The incidence of primary nonfunction in the early period of liver transplantation,is reported to be 2%-23%,which is the main cause of death of liver transplant patients.It can be induced by many complicated factors and is particularly associated with the pathological changes caused by acute immunological rejection.Current studies of liver transplantation are focused on how to reduce the injury and improve the function of the liver graft and raise the success rate of liver transplantation.

Objective To understand the basic situation of blood donors and their anti-HIV antibody screening in a hospital of Madagascar during 2008- 2009 in order to provide the reference information for giving better medical aid and promoting the devel-opment of the clinical transfusion in Madagascar.Methods The blood doner′s personal informations were understood by inquiry and the anti-HIV antibody was detected by the rapid testing,the results were recorded,the constituent ratio of the data was calcu-lated and the chi-square test was performed.Results There were 2 298 blood donors including teenages less than 15 years old dur-ing 2008-2009,female doners were more than male donors,among them 366 cases were pregrant women.The blood donors from village were more than those from city,the education level was dominated by the middle school students,accounting for 51.65%. There were 22 tuberculosis patients in 2008,none in 2009.The detection rate of anti-HIV antibody was 0.04% for these 2 years. Conclusion The protection on the blood donors in Africa area is not optimistic.The phenomenon of pregrant women and teenages as the blood donors exists in Madagascar.

Objective To detect the expression of Sushi Repeat Containing Protein X Linked 2 (SRPX2) in hunan endometrial carcinoma (EC) and its role in cell proliferation and apoptosis Methods Immunohistochemistry was applied to detect the expression of SRPX2 in EC and matched tumor-adjacent tissues (n =50).The correlation between SRPX2 expression and clinical features was analyzed by Pearson chi-square test.siRNA was used to down-regulate the expression of SRPX2 in HEC-1A cells.The effects of SRPX2 on cell proliferation and apoptosis were measured by CCK-8 assay and Flow cytometer,respectively.The FAK and Cyclin D1 expression were detected after silencing SRPX2 expression.Results The expression of SRPX2 was significantly up-regulated in EC tissues,and the high expression of SRPX2 was associated with the large tumor size (＞3 cm) and advanced FIGO stage (Ⅲ+Ⅳ stage).Silencing SRPX2 could inhibit cell proliferation,induce cell apoptosis and repress FAK and Cyclin D1 expression in HEC-1A cells.Conclusion The expression of SRPX2 is significantly higher in EC tissues,which is closely related with cell proliferation.Scilencing SRPX2 could achieve an anti-cancer role through inhibiting cell proliferation and inducing cell apoptosis by the down-regulation of SRPX2.

Nucleic acids, as a next generation of biotechnology drugs, not only can fundamentally treat diseases, but also own significant platform characteristics in view of technology and production. Therefore, nucleic acid-based drugs have broad clinical applications in biomedical fields. However, nucleic acids are degradable and unstable, and have very low intracellular delivery efficiency in vitro and in vivo, which greatly limits their applications. In recent years, ionizable lipid-based lipid nanoparticles have shown promising application potentials and have been successfully applied to COVID-19 (Coronavirus Disease 2019) vaccines in clinic. Lipid nanoparticles demonstrate high in vivo delivery efficiency and good safety profile due to their unique structural and physicochemical properties, which provides many possibilities for their clinical applications for nucleic acid delivery in the future. This review focused on the characteristics of nucleic acid drugs and their delivery barriers, and discussed the approved nucleic acid drugs to illustrate the key aspects of the success of their delivery carrier system. In addition, problems to be solved in the field were highlighted.

BACKGROUND: The renal transplanted recipients were in poor immunosuppressive state. Compared to common person, the bladder transitional carcinoma in recipients was aggressive and easy to recurrence. Looking for a more effective therapy method to decrease the recurrence of recipients' bladder transitional carcinoma is the hot and difficult problem in clinical study.OBJECTIVE: To analyze the efficacy and safety of submucosal injection epirubicin following transurethral resection of bladder tumor (TUR-Bt) to treat the recurrence of bladder transitional cell carcinoma in renal transplantation recipients.METHODS: Totally 9 renal transplantation recipients with transitional cell carcinoma of bladder were retrospectively studied. The patients' periods without cancer, the frequency of recurrence within one year, the rates of side effect, the changes of tumor grading following recurrence and allograff function were recorded when the routine method and submucosal injection epirubicin following TUR-Bt were used in different period respectively.RESULTS AND CONCLUSION: Submucosal injection epirubicin following transurethral resection of bladder tumor was safe and effective to treat bladder transitional cell carcinoma recurrence in renal transplantation recipients. Compared to the routine perfusion, periods without cancer and the frequency of recurrence within 1 year were significantly decreased, which can elevate recipients life quality and long-term survival rates.

Eukaryotic DNA element called Matrix Attachment Regions (MARs) can function on regulating the structure and activity of chromosome. Traditional quantitation in vitro and indirect functional analysis can not always reflect MAR-involved physiological state. In order to study transcription regulation and make a try in methodism,? 1-antitrypsin MAR (?1-AT MAR) is cloned and incorporated into pEGFP-C1 vector. Non-MAR-containing and MAR-containing plasmids were then transfected into HEK-293 cells with LipofectamineTM 2000 respectively. Positive cell clones were assayed after 20 days of selection by G418. Semiquantitative RT-PCR and fluorescence microscope analysis show that this MAR has a positive effect on modulating nearby gene expression. Further, co-localization with newly CMV promoter and RNA polymeraseⅡ(RNAPⅡ) was detected by chromatin immunoprecipitation (ChIP), The PCR result demonstrates that more RNAPⅡwas recruited to the CMV promoter to initiate transcription in presence of MAR. ChIP can be used to confirm the MAR-mediated transcriptional activation and provide more reliable information than RT-PCR in real time. The technology is also providing a platform for our research in gene expression regulation.

OBJECTIVE:To observe the effect of peripheral nerve block on the biology of spinal cord nerve cel s.
METHODS: Sixty New Zealand rabbits were selected and randomly divided into three groups: ischiadic nerve block group, subarachnoid block group and extradural nerve block group. Each group had 20 rabbits which were sub-divided into experimental group and control group with 10 rabbits in each subgroup. The rabbits in the three experimental groups were injected with lidocaine, bupivacaine and lidocaine at the middle point between femoral head and ischiadic tuberosity in ischiadic nerve block group, subarachnoid block group and extradural nerve block group, respectively. The rabbits in each control group were injected with normal saline at the same position.
RESULTS AND CONCLUSION:After ischiadic nerve block, extradural nerve block or subarachnoid block for 30 minutes, the Nigeria’s bodies of the little round cel s in the poliomyelia posterior horn laminaeⅢandⅣand the polyhedral cel s in the anterior horn laminaeⅨwere less than those in the control groups. Nuclei leaned towards one side. c-Fos protein expression was decreased or showed no expression, suggesting that the cord nerve cel function of corresponding spinal segments was inhibited. The spinal pia mater of spinal cord had a hierarchical or fracture phenomenon after subarachnoid block.
