1.Optimization of Macroporous Resin for Isolation and Purification of Total Alkaloids from Rhizoma Coptidis

Objective To select a best type of macroporous resin for the isolation and purification of total alkaloids from Rhizoma Coptidis.Methods With the adsorption rate and desorption rate of total alkaloids from Rhizoma Coptidis as the observation indexes,an initial selection of macroporous resin was carried out in AB-8,HP20,LD605,ADS-3,ADS-5,D151,DA-201,XAD7 and NKA-9.After the initial selection,the desorption rate of berberine hydrochloride was used as the index for the further selection.Results Of the nine types of resin,ADS-3 had good adsorption and desorption activities,with an adsorption rate being 97.26% and a desorption rate being 84.82%.Meanwhile,the amount of desorpted berberine hydrochloride was up to 9.311 mg.Conclusion ADS-3 shows good adsorption and desorption properties for total alkaloids and berberine hydrochloride,and can be used for the isolation and purification of total alkaloids from Rhizoma Coptidis.

4.Research Development of ERK1/2 Pathway and Its Mediated Multiple Sclerosis

Extracellular signal-regulated kinases 1 and 2 (ERK1/2) pathway, which was the first cell signal transduction pathway to be discovered, consisted of Ras, Raf, MEK1/2 and ERK1/2. After the activation of ERK1/2 pathway, extracellular signals can be transmitted from the cell membrane to the nucleus. It was involved in many physiological and pathological functions of cells, such as growth, proliferation, differentiation, apoptosis and etc. It was also related to the onset of many diseases, which included multiple sclerosis (MS) and experimental autoimmune encephalomyelitis (EAE). The activation of ERK1/2 pathway can induce the activation of astrocyte, MG, T cell and macrophage, which released a variety of inflammatory cytokines. It caused myelin damage which induced MS/EAE onset. A number of studies indicated that inhibiting ERK1/2 pathway can reduce the releasing of inflammatory cytokines and myelin damage for MS/EAE alleviation. It provided an important target for the development of MS treatment medication.
5.Role of MAP kinase cascades in pain modulation

Mitogen-activated protein kinases (MAPKs) are important kinases involved in the intracellular signaling transduction in mammalian. There are three major MAPK pathways: ERK/MAPK,JNK/SAPK and P38 MAPK. Current studies indicate that all of them play significant roles in the processing of pain especially in the development and maintenance of pathological pain. This review will focus on in the study of MAPK in pain modulation. The further insights into its mechanisms may highlight a nice prospect for the therapies of painful diseases.
7.Curative effects observation of the combined therapy of Folic acid,Vitamin B_6 and B_(12) in youth stroke associated with hyperhomocysteinemia

Objective To observe the curative effects of the combined therapy of Folic acid,Vitamin(Vit) B6 and Vit B12 in youth stroke associated with hyperhomocysteinemia.Methods 150 youth stroke patients with hyperhomocysteinemia were randomly devided into low dose Folic acid,Vit B6 and Vit B12 treatment group(low dose group);high dose Folic acid,Vit B6 and Vit B12 treatment group(high dose group) and control group,50 cases in each group.Low dose group was taken by the treatments of Folic acid 2.5 mg/d,Vit B6 10 mg/d,Vit B12 0.5 mg/d;high dose group was taken by the treatments of Folic acid 5 mg/d,Vit B6 30 mg/d,Vit B12 1.5 mg/d;control group was not taken by the treatments of Folic acid,Vit B6 and Vit B12.The plasma homocysteine(Hcy)concentration was detected before and after treatment.The level of plasma Hcy 0.05).No obvious side effect was found in the three groups.Conclusions It is effective in the combined therapy of Folic acid,Vit B6 and Vit B12 in youth stroke associated with hyperhomocysteinemia.The effectiv rates are same by the low dose and high dose drug.The therapy is free from side effect.
8.Progress of cancer stem cells in solid tumors

Cancer stem cell (CSC) theory supposes that CSCs are the origin of tumors occurrence, progression,drug resistance, recurrence and metastasis. Cancer is not only a genetic disease but also a stem cell disease. Increasing evidence suggests the existence of CSCs in some solid tumors.
9.Cloning, expression, purification and activity assay of Leucyl-tRNA synthetase from Trypanosoma Brucei

Purpose To clone and express Trypanosoma Leucyl-tRNA synthetase (LeuRS) gene and to complete the purification and activity assay of LeuRS. Methods We cloned the LeuRS gene from T. Brucei genomic DNA,and cloned it into pBS-T and then into the expression vector pET21a( + ) .The expression of T. Brucei LeuRS was carried out in E. coli BL21 (DE3) RIPL host by optimizing the expression conditions. The products are purified by His-Bind affinity column and verified by Western blot . Enzymatic activity was detected by isotope labeling. Results A DNA fragment at length of 3.2 kb was amplified. Both restriction analysis and sequencing analysis proved that recombinant plasmid pET21a( + )/LeuRS was correctly constructed. The expressed product contained about 20% of total somatic soluble protein and reached a purity of 85% after purification . It was verified by the Western blot. Enzyme unit per millilitre of purified products was about 72. Conclusion Here we report the successful clone, expression and purification of LeuRS from Trypanosoma Brucei ( T. Brucei) in bacterial host. In addition, we tested its enzymatic activity by isotope labeling.This work would be helpful to the design and in vitro screening of Trypanosoma LeuRS inhibitors.

Brain-derived neurotrophic factor (BDNF) is one of the most prevalent growth factors in the central nervous system (CNS).In the development and maturation processes of the nervous system,BDNF plays an important role in maintaining neuronal function,promoting neuronal regeneration after injury,and preventing neuronal degeneration,etc.At present,many researchers are being dedicated to the research of BDNF for treatment of brain ischemia and have achieved some progress.This article reviews the molecular biological characteristics and biological function of BDNF,roles and mechanisms in cerebral ischemia,and the possibility as an intervention target of cerebral ischemia.
