Journal of Applied Clinical Pediatrics 2004;0(11):-
Recent studies have revealed that tumor suppressor PTEN does not only closely relate to the development and differentiation of brain but also get extensively involved into the pathophysiological processes of ischemic brain injury.Through research on the mechanisms of molecular regulation mediated by PTEN in ischemic brain injury will provide new perspectives for the treatment of ischemic brain injury.

Objective To explore the effect of behavioral training on the differentiation of neural stem cells in the dental gyrus (DG) in rats with hippocampus infarction. Methods Seventy-eight Sprague-Dawley rats were randomized into infarction plus behavior training group, infarction group and control group. Photochemistry method was used to induce hippocampal infarction in rats of the infarction plus behavioral training group and infarc-tion group. At 1 day after surgery, Morris water maze training was used for infarction plus behavioral training group, free-movement without training was performed for infarction group. Double staining immunofluorescence was used to detect the co-expression of bromodeoxyuridine (BrdU) with neuronal nuclei ( NeuN ) or glia fibrillary acidic protein (GFAP) in the DG at different time points. Results Few BrdU/NeuN and BrdU/GFAP double staining cells were observed in the DG of control rats. In the infarction group and infarction plus behavioral training group the number of BrdU/NeuN and BrdU/GFAP double-stained cells increased in the DG on the opposite side compared with the control group on 14th, 21st, 28th and 35th days after surgery (P < 0.05 ). There observed significantly more BrdU/NeuN and BrdU/GFAP double-stained cells in the infarction plus behavioral training group than that in the infarction group on the 14th, 21st, 28th and 35th days after surgery ( P < 0.05 ). Conclusion Behavioral training can accelerate the differentiation of neural stem cells to neuron and astrocyte, by which to promote the re-covery of neural functions.

Objective To investigate the effects of reciprocal inhibition on motor function connectivity in the brains of stroke patients. Methods Thirty patients with stroke were randomly divided into a treatment group (n =15) and a control group (n =15).The control group underwent normal limb positioning, medium frequency electrotherapy, circulated compression of the limbs, etc. The treatment group received conventional rehabilitation treatment plus reciprocal inhibition treatment for 30 min daily,6 times a week for 4 weeks. All of the patients were assessed before and after treatment using the Canadian neurological scale (CNS),the Frenchay activities index (FAI),the motricity index (MI) and functional magnetic resonance imaging of the motor cortex in a resting state (rs-fMRI).Results In both groups the average CNS,FAI and MI scores improved significantly.Compared with the control group, the changes in FAI and MI scores in the treatment group improved significantly more. The coefficient of functional connectivity of the bilateral motor cortex decreased significantly after treatment in both groups. In the treatment group the motor cortex functional connectivity correlated significantly with the improvements in MI scores. Conclusions Reciprocal inhibition can accelerate the improvement of extremity motor function and ability in the activities of daily living significantly after stroke. It reduces functional connectivity in the bilateral motor cortex in ways significantly correlated with improvements in motor function.

Phosphatase and tensin homology deleted on chromosome 10 (PTEN) is a tumor suppressor with dual protein and lipid phosphatase activity.PTEN is involved multisystem diseases and cancer, though the mechanisms of PTEN regulation are far from clear.Recent advances concerning regulation of PTEN including protein-protein interaction, phosphorylation, ubiquitination, oxidation and acetylation will be discussed.

Phosphatase and tensin homologue deleted on chromosome 10(PTEN) is a tumor suppressor which can inhibit proliferation and migration and control apoptosis in a number of cell types, mainly through inhibiting the phosphoinositide 3-kinase(PI3K) signaling pathway.A number of in vitro and in vivo studies has been instrumental in uncovering a direct correlation between deregulated PTEN/PI3K signaling and changes in neuronal morphogenesis, which is likely to have profound bearings upon the pathogenesis of neurological symptoms. This review outlines recent work on the function of PTEN during the formation and maintenance of neuronal circuits in the brain.

Objective Comparatively studying the method, efficiency and anti-hypoxia ability of cardiomyocytes which are directionally induced from human embryonic stem cells and mouse embryonic stem cells in vitro so as to provide an experimental basis for further study of inducing the differentiation of embryonic stem cells into cardiomyocytes in vitro. Methods Human embryonic stem cells are induced into cardiomyocytes by suspension method which dosen't using inducersand adherence method which using inducers respectively.Mouse embryonic stem cells are induced into cardiomyocytes by hanging drop method which dosent using inducers and using inducer respectively.Staining the specific marker cTnT of cardiomyocytes by immunofluorescence.Comparing the time, percentage and beating frequency of cardiomyocytes by counting under the microscope.Using apoptosis-hoechst staining kit to detect the apoptosis ratio of beating cardiomyocytes which have been treated by hypoxia with 24 h.Results The group of hESC without inducers results in that the mean time days of appearing beating cardiomyocytes is (13.9 ± 0.9) days, the percentage is 20.8 % and the average frequency of beating is (63.8 ± 5.6) times/min.The group of hESC with inducers results in that the mean time days of appearing beating cardiomyocytes is (13.0 ± 1.1) days, the percentage is 66.7% and the average frequency of beating is (63.0 ± 7.0) times/min.The group of mESC without inducers results in that the mean time days of appearing beating cardiomyocytes is (14.3 ± 1.0) days, the percentage is 12.5% and the average frequency of beating is (80.2 ± 3.9) times/min.The group of mESC with inducers results in that the mean time days of appearing beating cardiomyocytes is (12.2 ± 1.2) days, the percentage is 81.3% and the average frequency of beating is (79.9 ±7.7) times/min.Beating cardiomyocytes of each group are positive to cTnT staining.Different apoptosis ratio are detected of beating cardiomyocytes of each group. Conclusion The four methods can all successfully induce the differentiation of embryonic stem cells into myocardiocytes, and the adherent method of hESC induced with activin A + BMP4 is the first successful induction in China. The groups adding inducers improve the differentiation efficiency more significantly than the groups without adding inducers.Inducing mouse embryonic stem cells into cardiomyocytes is more simple and efficient compared with human embryonic stem cells.Without the presence of other protective factor,anti-hypoxia ability of cardiomyocytes induced from human embryonic stem cells is stronger and the beating time are longer in vitro compared with mouse embryonic stem cells.

Journal of Applied Clinical Pediatrics 1986;0(02):-
Objective To analyze the etiologic factors and clinical manifestation of intraventricular hemorrhage (IVH) in preterm infants. Methods One hundred and seventy-two preterm infants from June 2005 to August 2006 were accrued to investigate their gestational age, birth weight, birth history, and clinical symptoms.Cranial chromatic ultrasound was used to scan the preterm infants and diagnose IVH.Results 1.The incidence of IVH was associated with gestational age (?2=6.40 P=0.011);2.The incidence of IVH was also associated with birth weight(?2=26.49 P=0);3.IVH usually occurred within 72 h with mild clinical manifestations and was diagnosed within 5 days after birth;4.IVH occurred more frequently and more severe in infants with severe asphyxia than those with mild asphyxia. Conclusions Early gestational age,low birth weight, and severe asphyxia are risk factors for IVH.The clinical symptoms of IVH are usually mild in most patients.Cranial chromatic ultrasound is a reliable, sensitive and convenient method of detection for IVH in preterm infants.
