1.Expression of GDNF and ChAT in the hippocampus of STZ-induced diabetic rats and relation between GD-NF and cognitive function

ObjectiveTo study the relationship between GDNF and ChAT Mrna and their potential role in the diabetic cognitive dysfunction by observing the change of the expression of GDNF and ChAT mRNA in brain under different blood glucose levels in STZ induced diabetic rats.MethodsForty male Wistar rats were randomly divided into diabetic group without blood glucose control( DM1 group),diabetic rats treated with insulin group( DM2 group),and normal control group( NC group).Blood glucose and body weight were detected every 4 weeks.Twelve weeks later,the hippocampus were submitted for cryostat sections which were subjected to the reverse transcription-polymerase chain reaction(RT-PCR) for GDNF and ChAT mRNA.ResultsThe expression of GDNF mRNA and ChAT mRNA in the hippocampus have negative correlation with HbAlc level( r =-0.962,-0.974,all P < 0.001 );The GDNF expression had positive correlation with the ChAT mRNA expression( r =0.974,P < 0.001 ).ConclusionLongterm chronic hyperglycation caused the “downregulation” of the GDNF expression in the hippocampal,which could cause the central cholinergic dysfunction symbled with the decrease of ChAT mRNA expression and the decline of learning and cognition ability.
2.Clinical efficacy for ptosis patients with severe undercorrection after frontal muscle flap suspension surgery

AIM:To observe the treatment for ptosis patients with severe undercorrection after frontal muscle flap suspension surgery. · METHODS: Totally 46 patients ( 67 eyes ) were randomly divided into 2 groups: 23 cases ( 33 eyes ) in Group A were given conjoint fascial sheath ( CFS ) suspension surgery; 23 cases ( 34 eyes ) in Group B Whitnall ligament suspension. Postoperative follow-up was 6mo. Corrected rate, recurrence rate and postoperative adverse reactions were compared and analyzed. ·RESULTS:The corrected rates of the two groups were both satisfied without significant difference between the two groups (P>0. 05). the Whitnall ligament suspension surgery had less recurrence than CFS suspension surgery, but the difference was not statistically significant (P>0. 05). The rate of hypophasis after CFS suspension surgery was significantly less than that after Whitnall ligament suspension (P<0. 05). ·CONCLUSION:The two operations both have satisfied corrected rate and low recurrence rate, the Whitnall ligament suspension surgery is less damaging and more easy to operate, while CFS suspension surgery is repeatable and less rate of hypophasis.
3.The effects of sustained +Gz condition and administration of depside salt from salvia miltiorrhiza on the dental pulp of rats

Objective:To investigate effects of depside salt from salvia miltiorrhiza(DSSM)on the dental pulp of rats under repeated exposure to +Gz acceleration.Methods:27 male SD rats were randomly divided into 9 groups(n =3).The rats in negative control group was only fixed with rats kept-keeling device,in positive control group were exposed to +5 Gz and +10 Gz respectively for 30 s each time,5 times per day,4 days per week,for 3 consecutive weeks.The rats in other groups were respectively given DSSMat 2,6 and 18 mg/kg respectively 30 min before each +Gz exposure.The rats in negative and positive control groups were injected with 0.9%sodium chloride solution 30 min prior to +Gz exposure.After the experiment,The pulp changes were observed by HE staining.Real time qPCR was applied to determine the relative expression of heat sheck protein(HSP70)mRNA.Results:+Gz exposure decreased the activity degree of the rats,and resulted in damage of rat dental pulp.With the increase of +Gz value,the relative expression of HSP70 mRNA was increased(P <0.05).HSP70 mRNA expression in drug groups was less than that in their own positive control groups,but showed statistical significance only in the 18 mg/kg and +10 Gz group(P <0.05).Conclusion:+Gz exposure may dam-age dental pulp.DSSMhas certain protective effect under +Gz exposure.

5.Clinicopathological observation and analysis of donor liver and biopsy in liver transplantation

Objective To investigate the relationship between histopathological changes of donor liver and those after liver transplantation and explore its effects on prognosis.Methods The histopathological changes of donor liver and post-transplantation live biopsy were observed in 87 cases.Results The percentage of hepatic degeneration at immediate biopsy of donor liver was 100%,the injury of sinusoid endothelial cell accounted for 41.3%,the hepatic steatosis for 18.3%and the necrosis of liver cells for 10.3%.Conclusion Most of the pathological changes of the donor liver during and after liver transplantation are reversible.The damage of the hepatic sinusoidal cells is the important criterion for the observation of ischemia reperfusion injury.Moderate and heavy hepatic steatosis can be used in the liver transplantation. After liver transplantation,if the necrosis of liver cells gradually gets serious,the risk of transplanted liver losing its function gets bigger,so the comparative observation has very important value in diagnosis.
7.Comparing the analgesic efficacy of femoral nerve block and sufentanil for positioning patients with hip fractures before spinal anesthesia

Objective To observe and compare the analgesic efficacy of femoral nerve block (FNB)and sufentanil for positioning patients with fractured hip.Methods Sixty patients scheduled for hip fracture surgery were randomized into three groups.Fifteen minutes before positioning for subarachnoid block,the FNB group received nerve ultrasound guided FNB using 20 ml ropivacaine 0.5% .Five minutes before positioning,the S group received a dose of IV sufentanil 0.15μg/kg,the C group didn't accept any intervention.The visual analog scale(VAS)following positioning (T2 ),OAA /S scale before positioning (T1 ),MAP,HR,SpO2 entering operating room (T0 )and T1 ,T2 ,adverse reactions were recorded.The plasma concentration of noradrenaline(NA)and cortisol(Cor)by T0 and T2 were measured.Results MAP,HR,the level of NA,Cor by T2 in group C increased significantly by comparing with T0 (P <0.05),while there were no obvious change in group F,and the same to group S except NA.The obvious change of SpO2 couldn't be found at each time point in every group(P >0.05).There were no significant differences in MAP,HR,the level of NA,Cor in all instances among the three groups(P >0.05).The OAA /S of S group was higher than the other groups(P <0.05).Compared with group C,group F and S showed markedly lower VAS at T2 (P <0.05),there were no significant differences between the two group.Side effects of group S included respiratory depression in 3 patients,nausea vomiting in 1 and dizzy in 1,no adverse reaction were found in other groups.Conclusions Both FNB and intravenous sufentanil can reduce the pain of positioning patients with fractured hip,showing almost the same analgesic efficacy,FNB can more inhibit stress reaction caused by pain.
8.A comparison between endoscopic variceal ligation and somatostatin for esophageal varices bleeding

Objective To evaluate the efficacy of endoscopic variceal ligation and somatostatin for esophageal varices bleeding.Methods Eighty patients with hepatic cirrhotic esophageal varices bleeding were allocated into two groups,forty in endoscopic variceal ligation(EVL)group and the others in somatostatin(stilamin)group.Results Initial hemostasis rates(including emergency hemostasis and no bleeding for 72 hours)and one-month rebleeding rates in EVL group and somatostatin group were 97.5% vs 80%(P0.05).The hepatic encephalopathy rates were 2.5% vs 15%(P0.05).Conclusion Endoscopic variceal ligation is superior to somatostatin for hepatic cirrhotic esophageal varices bleeding.EVL can lower the rate of hepatic encephalopathy.
9.Evaluation of retinal function after retinal transplantation for light-induced retinal degeneration in miniature pigs

Objective To investigate the possibility that transplanting the full-thickness neonatal piggy retinas that are completely differentiated but immature improves the retinal function after light-induced retinal degeneration in pigs.Methods Retinas from normal Guangxi Bama pigs aged 1-6 days were used as donor tissues.Neuroretina-RPE cografts were obtained from newborn pigs by using excimer laser for microablation of choroidal tissue and transplanted into the subretinal cavity of adult Bama pigs after light-induced retinal degeneration through vitrectomy and retinotomy.On days 5-7 and in 1st to 5th month after retinal transplantation,the survival of the cografts in the recipients and whether the host retinas have rejection were observed by ophthalmoscope,colour fundus photography and fundus fluorescein angiography,and the amplitude and lantency of N1,P1 waves between different periods were measured by mfERG.Results The retinal transplantation was performed in 15 eyes of 8 Bama pigs after light-induced retinal degeneration.The subretinal transplantation of the cografts was performed successfully in 11 eyes,with the operation success rate of 84.6%(11/15).In host retina,the gray-black graft inside transplantation bed could be seen clearly in 1st to 2nd month after transplantation and the leaked fluorescence in transplants was checked with FFA.The vertical comparison between different periods showed the amplitude of N1,P1 waves of retinal transplants rose with the extension of the survival time,and the areas where active response was observed were ring 2 and ring 3;but the latency of N1,P1 waves was shortened significantly in each ring as compared with that before operation,especially in late survival period.Conclusion The function measurement and the observation of living body together confirm the transplantation of completely differentiated retina from newborn pigs improves the retinal function of pigs after light-induced retinal degeneration.
10.Screening of proteins interacting with the cytoplasmic tail of NECL1 by yeast dual-hybrid system

Objective To screen the proteins interact with cytoplasmic tail of NECL1.Methods The cytoplasmic tail sequence of human NECL1 gene was inserted into the "bait" vector pGBKT7 in frame.The recombinant pGBKT7-NECL1C was then transformed into yeast cells and followed by screening of human fetal brain cDNA library.Then the GST pull down assay was used to confirm the interactions between the proteins.Results Nine different sequences by sequencing the positive clones and five alternative proteins were obtained by the amino acid sequence blast.Using the GST pull down analysis,we confirmed that two of them can interact with the cytoplasmic tail of NECL1 in vitro.Conclusion The alternative proteins obtained with the yeast dual-system may interact with the cytoplasmic tail of NECL1.
