1.Study on Ancient Literature of Correlation Between Urticaria and External Infection of Six Evil Factors

Urticaria, which is called as“yin zhen”in traditional Chinese medicine, is a kind of allergic skin disease with comings and goings of wind clumps patches. Its episodes have close relationship with external infection of six evil factors. This article collected, classified, and studied literature of TCM doctors in different dynasties about urticaria and external infection of six evil factors, and expounded pathogenesis of urticaria in the aspects of wind pathogen, dampness pathogen, fire evil and other pathogenic factors. In the treatment of urticaria, internal treatment is divided into dispelling wind method, eliminating dampness method, clearing heat method and other methods. External treatment is concluded form the aspects of washout method, medicated ironing method, and moxibustion method. This article classified and dig some valuable information, with a purpose to provide reference and evidence for prevention and treatment of urticaria.
2.Primary research of the importance of ALT testing on the safety of blood donor screening system

Objective To analyze the second time blood screening results of ALT deferred donors,and to evaluate the importance of alanine aminotransferase(ALT) testing on the improvement of blood safety.Methods The ALT testing results of 565 360 blood donors from Feb.2006 to Jan.2008 of Shanghai Blood Center were studied retrospectively.The screening results and donation intervals of such donors who delayed their donation just because of their former unqualified ALT level were also analyzed.Results A total of 32 042 donors(5.67%) failed in ALT testing among 565 360 donors.And 3 395 ALT deferred donors participated the second time blood donation,among which 2 205(64.95%) passed the blood screening tests,while the other 1 190(35.05%) failed.Among the 1 190 unqualified blood donors,1 151(33.90%)failed again in ALT testing,and 11(0.32%) in Syphilis,12(0.35%) in HBsAg,7(0.21%) in anti-HCV and 1 in anit-HIV(0.03%).Meanwhile,donors failed both in ALT testing combined with HBsAg,anti-HCV,and anit-HIV sero-converted were 1(0.03%),2(0.06%) and 1(0.03%),respectively.And 72.64% of ALT deferred donors participated the second time blood donation within 6 months.The average donation intervals of donors with qualified ALT level but sero-converted were 140 days(from 24 to 267 days),and those with both unqualified ALT level and sero-converted were 158 days(from 91 to 220 days).Conclusion Before the new methods such as NAT were applied to blood donation screening system,ALT test could prevent the window-period failure of ELISA screening so as to improve the blood safety.
3.Observation of the effect of thoracoscopic esophagectomy for 22 patients with esophageal cancer

Objective To explore the safety and outcomes of thoracoscopic esophagectomy for esophageal cancer.Methods The clinical data of 22 patients with esophageal cancer received thoracoscopic minimally invasive esophagectomy ( study group ) were retrospectively analyzed and compared with 46 patients underwent conventional open thoracic esophagectomy( control group) .The medium-term clinic outcomes of the two groups were compared. Results Compared to the control group,the study group showed significantly longer operation time[(183 ±37)mins vs (239 ±41)mins,t=6.19,P=0.002],less intraoperative blood loss[(95 ±33)mL vs (115 ±27)mL,t=5.04, P=0.004],shorter postoperative hospital stay[(15.8 ±6.2)d vs (17.5 ±7.1)d,t=5.62,P=0.003],and more removed lymph nodes[(11.6 ±4.2) vs (10.3 ±5.8),t=-3.19,P=0.018)].No significant difference was observed in the survival rate between the two groups (χ2 =0.358,P=0.449) .Conclusion There is no difference in medium-term clinic outcomes between the two kinds of esophagectomy.The patients with esophageal cancer received thoracoscopic minimally invasive esophagectomy has less complication and quick recovery.And the technology should be widely applied.
4.Effect of paracrine of hypoxia preconditioned umbilical cord mesenchymal stem cells on function of osteoblasts

Objective To study the paracrine effect of hypoxic preconditioned umbilical cord mesenchymal stem cells (UCMSCs) on the proliferation,migration and osteogenic differentiation of osteoblasts.Methods UCMSCs were cultured under hypoxia and normal oxygen condition before two UCMSCs conditioned media were obtained.After that,MG-63 cells were cultured in three groups:hypoxia conditioned medium group,normoxia conditioned medium group and DMEM control group.The proliferation of MG-63 cells was detected by mosmann tetrazoline colorimetry( MTT) method after 1,3 and 5 days.The migratory ability of MG-63 cells was detected by scratch assay .After 21 days′culture , the formation of osteogenic calcium nodules was detected by Alizarin red staining.ELISA method was used to detect the content of vascular endothelial growth factor ( VEGF) in hypoxia and normoxia conditioned medium.Results The MTT test showed that the proliferation ability of MG-63 cells in hypoxia conditioned medium group and normoxia conditioned medium group was greater than in the DMEM control group.The difference was statistically significant ( P <0.05).Furthermore, the proliferation ability of cells in hypoxia conditioned medium group were much greater than cells in normoxia culture medium group.The difference was statistically significant (P<0.05).Scrath assay showed that the migratory ability of MG-63 cells in hypoxia conditioned medium group and normoxia conditioned medium group was greater than cells in DMEM control group,and cells in hypoxia conditioned medium group was much greater than cells in normoxia conditioned medium group. After 21 days′culture,we found that the number of calcium nodules was the largest in hypoxia conditioned medium group, followed by normoxia conditioned medium group and DMEM control group.ELISA showed that the content of VEGF in hypoxia conditioned medium was higher than that in normoxia conditioned medium and the difference was statistically significant (P<0.01).Conclusion The paracrine function of UCMSCs can be enhanced by hypoxia,thus improving the proliferation,migration and osteogenic differentiation of osteoblasts.
5.Tanreqing Injection in Combination with Antibacterials for Hospital-acquired Pneumonia

OBJECTIVE:To observe the therapeutic effect of Tanreqing injection in combination with Piperacillin sodium and Amikacin sulfate for hospital-acquired pneumonia.METHODS:104 cases of hospital-acquired pneumonia were enrolled and randomly assigned to receive Tanreqing Injection+Piperacillin Sodium+Amikacin Sulfate(treatment group,n=52) or Piperacillin Sodium+Amikacin sulfate(control group,n=52) for 7~10days.The clinical indexes including the total effective effect,cardinal symptoms,physical signs,and chest X-ray film in two groups after treatment were observed.RESULTS:The total effective rate in the treatment group was 86.54% as compared 67.31% in control group(P
6.The Inhibiting Effect of Photofrin-Diomed 630-PDT Photodynamic Therapy on Human Immortalization Eesophageal Epithelial Cell Line SHEE and Its Malignant Transformation Cell Line SHEEC

Objective To investigate the sensitivity of tumor cells to photodynamic therapy(PDT), and to select the optimal photo-dose and photo density for PDT to the cultured cells. Methods Cells of SHEE and SHEEC cell lines were divided into 21 groups randomly,after 24 h inoculation, the cells accreted on paries of culture capsule completely, we replaced M199 complete culture solution with 30 μg/rnl Photofrin-Ⅱ solution 100 μl and then replacd with M199 complete culture solution without Photofiin-Ⅱ after 150 min of incubation in Photofrin-Ⅱ. Within 15 min,we dealed the cells with PDT using Photofrin-Diomed 630 under three different power densities:25 raW/cm~2,50 mW/cm~2 and 100 mW/cm~2 for 10 s,20 9,30 s,50 s, 100 s, 150 s and 200 s respectively,and then continue for 24 h culture. We examined the inhibiting effect on cell line SHEE and SHEEC under PDT by the method of CCK-8. Results There was no significant difference in the inhibition ratio with same power density of PDT between cell line SHEE and SHEEC under the concentration of 30 μg/ml Photofrin-Ⅱ. However, the inhibition ratio increased with the raising of photo-dose of PDT,but there was a platform stage after the 2.5~30 J/cm~ 2 photo-dose. Conclu-sion The difference for photodynamic sensitivity between human immortalization esophageal epithelial cell line SHEE and the malignant transformation cell line SHEEC is not significant. The targeting of PDT to malignant tumor cell may not be involved in the photosensitivity for tumor cell.
7.Relationship between expression of DAS-1 protein in intestinal metaplasia and gastric cardia adenocarcinoma

Objective To investigate the relationship between expression of DAS-1 in gastric cardia intestinal metaplasia(CIM)and gastric cardia adenocarcinoma (GCA). Methods The cancerous tissues and CIM tissues (2 cm apart from caneer) obtained from 65 patients with GCA were examined for the expression of DAS-1 protein using immunohistoehemistry. The CIM tissues (<2 cm below Z line) obtained from 15 outpatients and inflammatory mucosa from 25 outpatients were also examined for expression of DAS-1 protein. Results The type Ⅲ IM was accounted for 55.4% (36/65) in GCA patients, which was significantly higher than that in outpatients [13.3% (2/15), P<0.01]. The positive rate of DAS-1 expression in cancerous tissues [78.5 % (51/65)] was also significantly higher than that in CIM tissues [38.8 %(30/80), P<0.01]. The expression of DAS-1 protein in IM tissues was gradually increased from type Ⅰ (0%) to type Ⅲ (71.1%) with positive correlation (P<0.01). Conclusions The type Ⅲ IM with over-expression of DAS-1 is closely related to GCA, which might be one of important precancerous lesions for GCA.
8.Establishment of an osteoporosis model in tree shrews by bilateral ovariectomy

Objective To establish an osteoporosis model in tree shrews by bilateral ovariectomy.Methods One hundred healthy 6-24 month old female tree shrews were used in this study.The diagnosis and treatment of osteoporo-sis in the model tree shrews was assessed according to the Interpretation of the Clinical Practice Guideines for Primary Oste-oporosis 2011 released by the Chinese Medical Association of Osteoporosis and Bone Mineral Salt Disease Branch.① Ac-cording to the human natural life span, sexual maturation time and peak bone mineral density ( PBMD) and the tree shrew natural life span and sexual maturation time, we estimated the PBMD of tree shrew in the age of months.Six 6-, 12-, 18-and 24-month old tree shrews in each group were randomly taken for measurement of BMD, and compared the differences between groups to verify the PBMD age of tree shrews.②We chose 62 6-month old tree shrews from the rest of tree shrews to measure their BMD to determine the mean and standard deviation of BMD.Then we divided them into study group ( n =32) and control group (n =30).Bilateral ovariectomy was performed in the study group, while omentectomy was done in the control group.BMD was measured at 3, 6, 9, 12 months after the surgery, 6 animals from each group.The animals which met the criteria of guideline were taken for pathological examination.Results ①The PBMD of 6-, 12-, 18-, 24-month old tree shrews was 0.138 ±0.012, 0.143 ±0.010, 0.141 ±0.009, and 0.139 ±0.009) g/cm2 , respectively, showing no significant differences among them (P>0.05).②The mean PBMD of the tree shrews was 0.143 ±0.016 g/cm2 .③In the model groups, the number of tree shrews whose BMD met the criteria of guideline was 0, 5, 4, 5, respec-tively.The pathological examination of the proximal tibia from the model group showed sparse and some disrupted trabecu-lae and enlarged inter-trabecular space.In the control group, the tibia tissues showed normal histology of bone structure and no one tree shrew showed a BMD value which met the criteria of diagnosis of osteoporosis.Conclusions An osteopo-rosis model in tree shrews can be established at 6 months after bilateral ovariectomy, showing similar manifestation of osteo-porosis in postmenopausal women.The success rate of this modeling method is high and the model is stable.
9.Characterization of influenza B viruses isolated in Hunan Province after pandemic influenza A (H1N1)

Objective To analyze the prevalence status and the genetic characterizations of influenza B viruses isolated in Hunan Province after pandemic influenza A (H1N1) 2009,and to explore possible reasons for the prevalence.MethodsThroat swabs were collected from outpatients with influenza-like illness in 23 sentinel hospitals of Hunan Province in 2010.Influenza viruses were isolated with Madin-Darby canine kidney (MDCK) cells and identified by haemagglutination inhibition test.The genomes of 10 selected influenza B viruses were sequenced and analyzed for phylogenetic and molecular characterization.ResultsWith the reduction of isolation of pandemic influenza A (H1N1)2009 viruses,influenza B virus became the predominant isolated strain in the first half of 2010.Epidemic viruses mainly belonged to the B/Victoria lineage,and both two lineages co-circulated.Seven out of 11 influenza outbreaks caused by type B.Ten strains were filled into 2 branches of BV and BY which were classified by their lineage types in polymerase (PB2,PB1,PA),hemagglutinin (HA),neuraminidase (NA),NB,membrane protein (M1),influenza B virus membrane protein M2 (BM2),and non-structural protein (NS1,NS2) phylogenetic trees except the NP phylogenetic tree in which 10 strains were all in the BY branch.Compared with World Health Organization (WHO) vaccine strains,the amino acid identity of 11 proteins of the 10 strains was high (97.2%-100.0%).However,some amino acid point mutations were found.No mutation was found in drug resistance mutation sites.Some mutations in NA,NB,PB1,PB2 and NS2 molecules were found in 2 strains isolated from outbreaks compared with strains from sentinel surveillance.Conclusions The point mutations,insertions and genetic reassortment indicate viruses sustaining evolution,which is probably the reason for predominant influenza B viruses after pandemic influenza A (H1N1) 2009 in Hunan Province.
10.The analysis of gene expression profile and related signal pathways in focal cortical dysplasia

Objective To investigate the potential pathogenesis of Focal cortical dysplasia (FCD), we performed cDNA microarray analysis to obtain gene expression profile of FCD. Methods Three FCD samples and three normal controls were enrolled. Total RNA of the brain tissues were extracted. The difference gene expressions between FCD group and control group was detected using Affymetrix gene chip. The up and down-regulated genes were confirmed by Real-time PCR. Further, the related signal pathways involved in the pathogenic mechanisms of FCD were predicted by bioinformatics. Result In FCD, two up-regulated genes C21orF2 and AU152162 and 5 down-regulated genes ENPP2, ANLN, IP6K3, UGT8, and AZGP were found. Compared the FCD samples with the normal controls , there were significantly different in all down-regulated genes (P < 0.05), while the up-regulated genes were not (P > 0.05). Using bioinformatics analysis, the ENPP2 , UGT8 , and AZGP1 protein which located in the cell membrane or secreted into the extracellular matrix may be involved in the formation of the myelin sheath and the development of the nervous system by the lipid metabolism and LPA signaling pathway. Conclusion ENPP2, UGT8 and AZGP1 may be involved in pathogenesis of FCD through the process of myelin sheath formation and LPA signal pathway , which warrants further study to know their roles in the pathogenesis of FCD.
