Objective To investigate the effect and the underlying mechanism of miRNA (miR)-506 regulating transforming growth factor-β1 (TGF-β1)-induced endothelial-mesenchymal transition of human aortic endothelial cell lysates. Methods In this study,miR-506 and inhibitory members of the ASPP family (iASPP) were selected as the study objects.First, the expression of miR-506 in human aortic endothelial cell lysates (HAEC) lysate was detected under different concentrations of TGF-β1.The expression of platelet endothelial cell adhesion molecule-1 (CD31),endothelium-cadherin (VE-cadherin),fibroblastspecific protein-1 (FSP1),α-smooth muscle actin (α-SMA) and N-cadherin was determined after miR-506 mimics/mimic negative control (NC) transfection into HAEC treated with or without TGF-β1.Then the expression levels of iASPP in miRNA-506 mimics/mimic NC-transfected HAECs treated with or without TGF-β1 were determined. Finally, the protein expression of CD31,VE-cadherin,FSP1 and α-SMA in HAEC transfected with miR-506 mimics/mimic NC was determined after iASPP overexpression. Results TGF-β1 can induce human aortic endothelial cell mesenchymal transition,down-regulate CD31 and VE-cadherin expression while up-regulate FSP1,α-SMA and N-cadherin expression;TGF-β1 inhibited miR-506 expression, promoted iASPP expression;miR-506 transfection into HAECs up-regulated CD31 and VE-cadherin expression while up-regulate FSP1,α-SMA and N-cadherin expression;miR-506 could downregulate the expression of iASPP;the overexpression of iASPP increased the expression of FSP1,α-SMA and N-cadherin,down-regulated CD31 and VE-cadherin and promoted the transformation of human aortic endothelial cells. Conclusions The transfection of iASPP plasmid promoted the endothelial-mesenchymal transition of human aortic endothelial cell lysates. The transfection of miR-506 inhibited the expression of iASPP and inhibited the endothelial-mesenchymal transition of human aortic endothelial cell lysates induced by TGF-β1.

Aim To investigate the effect of chlorogen-ic acid, caffeic acid, and ferulic acid on expression of molecules related with inflammatory response of HMECs induced by activated complement alternative pathway. Methods CVF was used to activate the al-ternative pathway of serum complement. After exposure of HMECs to activate complement for various times, supernatant of cell culture was removed and assayed for content of ICAM-1,IL-6,IL-8,t-PA,and PAI-1 u-sing ELISA kits. The expression of the above mole-cules induced by activated complement was measured after HMECs were pre-treated with 50,100,250 μmol ·L-1 of CGA,CA,and FA.Results After HMECs were exposed to the product of the activated comple-ment alternative pathway, the expression of ICAM-1 , IL-6,IL-8,t-PA,and PAI-1 was up-regulated.The expression of ICAM-1,IL-6,IL-8,t-PA,and PAI-1 was down-regulated by various concentrations of CGA, CA,and FA.ICAM-1 and IL-8 were inhibited most significantly in all molecules mentioned above.CA ex-hibited the best intervention effect, followed by FA. Conclusion Certain concentration of CGA,CA,and FA can inhibit the expression of ICAM-1,IL-6,IL-8, t-PA,and PAI-1 in HMECs induced by the activation of the alternative complement pathway, indicating that CGA,CA,and FA can inhibit inflammatory response of HMECs.

Fulminant myocarditis is one of the most common diseases causing sudden cardiac death in pediatrics and young adults. The pathophysiology of fulminant myocarditis involves both direct myocyte dysfunction and immune-mediated tissue injury, which result in severe damage in cardiac structure and func-tion.It always rapidly developed into cardiogenic shock in several hours.Despite the high mortality rate,if pa-tient response well to a series of progressive treatment, most of them would get complete resolution.Better understanding of the hemodynamic changes of fulminant myocarditis is the key point of rapid diagnosis and proper interventional treatment. In this article we made a summary introduction of those hemodynamic chan-ges in fulminant myocarditis.

Chinese Journal of Medical Imaging Technology 2010;26(4):712-714
Objective To explore the value of contrast-enhanced ultrasonography (CEUS) in diagnosis of esophagogastric varices (EGV) compared with color Doppler flow imaging (CDFI). Methods CDFI and CEUS were performed on 23 variceal patients based on the B mode ultrasound examination. The blood flow of esophageal varices (EV) or gastric varices (GV) displayed with CDFI and CEUS were compared. Results The flow signal demonstration rate of EV with CDFI and CEUS was 42.11% (8/19) and 73.68% (14/19), while that of GV with CDFI and CEUS was 76.47% (13/17) and 94.12% (16/17), respectively. CEUS displayed the flow signal of EV better than CDFI did (χ~2=4.17, P<0.05), but the flow signal demonstration rate of GV was not significantly different (χ~2=1.33, P>0.05). Conclusion CEUS can be used more extensively in clinic practice for diagnosis of EGV.
