2.Antibacterial Cecropin and Its Application on Transgenic Plants for Bacterium Resistance

Cecropin is a kind of heat-durable and broad-spectrum antibacterial polypeptides which has strong effect against bacteria,fungi,virus and some pathogenic microorganisms.Today Cecropin has been widely applied into plant genetic engineering,antiviral study,and inhibiting tumor cell proliferation.Its Structure-function relationship,antibacterial mechanism,and the application on transgenic plants for bacterium resistance was reviewed.Expression of Cecropin in plants has a great application potential in bacterium resistance.Deep analyses and research of molecular structure and action mechanism can promote the transgenic plants antibacterial research.
3.Analysis of the Application of TCM Injection in 3 Hospitals from Jiangsu Changshu during 2013 to 2015

OBJECTIVE:To provide reference for rational use of TCM injection in the clinic. METHODS:In retrospective survey,the application of TCM injections in 3 hospitals from Changshu area during 2013-2015 were analyzed statistically in re-spects of consumption sum,annual growth rate,constituent ratio,DDDs,B/A,DDC,etc. RESULTS:During 2013-2015,in the sample hospitals,consumption sum of TCM injections in 2014 and 2015 declined by 5.27% and 15.92%,compared with last year. The proportion of consumption sum of TCM injections in total consumption sum were 4.40% in 2013,3.86% in 2014 and 3.13%in 2015. Top 3 TCM injections in the list of consumption sum and DDDs were blood activating and stasis eliminating drugs,the re-suscitation and reviving yang first aid class drugs and tonic drugs. The top single types in the list of consumption sum were Xing-naojing injection,Shuxuetong injection,Xuebijing injection,Shenmai injection and Xue shuantong for injection;the top single types in the list of DDDs were Shenmai injection,Reduning injection,Shuxuetong for injection,etc. B/A of Shenmai injection and Reduning injection were higher,while DDC of Xuebijing injection was the highest. CONCLUSIONS:The utilization of TCM injec-tion is basically reasonable,and shows decreasing tendency in sample hospitals,but great importance should be attached to the safe-ty and effectiveness of TCM injections and reasonable choice.
4.Effects of Lung Recruitment Maneuvers in Patients with Acute Respiration Distress Syndrome

Objective To investigate the clinical effects, therapy safety and maintenance time of recruitment maneuvers (RM) combined with lung protective strategy in patients with acute respiratory distress syndrome (ARDS). Methods Thirty-two ventilated patients with ARDS underwent RM using a pressure of 30cmH_2O for 30s, without any sedatives and neuromuscular blocking agents. Results Compared with before RM, both oxhemoglobin saturation measured by pulse oximetry (SpO_2) and partial pressure of arterial oxygen (PaO_2) were significantly increased during one hour after applying RM. SpO_2 increased significantly within two hours after RM. 5.Effect of lymphovascular invasion on prognosis in high-grade pT1 bladder cancer after first transurethral resection of bladder tumor

Objective To explore the effect of lymphovascular invasion (LVI) on prognosis in the high-grade stage pT1 bladder cancer after first transurethral resection of bladder tumor (TURBT). Methods Thirty-five high-grade stage pT1 bladder cancer patients who underwent first TURBT and showed LVI positive according to the postoperative pathological (observation group) and 70 high-grade stage pT1 bladder cancer patients who underwent first TURBT and showed LVI negative according to the postoperative pathological (control group) were enrolled into the study. The tumor recurrence rate, tumor progression rate, recurrence-free survival (RFS) and overall survival (OS) were compared between 2 groups. Results The follow-up time in observation group was 15 - 71 (42.1 ± 18.1) months, and in control group was 20-72 (50.9 ± 12.2) months. The tumor recurrence rate and tumor progression rate in observation group were significantly higher than those in control group:88.6%(31/35) vs. 34.3%(24/70) and 54.3%(19/35) vs. 22.9%(16/70), and there were statistical differences (P<0.01). The RFS and OS in observation group were significantly shorter than those in control group: (19.7 ± 4.5) months vs. (46.8 ± 9.2) months and (43.4 ± 12.6) months vs. (63.4 ± 7.1) months, and there were statistical differences (P<0.05). Conclusions There is a significant correlation between LVI and tumor recurrence and progression in the high-grade pT1 bladder cancer after first TURBT, and the LVI can predict disease progression and poor prognosis.

7.Acupuncture Treatment of 106 Cases of Peripheral Facial Paralysis in Different Stages

In the treatment of facial paralysis in the acute stage and restoration stage by shallow needling technique and penetrating needling technique respectively, totally 106 cases, the effective rate is respectively 98.6% and 83.9%, indicating that the curative rate is high if early treatment is given.
8.Expression and clinical significance of SIRT1 in hepatocellular carcinoma

Objective To investigate the clinical significance of silent mating-type information regulation 2 homologue 1 (SIRT1) in hepatocellular carcinoma (HCC).Method We analyzed p53 mutation by gene sequencing and activation of SIRT1 and AMP-actived protein kinase (AMPK) using western-blot in 252 patients with hepatitis B virus-positive HCC.Results A higher proportion of tissues with mutant p53 were demonstrated to harbor activated SIRT1 (64.8% vs 31.8% ; P < 0.01).Activated SIRT1 predicted a longer relapse-free survival.On multivariate analysis,activated SIRT1 remained significant (OR:0.339,CI:0.160-0.720,P =0.005).Analysis of 252 paired specimens revealed a significant correlation between activated SIRT1 and activated AMPK in HCC tissues harboring mutant p53 (P =0.007).Conclusion SIRT1 exerted anti-carcinogenic effects through the AMPK pathway in HCC in the context of mutant p53.
9.Study on Quality Control of Ethyl-acetate Parts ofFerula Sinkiangensis

Objective To establish the method for quality control of ethyl-acetate parts of Ferula sinkiangesis. Methods HPLC was used to detect the contents of ferulic acid, farnesiferol A and farnesiferol C. Waters XTerra RPC18 column (250 mm×4.6 mm, 5 μm) was used; acetonitrile-0.1% phosphoric acid was as mobile phase with gradient elution; flow rate was 1.0 mL/min; detective wavelength was 324 nm; temperature was 30℃; sample volume was 10 μL ResultsFerulic acid, farnesiferol A, and farnesiferol C showed a good linear relationship range from 0.05– 1.0 mg/mL, 0.132–2.64 mg/mL, and 0.118–2.36 mg/mL, respectively. The average recovery rates were 99.34%, 98.96% and 99.24% respectively. The contents of ferulic acid, farnesiferol A and farnesiferol C were 33.4, 76.5, 72.3 mg/g respectively.Conclusion The method was simple, accurate and repeatable, with good repeatability and stability, which can be used for the quality control of ethyl-acetate parts ofFerula sinkiangesis.
10.The effect of microwave ablation combined with dendritic cell tumor vaccine treatment on mouse mammary carcinoma and its immunological mechanism

Objective To investigate the therapeutic effects of microwave ablation combined with dendritic cell (DC) tumor vaccine on a murine breast cancer model and to explore its immunological mechanism.Methods Forty-eight Balb/c mice were used to establish models of mouse mammary carcinoma,and then randomly divided into a sham-operation group,a microwave ablation treated group,a DC tumor vaccinated group and a microwave ablation plus DC tumor vaccinated group.Microwave ablation was conducted with the microwave ablation group on day 14 after establishing the model.DC tumor vaccination was performed on day 7 in the vaccinated group.In the microwave ablation plus DC tumor vaccinated group,DC tumor vaccination and microwave ablation were consecutively performed on days 7 and 14,respectively.Tumor growth,lung metastasis and survival were observed.The proportion of CD3+ and CD4+ T cells in splenocytes were analyzed by fluorescence Activated Cell Sorter (FACS) on the 14th day after microwave ablation.Serum levels of interferon-γ (IFN-γ) and interleukin-4 (IL-4) were also detected using ELISA.Results The survival time (56.4 ± 4.3 days),tumor size (67.3 ± 6.1 mm2) and lung metastasis index (0.6 ± 0.4) in the microwave ablation plus DC tumor vaccinated group were all significantly lower than the corresponding averages in the microwave ablation and DC tumor vaccinated group.The proportions of splenic CD3 + T cells (27.8 ±10.3 %) and CD4+ T cells (16.7 ± 8.3 %) in the microwave ablation plus DC tumor vaccinated group were significantly larger than in the control group.Average serum levels of IFN-γ (41.8 ± 22.9 pg/ml) in the microwave ablation plus DC tumor vaccinated group were also significantly larger than in the control group,while the average serum level of IL-4 (15.4± 6.4 pg/ml) was significantly lower.Conclusions Microwave ablation and DC tumor vaccination together could have synergistic effects in treating experimental breast cancer,which might be closely related to the enhanced immune response.
