1.Inhibition effects of gefitinib on non-small cell lung cancer cell lines in vitro

Objective To explore the mechanism of gefitinib in the treatment of non-small cell lung cancer (NSCLC) cell lines in vitro. Methods The effects of the gefitinib in five human NSCLC cell lines (A549, SPC-A-1, H460, H1229, 95D) were studied. The inhibition of cell proliferation in each group were measured by CCK8;The status of apoptosis cells were observed using flow cytometry after PI marked;invasion of lung cancer cell inhibited by gefitinib were assessed by transwell technique;The drug was detected by western blot on the proliferation-related signaling protein. Results The proliferation and invasive capacity of NSCLC cells were inhibited by gefitinib (P < 0.05). In gefitinib group, the apoptosis rates of A549 [(9.6±0.73) %]and SPC-A-1[(14.3±1.12) %]were higher than that of control group[(3.1±0.29) %](t =11.16,P =0.001;t =4.726, P =0.009). Expression of p-AKT, p-EGFR, p-MAPK protein levels were significantly down regulated in A549 cells when gefitinib was given after 72 hours (t =6.656, P =0.003;t =16.441, P =0.0001;t =3.736, P =0.020). Conclusion Gefitinib can inhibit the proliferation and invasion of lung cancer cell, also can induce apoptosis in vitro and most likely to contribute to the inhibition of key enzymes in EGFR signaling transduction pathway.

Methods:According to the needs of medical equipment management, the problems of medical equipment archives system would be analyzed and addressed. Results:In recent medical equipment archives system, the database of usage rate, cost-effectiveness, maintenance costs and property management should be improved. Conclusion:The improved archives system could provide reliable basic data and multi-faceted sharing for medical equipment dynamic management.
4.The Distribution of Ammonia Nitrogen in Groundwater of Shunyi and Its Influencing Factors

Objective To discover the distribution of ammonia nitrogen in groundwater of Shunyi District and its influencing factors. Methods 286 monitored points in low-flow period and 100 in high-flow period were chosen in 19 towns in Shunyi District. Concentrations of ammonia nitrogen, iron and manganese, and the total bacterial count of water samples were determined and evaluated based on the Guidelines for Drinking Water Quality and related standard during Jan.-Mar., Aug.-Sep., in 2005. Results All the monitored points were deep groundwater. The median of ammonia nitrogen concentration in groundwater was 0.010 mg/L in low-flow period, and 0.140 mg/L in high-flow period. The relativity was not found between the concentration of ammonia nitrogen,total bacterial count in water sample and pollution source around the well. Fourty - seven cases were treated with this remedy, a~ mong them, 26 cases were markedly effective, 12 cases abated, 4 cases ineffective, the total effective rate being 91. 5%. The results were apparently superior to the blank control group. The mechanism of such effectiveness lies in the improvement of evacuation of gastric content and gastrointestinal movements.
6.Effects of nimodipine on the changes of calcium, magnesium and calmodulin contents after cerebral ischemia in rats

The changes of calcium and magnesium content in brain tissue (BCa;BMg) and in the mitochondria (MCa,MMg) and calmodulin content in the cortex and hippocampus (CCaM,HCaM) were determined in the rats with 4-vessel-ligation cerebral ischemia.And then the therapeutic effects of nimodipine were observed during ischemic and.reperfusion stages of the brain.It was found that BMg and BMg were decreased and CCaM HCaM increased during both the ischemic and reperfusion stages and BCa and MCo were increased during the reperfusion stage.The fact that calcium content was only increased during the reperfusion stage indicates that the influx of calcium plays an important role to precipitate injury during the reperfusion stage.Adminstration of nimodipine could prevent the increase of calcium and calmodulin and the decrease of magnesium after cerebral ischemia.
7.The changes of brain natriuretic peptide in patients with myocardial infarction

Objective To observe the changes of brain natriuretic peptide(BNP)in patients with myocardial infarction,and to assess the clinical value of BNP in the patients with myocardial infarction.Methods BNP was determined in 91 patients with myocardial infarction and in 90 normal cases,and the difference was studied between the area of myocardial infarction and the number of coronary arteries stenosis.Results BNP(ng/L)(155.03 ± 113.45,71.03 ±37.01)and LVEDd(cm)(5.11 ±0.51,4.56 ±0.43)were significantly increased in patients with myocardial infarction than that in normal cases(P<0.01),but EF(%)(50.84 ±7.98,57.09 ±6.29)was lower than that in normal cases(P<0.01).BNP and LVEDd were also significantly higher in patients with wide-anterior myocardial infarction than that in antero-septal,inferior and posterior wall myocardial infarction,but LVEF was lower.Conclusion The BNP level was higher in patients with myocardial infarction,especially in patients with wide-anterior myocardial infarction.BNP was a reliable bio-chemical marker which can evaluate the degree and prognosis of patients with myocardial infarction.

9.Down regulation of miR-33a is involved in gemcitabine chemoresistance in human pancreatic cancer

Background and purpose:Pancreatic cancer is one of the most deadly human malignant neoplasms. Resistance to chemotherapeutic drugs is a major reason responsible for poor prognosis in the treatment of pancreatic cancer patients. MicroRNA (miRNA, miR) is a family of small non-coding RNA molecules, dysregulated miRNA is associated with various tumor biological function. miR-33a has been widely reported as a metabolism-related miRNA, while its relationship with drug resistance has little understand. This study was focused on the effect of miR-33a on gemcitabine chemoresistance in pancreatic cancer to bring the novel theoretical basis to chemotherapy for pancreatic cancer.Methods:In situ hybridization and Real-time PCR were used to analyze the miR-33a expressions in pancreatic cancer tissue sample and cell lines, respectively. Cell counting kit 8 (CCK-8) assay was used to calculate the IC50 value of different pancreatic cancer cells.Results:miR-33a was down-regulated in pancreatic cancer tissue and cell lines compared with para-cancerous tissues and normal HEK293T cells. Moreover, miR-33a over expression not only could enhance the chemosensitivity to gemcitabine in pancreatic cancer cells, but also rescue the gemcitabine resistance in pancreatic cancer cells.Conclusion:Down regulation of miR-33a in pancreatic cancer decreases the chemosensitivity to gemcitabine, resulting in development of acquired gemcitabine chemoresistance. It provides the theoretical basis to develop a new molecular targeted drug to combine with chemotherapy for pancreatic cancer.
10.Images change of pulsed Doppler and tissue Doppler in evaluation of right ventricular diastolic function in patients with essential hypertension

ObjectiveTo assess right ventricular diastolic function in patients with essential hypertension using tricuspid valve flow spectrum and tricuspid annulars motion dectected by echocardiography. MethodsAccording to Ganau′s categories, 124 patients with essential hypertension were enrolled,which were divided into four groups:normal left ventricular configurational group (group Ⅰ, n =32 ), centripetal reconstruction group (group Ⅱ, n =30), centripetal hypertrophy group ( group Ⅲ, n =32), eccentric hypertrophy group ( group Ⅳ, n =30 ). Control group consisted of 35 normal subjects pulsed wave Doppler(PW) data and tissue Doppler imaging(TDI) data were acquired from tricuspid and its' annulus respectively on apical four chamber view. ResultsEPA > 1 was observed in control group as well as group Ⅰ and Ⅱ ,while E/A < 1 in group Ⅲ and group Ⅳ ,there were significant differences between them. Em/Am < 1 was observed in all of patients with hypertension,there were significant differences among them( P < 0.05 ) .ConclusionTissue Doppler imaging was more sensitive than pulse Doppler in evaluating decreased right ventricular diastolic function in patients with essential hypertension, and the decrement of the function was different in degree based on variant left ventricular configurations.
