) were treated with enalapril,30(Group B) with groLosartan,and 30(Group C) with metoprolol,all with a treatment course of 6wk.The clinical effects were observed and the cost-effectiveness analysis was conducted among the three schemes. RESULTS:The total effective rates of the 3 groups (A,B,and C) were 93.1%,96.7%,and 83.3%,respectively;the incidence rates of ADR were 10.3%,10.0%,and 16.7%,respectively;the cost-effectiveness ratios were 77.39,338.78,and 180.40,respectively;The incremental cost-effectiveness ratio of Group B and C were 7 098.61 and —798.16,respectively as against Group A.CONCLUSION:Group A(enalapril) treatment programs is a more reasonable drug program.
4.Effects of low-molecular weight heparin on MMP-2, TIMP-2 expression and invasiveness of cytotrophoblastic cells

AIM: To investigate the hypothesis that low-molecular weight heparin (LMWH) regulates in vitro cytotrophoblast invasiveness and production of metalloproteinases-2 (MMP-2), tissue inhibitor of metalloproteinas-2 (TIMP-2). METHODS: Chorionic villi tissue of normal 6-8 weeks pregnancy was obtained. Trophoblastic cells were collected by trypsin-collagenase digestion and Percoll gradient centrifugation. The cytotrophoblastic cells were cultured for 24 h and divided into 4 groups according to the concentrations (1.0×10~2 IU/L, 1.0×10~3 IU/L or 1.0×10~4 IU/L) of LMWH adding into the medium. The contents of MMP-2 and TIMP-2 in cell culture supernatants were measured by the method of ELISA. Cytotrophoblast invasiveness was determined by Transwell chamber assay. RESULTS: With the increasing concentrations of LMWH, the invasion activity of cytotrophoblastic cells and MMP-2 secretion were increased. At concentration of 1.0×10~3IU/L, LMWH greatly enhanced cytotrophoblast invasiveness and the expression of MMP-2 (P<0.05). The levels of TIMP-2 were decreased after intervention with LMWH. At concentration of 1.0×10~3IU/L or 1.0×10~4 IU/L, LMWH induced a significant decrease in TIMP-2 expression. No significant difference between group 1×10~3IU/L and group 1.0×10~4 IU/L was observed (P>0.05). CONCLUSION: LMWH might regulate cytotrophoblast invasiveness in vitro by influencing the expression of MMP-2 and TIMP-2 in cytotrophoblastic cells.
5.Microanatomy,histology and CT of arachnoid granulations in middle cranial fossa

Objective To further improve the morphological materials of AGs by micro-dissection, histology and CT, we observed the arachnoid granulations (AGs) in middle cranial fossa. Methods Thirty-three adult cadaveric heads were used for microsurgical dissection;Histological sections of AG specimens from 3 cadaver heads were examined. Forty patients who had both normal conventional brain CT and computed tomographic venography (CTV) were retrospectively reviewed. Results In middle cranial fossa the AGs occur in the following situations in order of frequency: the middle meningeal sinus, sphenoparietal sinus, lateral foramen rotundum and cavernous sinus. AGs usually show round, oval in shape and irregular in shape. AGs can be divided into individual type and leaflet type under light microscope. The numbers of AGs were observed by microanatomy and CTV were 8.72 and 3.52 respectively. The AGs of cavernous sinus was not localized precisely on CTV. Conclusion Study of the AGs in the middle cranial fossa systematically and comprehensively enriches anatomy and image knowledge. It is helpful in neurosurgical planning and choosing operalion procedure to avoid postoperative complications.
6.Effects of angiotensin converting enzyme inhibitor on expression of transforming growth factor β1 and β1-integrin in glomerular mesangial cells induced by lipopolysacchatide

Objective To observe the effects of one kind of angiotensin converting enzyme inhibitor (ACE1) drugs fosinopril (FOS) on transforming growth factor β1 (TGF-β1)and β1- integrin( Itg-31 ) expression in rat glomerular mesangial cells (GMC)induced by lipopolysacchatide (LPS).Methods We established the cultured glomerular mesangial cells of rat in vitro and passages 3 ~ 10 of cells were used in the experiment after identification.The experiment included the following groups:Control group,LPS induced group (LPS group) and FOS intervened group.According to the different concentrations of FOS,FOS intervened group was divided into high,middle and low dose FOS groups,which were FOS1 group,FOS2 group and FOS3 group respectively.The changes of TGF-β1 protein secretion was detected by the enzyme-linked immunosorbent-assay; The changes of TGF-β1 and Itg-β1 mRNA expression was detected by quantitative real-time RT-PCR.Results (1) TGF-β1protein secretion in rat GMC at 6h,12h,24h three time points:They were 958.55 ± 34.67 ( ng/L),1052.05 ±48.59( ng/L),1166.06 + 35.39 (ng/L) respectively in Control group.They were 1342.12 + 39.87 ( ng/L),1432.31 + 39.33 (ng/L) and 1 537.77 + 43.79 (ng/L) respectively in LPS group,which were higher significantly than those in Control group ( all P < 0.01 ).They were 779.58 ± 48.64 ( ng/L),878.33 ± 29.50 (ng/L) and 962.57 ±31.94( ng/L) in FOS1 group,989.311±73.56(ng/L),1073.29±66.89(ng/L) and 1210.75 ±61.68(ng/L) in FOS2 group,1 253.78 ±45.32( ng/L),1 348.18 ±45.81 (ng/L) and 1450.06 ±46.24( ng/L) in FOS3 group respectively,which were lower significantly in all FOS intervened groups than that in LPS group (all P<O.01).(2)TGF-β1 mRNA expressions in rat GMC at6h,12h,24h three time points were higher significantly than that in Control group.TGF-β1 mRNA expressions were lower significantly in all FOS intervened groups than that in LPS group.( 3 ) Itg-β1 mRNA expressiones in rat GMC at 6h,12h,24h three time points were higher significantly than that in Control group.Itg-β1 expressions were lower significantly in all FOS intervened groups than that in LPS group.Conclusions LPS can induce the increase of TGF-β1 secretion and mRNA expression.FOS can inhibit the TGF-β1 secrection and mRNA expession in GMC as dose-dependent manner,at the same time down regulated the Itg-β1 mRNA expression iuduced by LPS.All above supply the theoretical evidence for the renal protection of FOS by non-hemodynamics mechanism.
7.Effect of MG132 on the expression of ERK1/2 and connective tissue growth factor in rat peritoneal mesothelial cells induced by high glucose

Objective To observe the effect of MG132 on the expression of extracellular regulated kinase 1/2 (ERK1/2) and connective tissue growth factor (CTGF) in rat peritoneal mesothelial cells (RPMCs) induced by high glucose.Methods RPMCs were isolated,cultured and passaged by trypsin,then identified.The second generation of cultured RPMCs were used in the experiment.RPMCs were divided into normal control group,high glucose (1.5%,2.5%,4.25%) for 24 hours,high glucose (2.5%) for 0,12,24,48 hours,incubated with MG132 (0.5,1,2 μmol/L) for half an hour and then with high glucose (2.5%) for 24 hours.ERK1/2 protein was detected by Western blotting,and CTGF protein in supernatant was detected by ELISA.Results Compared with the control group,the expression of p-ERK1/2 was significantly increased in the groups stimulated by high glucose (P <0.01),reached the peak at 24th hour (P < 0.01),and then the expression decreased at 48th hour,but still was higher than that in the normal control group (P < 0.01).CTGF protein expression of RPMCs induced by high glucose increased,in time-and dose-dependent manner (P < 0.05).MG132 could significantly decrease the expression of ERK1/2 and CTGF induced by high glucose (P<0.05).Conclusions MG132 can decrease the expression of p-ERK1/2 and CTGF in RPMCs induced by high glucose.The ubiquitin proteasome pathway participates in the development of peritoneal fibrosis,and blocking the way may contribute to the prevention of peritoneal fibrosis.
8.Effects of Electrolyte on the Stability of Neonatal Parenteral Nutrition

OBJECTIVE:To study the effects of electrolyte on the stability of the neonatal parenteral nutrition. METHODS:Under room temperature(25 ℃),the neonatal parenteral nutrition containing only monovalent or bivalent ion electrolyte(10% So-dium chloride injection,10% Potassium chloride injection,25% Magnesium sulfate injection,and Calcium gluconate injection) and containing both monovalent and bivalent ion electrolyte were investigated by the change of appearance to determine the pH val-ue,insoluble particles and the size and distribution (polydispersity index,PDI) within 24 h. RESULTS:The pH of the nutrition with electrolyte was over 5 and also met the quality requirements;there were no precipitate,flocculation and discoloration in the appearance;the neonatal parenteral nutrition containing only monovalent ion electrolyte appeared a small amount of hanging wall phenomenon for 24 h,but did not appear demulsification phenomenon;the neonatal parenteral nutrition containing only bivalent ion electrolyte appeared a small amount of hanging wall phenomenon and demulsification phenomenon for 24 h;the neonatal paren-teral nutrition containing both monovalent and bivalent ion electrolyte appeared a small amount of hanging wall phenomenon and de-mulsification phenomenon for 12 h and the hanging wall phenomenon was more obvious for 24 h. Meanwhile,a size bigger than 5μm microns and particle size bigger than 25 μm microns of insoluble particles appeared,and both the average particle size and PDI value were higher than those in the previous two situations(P<0.05). CONCLUSIONS:As more and more monovalent and biva-lent ion electrolyte being added into the neonatal parenteral nutrition,especially divalent ion electrolyte,the stability of the neona-tal parenteral nutrition decreases,which behaves as a phenomenon that the size of grains and the number of insoluble particles in-crease.
9.Role of B cells in CD45 RB antibody-induced transplantation immune tol-erance

AIM:To investigate the role of B cells in CD45RB antibody-induced transplantation immune toler-ance.METHODS:Single cell suspension was made from the spleen of BALB/c nude mice disposed by CD45RB antibod-y, then mixed cultured with T cells of BALB/c mice and spleen cells of C57BL/6 mice.The Th1, Th2, Treg and Tm cells were monitored by flow cytometry during the culture process .The skin graft model was set up with B 6.μMT-/-mice as re-ceptors and BALB/c mice as donors.CD45RB antibody was intraperitoneally injected into the receptors after transplantation and then CD3+CD45RBhi cells were detected by flow cytometry .In another mixed lymphocyte culture , CD45RB antibody was added, and then B cells were isolated and injected into B6.μMT-/-mice through the tail vein.The heart transplanta-tion model was established with B 6.μMT-/-mice as receptors and BALB/c mice as donors, and then the survival and the migration of B cells to the thymus were observed .RESULTS:When T lymphocytes were co-cultured with B lymphocytes treated with anti-CD45RB monoclonal antibody (mAb) in vivo, the percentages of Th2 and Treg cells were up-regulated and Th1 cells were down-regulated, but Tm cells were not altered as compared with the control .In vivo without B lympho-cytes, anti-CD45RB mAb also down-regulated the expression of CD45RB in T lymphocytes.The reduction was faster and the percentage of CD3 +CD45RBhi T cells was not altered as compared with the control .The B lymphocytes treated with an-ti-CD45RB mAb in vitro prolonged the lifetime of receptor in heart transplantation model but failed to induce complete toler -ance.After recieving B cells treated with anti-CD45RB mAb and allogeneic heart transplantation , B cells migrated to the thymus in B6.μMT-/-mice.CONCLUSION:B lymphocytes play a definite role in the transplantation immune tolerance induced by anti-CD45RB mAb through their affection on T-cell subgroups and also in the central tolerance .However, the induction of immune tolerance can not only rely on B cells .
10.Investigation on deciduous dental caries among preschool children in Chongqing city.

OBJECTIVETo investigate the status of deciduous dental caries among preschool children in Chongqing city and to compare the oral health status of preschoolers from public and private kindergarten.
METHODSAccording to the criteria recommended by The Third National Oral Health Investigation, the deciduous caries of 5 030 preschool children aged 3 to 6 years old from 40 private kindergartens and 16 public kindergartens in Yuzhong district of Chongqing were examined. The prevalence of dental caries and the mean dmft and filling rate were calculated. The results were analyzed using SPSS 21.0 software package.
RESULTSMaxillary deciduous central incisors and mandibular deciduous molar were most prone to decay in 5,030 children. The prevalence of dental caries was 54.08% (2,720/5,030). The mean dmft score was 2.58 and the filling rate was 0.98% (127/12,993). The prevalence of dental caries and mean dmft increased gradually as the children were growing up (P < 0.05). The rate of filling teeth of the 6 year old group was higher than other groups (P < 0.05). No significant difference between males and females were noted (P > 0.05). The prevalence rate of deciduous dental caries among private kindergartens was 61.04% (1,656/2,713), with a mean dmft of 3.12 and filling rate of 0.35% (30/8,465). The prevalence rate of deciduous dental caries among public kindergartens was 45.92% (1,064/2,317), with a mean dmft of 1.95 and filling rate of 2.14% (97/4,528). Statistical analysis revealed significant differences in the prevalence rate, mean dmft, and filling rate between two kinds of kindergarten (P < 0.05).
CONCLUSIONDeciduous dental caries of preschool children is an important problem in Chongqing city and the children from private kindergarten had very low filling rate.
