OBJECTIVE:To optimize the approach and methods for the selection of national essential drugs and to improve national essential drug system.METHODS:The concept of national essential drugs and the principle of selecting national essential drugs and the problems within were expounded,and the rationale and methods of pharmacoeconomics were adopted to analyze its significance and role in selecting national essential drugs.RESULTS & CONCLUSION:Simplifying National Essential Drug List can facilitate peoples’ access to essential drugs.

3.Analysis on moxibustion papers in SCI journals during the recent 5 years.

The papers regarding moxibustion published in science citation index (SCI) journals for the recent 5 years were searched to explore the international tendency of moxibustion researches, which provided references for moxibustion to have a better internationalization. With methods of internet search and database search, a total of 116 papers regarding moxibustion were included. These papers were published in 40 kinds of journals, mostly in Britain and the United States. The journal with the highest impact factor was Stroke, which had 5729 points. The number and impact factor of these journals were inferior to those of acupuncture journal. Compared among these journals, the depth and width of moxibustion research were increasing year by year. The category of diseases related with moxibustion is mainly digestive system diseases, motor system diseases and urinary-genital system diseases. The type of papers was characterized with clinical observation and mechanism research. Researches related with moxibustion included moxibustion dose and safety. It was believed that the international recognition of moxibustion effectiveness, standardization of moxibustion manipulation, standardization research, etc. were needed to be solved in the further.

4.Effects of internal and external biliary drainage on inducible nitric oxide synthase mRNA expression of Kupffer cell and serum tumor necrosis factor-alpha in rats with obstructive jaundice

Objective To study the effects of internal and external biliary drainage on inducible nitric oxide synthase (iNOS) mRNA expression of Kupffer cells and serum tumor necrosis factor-alpha (TNF-α) in rats with obstructive jaundice. Methods Forty-eight male Sprague-Dawley rats were randomly assigned to obstructive jaundice (OJ), sham operation (SH), internal biliary drainage (ID) and external biliary drainage (ED) groups with 12 each. Kupffer cells were isolated by in situ hepatic perfusion and digestion with collagenase type Ⅳ, and purified by cell culture attachment. The expression of iNOS mRNA of Kupffer cells was detected by reverse transcription polymerase chain reaction (RT-PCR) and the serum TNF-α concentration was measured using ELISA method. Results The serum TNF-α level was increased in OJ group[ (110.84±26.3) pg/ml ]compared with SH group [-(88.4±17.9) pg/ml, (P=0.045)]. The TNF-α level in ID group[ (89.8±28. 3) pg/ml ]was significantly lower than that in ED group[ (118.64±22.7) pg/ml, (P=0. 011) ]and OJ group (P=0. 059). Expression of iNOS mRNA of Kupffer cells was significantly higher in OJ group (0. 824± 0. 24) compared with SH group (0. 384±0.35,P=0. 005). After relieving the OJ, the iNOS mRNA expression in ED group (0. 974± 0.48) was not suppressed (P=0. 321). On the contrary, the iNOS mRNA expression in ID group was suppressed and significantly lower than that in ED group (0. 59±0. 35) (P=0. 016). Conclusions Internal biliary drainage is superior to external drainage in terms of reversing the elevated serum TNF-α and in suppressing the iNOS mRNA expression of Kupffer cells in rats with obstructive jaundice.
5.Association of human leukocyte antigen DP and DQ with postpartum thyroiditis:an analysis of 52 cases

Objective To study the genetic susceptibility in patients with postpartum thyroiditis(PPT)by genotyping their human leukocyte antigen(HLA)-DP and -DQ alleles.Methods The polymerase chain reaction-sequence based typing(PCR-SBP)method was used to analyze the distribution of HLA-DPA1,HLA-DPB1,HLA-DQA1 and HLA-DQB1 alleles among 52 PPT patients(31 clinical patients,21 subclinical patients)and 82 healthy controls.Compare the allele frequencies between various patient groups and the control population.Results There was no significant difference between patient group and control group.Conclusion Perhaps PPT is not associated with HLA-DP and HLA-DQ alleles.
6.Study on the Localization of a Major Allergen Der f 2 by Fluorescence Immunohistochemical Method

Objective To localize Der f 2 in the body of Dermatophagoides farinae.Methods Live mites were embedded.Serial mite paraffin sections were made and checked under light microscope.The sections were incubated with anti-recombinant Der f 2 monoclonal antibody as the first antibody.They were then reacted with the fluorescent isothiocyanate conjugated goat anti-mouse IgG as the secondary antibody.The sections were examined with fluorescent light microscopy.Results Der f 2 reacting with immunofluorescent antibodies was found localized in the tissue and contents of the mid-gut of the mite.Conclusion The major allergen Der f 2 distributes in the gut and fecal pellets of Dermatophagoides farinae.
7.Discussion on Standardization ISO 15189 quality management system as the teaching content for laboratory medicine

ISO 15189 standard quality management system developed by the International Organi-zation for Standardization (ISO) embodies the international medical laboratory's latest quality management concept, and puts forward the quality improvement method of the standard. In this research, ISO 15189, as an important content in practice teaching for laboratory medicine, was designed according to the elements of this system. Students were requested to participate in every linkage of laboratory quality management by writing system document, taking part in quality activity, and practicing the key and difficult points of tech-nique in quality system in order not only to cultivate their basic idea of ISO 15189 standard, but also to make them familiar with the basic patterns of laboratory accreditation system and process, which could make them become the backbone of the laboratory quality management quickly in their vocational positions in the future.
8.New strategy of constructing the ?_2m gene targeting vector

AIM: The purpose of this study was to establish a new strategy for constructing the mouse ? 2m gene targeting vector in order to increase the homologous recombination frequency in contrast with our previous one, which was successfully constructed in the normal way.METHODS: A 4.2 kb 3' arm and a 0.8 kb 5' arm were amplified by PCR from the mouse ? 2m-pSV2△HXgpt genomic clone. They included the start region and the three exons, which were separated into two parts from exons 2 (the main coding block) for the two arms——5' arm and 3' arm.RESULTS: The two fragments, in reverse orientation to the Neo gene, were cloned into pPNT respectively on either side of Neo. They were identified by PCR, restriction analysis and sequence analysis as well. CONCLUSION: The mouse ? 2m gene targenting vector has been cloned successfully.
9.The expression of the mouse ?_2 microglobulin in NIH3T3 cells transfected with antisense RNA

AIM: The expression of the mouse ? 2 microglobulin (? 2 m) in NIH3T3 cells transfected with the mouse ? 2m sense and antisense RNA was detected to clarify the effect of mouse ? 2m sense and antisense RNA on the expression of MHC classⅠgene. METHODS: The mouse sense and antisense RNA, pcDNA3-? 2mSN and pcDNA3-? 2mAN, were constructed and were transfected into NIH3T3 cells by lipofectamine. RT-PCR and Western blot were used to detect the expression of ? 2m in those cells. RESULTS: The expression of the mouse ? 2m in the cells transfected with pcDNA3-? 2mSN was increased, while it was decreased in those cells transfected with pcDNA3-? 2mAN. CONCLUSION: pcDNA3 -? 2mAN can downregulate the expression of the ? 2m in NIH3T3 cells.
10.Study on excretion of 20 (S) -protopanaxadiolocotillol type epimers in rats.

Gindenosides are the active ingredients of Panax ginseng. 20 (S) -protopanaxadiolocotillol type epimers are the main metabolites of 20 (S) -protopanaxadiol. The previous studies showed that there are stereoselectivity difference in pharmacodynamics and pharmacokinetics between 24R-epimer and 24S-epimer. The purpose of this study was to explore the excretion of the epimers in bile, feces and urine of rat. Liquid chromatography tandem mass spectrometry method has been performed for determination of 24R-epimer and 24S-epimer in bile, feces and urine. 24R-epimer or 24S-epimer was intragastric administered to rats at a single dose of 10 mg x kg(-1). Results showed that after administration the recovery of 24R-epimer and 24S-epimer in feces was 17.69% and 17.09%, respectively, while both of the two epimers were hardly detected in urine. The 48 h cumulative biliary excretion rate of 24R-epimer was 8.01% after administration, while only 1.47% for 24S-epimer. It indicated that there are stereoselectivity in biliary excretion of the epimers with intragastric administration.
