1.The study of mitochondrial DNA in the pathopoiesis mechanisms of paraquat poisoning in vitro

Objective To investigate the effects of mitochondrial DNA (mtDNA) on the pathopoiesis mechanisms of paraquat poisoning in vitro.Methods Firstly,the survival rate of A549 cells (human type Ⅱ alveolar epithelial cells) was measured with cell counting kit-8 after exposure to paraquat.Afterwards,the concentration of mtDNA in supernant of culture medium for culturing A549 and the chauge of mitochondrial membrance potential were detected with absolute quantitative PCR and confocal laser microscopy,respectively.Then,The chemotactic activity of mtDNA in peripheral blood mononuclear cells (PBMC) and neutrophils (PMN) were detected by transwell chemotaxis,and the subtype of chemotactic cells was measured with flow cytometry.Meanwhile,the role of mtDNA in vascular permeability was measured by using Xcelligence system and in vitro using vascular permeability kits.Finally,the effects of mtDNA in cell proliferation were to verify.Results The 50% of lethal concentration (LD50) of paraquat for A549 was 600 μmol/L.Cell viability and concentration of mtDNA following challenge of PQ revealed in a concentration-and time-dependent manner (P < 0.05).The mtDNA had a power in aggregating PBMC nonspecifically,but there was no effect on the vascular permeability was found.Moreover,the proliferation of human fibroblasts was not stimulated directly by mtDNA,but TGF-β1 (transforming growth factor-beta 1),a major pro-fibrotic factor,was increased compared to control group (P < 0.05).Conclusions The mtDNA could play an important role in the inflammatory and proliferation responses to paraquat poisoning.

3.The effects of different pacing sites of right ventricle on serum N-terminal Pro brain natriuretic peptide level and left ventricular systolic function

Objective To investigate the effects of different pacing sites of right ventricle on serum N terminal Pro brain natriuretic peptide (NT-ProBNP) and left ventricular systolic function.Methods A total of 76 patients with an implanted DDD pacemaker were randomly divided into right ventricular septal pacing group (RVSP group,n=40) and right ventricular apex pacing group (RVAP group,n=36) according to the ventricular leads position.Serum NT-proBNP level,left ventricular end diastolic dimension(LVEDD)and left ventricular ejection fraction(LVEF)were analyzed before and 6 months after operation in the two groups.Results There was no difference in serum NT-proBNP level between the two groups before operation,but the serum NT-proBNP level increased in both groups 6 months after operation,and it was higher in RVAP group than in RVSP group (P<0.05).There were no significant differences in LVEDD and LVEF in RVSP group before and after implantation (P>0.05).Compared with pre-implantation,LVEDD was increased and LVEF was decreased in RVAP group 6 months after implantation (both P<0.05).Linear correlation analysis showed that serum NT-proBNP level was negatively correlated to LVEF (2γ=-0.76,P<0.05).Conclusions Compared with RVAP,RVSP can keep the normal sequence of electrical activity and exert less adverse effects on left ventricular systolic function.Therefore,RVS is an ideal pacing location.
4.The expression and clinical significance of matrix metalloproteinase-2 in human pancreatic carcinoma.

Objective To investigate the expression of matrix metalloproteinase-2 (MMP-2)in pancreatic carcinoma and the relationship between MMP-2 with tumour clinicopatholngical features. Methods The expression of MMP-2 was detected by S-P immunohistochemistry in 36 cases with pancreatic carcinoma and 14 normal pancreat-ic tissues. Results The positive expression rate of MMP-2 was 66.7% (24/36) in pancreatic carcinoma tissue and 14.3% (2/14) in normal pancreatic tissues (χ2 = 3. 587, P < 0.01 ) ;The expression rate of MMP-2 in pancreatic carcinoma tissue with positive-node was 86.7% ( 13/15 ) ,which was higher than that with negative-node,which was 52.4% ( 11/21 ) ( P < 0.05 ) ; As to TNM staging in pancreatic carcinoma, the expression rate of MMP-2 was 41.2% (7/17) with Ⅰ,Ⅱ staging and 89.5% (17/19) with Ⅲ,Ⅳ staging(χ2=9.418,P <0. 01 ) ;The expression rate of MMP-2 was 50.0% (5/10) ,66.7% (10/15) and 72.8% (8/11) in highly,moderately and poorly differentiated pancreatic carcinoma(P > 0.05 ). Conclusions The expression of MMP-2 is strengthened significantly in pancreatic carcinoma tissue and involved in turnout invasion and metastasis features; MMP-2 might be regarded as one more marker for the invasive properties of pancreatic carcinoma.
5.The correlation of father involvement with social adaptation of preschool children

Objective To investigate the correlation of father involvement with early social adaptation behaviors.Methods With social adaptation scale for infant-junior middle school' s student and a self-made questionnaire,1246 normal children,age from 16 to 72 months,were investigated.Regression analysis was performed to get the correlation of social adaptation with time and activities of father involved.Results After controlling for mother-,child- and family-related factors,the regression analysis showed that standard score of social adaptation was positively rehted with the time of father involved ( β =0.05,P=0.03 ),the time father invloved explain 12%of the variation of social adaptation.The father-child activities were related with social adaptation behavior,playing was related with work skills (β =0.09,P<0.01 ) and socialization ( β =0.06,P<0.05),story listening related with communication ( β =0.07,P < 0.05) and self-management ( β =0.06,P < 0.05 ),other activities related with self-dependence ( β =0.07,P<0.05) and work skills ( β =0.06,P<0.05).Conclusion The time and activities of father involved are related with children' s social adaptation.The effect of father involvement on early social adaptation is positive.
6.Establishing the System of Academic Assessment and Evaluation on Introduced Talented Personnel

Objective To evaluate the system of academic assessment and evaluation on introduced talented personnel so as to establish a consummate talented personnel introduction mechanism and to further standardize the introduction work. Methods On the basis of referring to document papers and soliciting management as well as human resource experts iteratively,the research method of expert consultation was adopted.(Results)The index system of academic assessment and evaluation on introduced talented personnel in school of medicine was preliminary established,and the objective index and subjective evaluation were scientifically combined to form the method of academic assessment and evaluation on introduced talented personnel. Conclusion Through simulative assessment and evaluation as well as probational demonstration,there exits definite scientificality and applicability on this academic assessment and evaluation index system,and it can provide a properly objective evaluation and essential reference foundation in the aspect of introducing more advanced talented personnel in the near future.
7.ABCF2 positive expression and its relationship with chemotherapy resistance in ovarian carcinoma

Objective To investigate the positive expression of ATP binding cassette transporter protein F2 (ABCF2) in ovarian cancer tissue and its relationship with chemotherapy resistance.Methods 91 patients with ovarian cancer specimens were selected and 42 cases from the tumor 5cm adjacent tissues as normal controls, ABCF2 expression in each sample was detected by immunohistochemical staining , the relationship between ABCF2 expression and clinicopathological features of patients with ovarian cancer were analyzed .Results The positive expression rate of ABCF2 in ovarian cancer tissue was 68.13%, which was significantly higher than that in normal ovarian tissue (9.52%), and the difference was statistically significant (P<0.05).The positive expression of ABCF2 was not related to the age and organization type of ovarian cancer patients.The positive expression rate of ABCF2 in stage III~IV was 86.54%, which was significantly higher than that of phase I~II (43.59%), the difference was statistically significant (P<0.05). The positive expression rate of ABCF2 in patients with chemotherapy resistance was 88.89%, which was significantly higher than that in patients with chemotherapy sensitivity ( 59.38%) , and the difference was statistically significant ( P<0.05 ) .The results of Logistic regression analysis showed that ABCF2 positive expression was the independent influencing factor of ovarian cancer drug resistance (OR =4.586,95% CI:1.121 ~3.392,P<0.05). Conclusion ABCF2 is highly expressed in ovarian cancer, which may be associated with chemotherapy resistance in patients with ovarian cancer .
8.Determination of Yohimbine in Corynanthe Yohimbe K. Schum. by HPLC

OBJECTIVE:To establish a HPLC method for the determination of yohimbine in corynanthe yohimbe K.schum.METHODS:The analysis was performed on C18 column with column under room temperature.The mobile phase consisted of methanol-phosphoric acid buffer-triethylamine(30∶ 70∶ 0.5,pH5.0)with a flow rate of 1.0mL? min-1.The wavelength was set at 270nm.RESULTS:There was a good linear relationship when the sample size of yohimbine was over a range of 10~ 1 000? g? mL-1(r=0.999 8),with recovery at 96.5% and precision at 1.23%.CONCLUSION:This method was simple and reliable,and suitable for the determination of content of yohimbine.

10.Prediction of Elimination of Leprosy in Leprosy Endemic Areas of China

Objective To have insight into the epidemiological situation and to predict the possibilities for achieving the national goal of basic eradication or the WHO target of elimination of leprosy as a public health in the leprosy high- endemic areas in China. Methods Based upon the data during 1983- 1996 from the National Leprosy Recording and Reporting Surveillance System and using the appropriate mathematical models, the expected calendar year of reaching the national goal in terms of detection and prevalence rates and the WHO target of elimination under different conditions was predicted. Results Of 337 counties where the national goal of basic eradication of leprosy had not reached and 40 counties where the WHO goal of leprosy elimination had not achieved in 1996, the detection rates in calendar years followed the negatively exponential models with a significant goodness- of- fit in 67 and 11 counties, respectively. In the former, the national goal can be met in 6% of counties before the year 2000 or 34.4% before 2010 in terms of detection rate, or 31.3% before the year 2010 in terms of prevalence rate. In the latter, the WHO target can be met in 8- 10 counties within this century when the duration of disease was determined with the WHO definition. While the MB proportion among new cases increased by 10% , the target would be met one year later. However, at the same MB proportion, the change of the duration of completing the fixed treatment from PB 0.5 year and MB 2 years to PB 0.75 year and MB 3 years will result in the achievement of the goal 2- 10 years later. Conclusion The results imply that WHO goal of leprosy elimination can be reached in more than 95% of counties by the end of this century, but the national goal of achieving basic eradication of leprosy in more than 95% of counties by this century will not be met, indicating that leprosy control will go beyond the century in China.
