Mesenchymal stem cells (MSC) possess an innate ability for self-renewal and can differentiate into multiple cell types.MSC have been used in cell-based therapy in various disease conditions including tumors.However,as MSC therapy gained popularity among practitioners and rescarchers,there have been reports on thre adverse effects of MSC especially in the context of tumor modulation and malignant transformation.Recent studies have shown that MSC also play an important role in the resistance to various anti-cancer drugs.This review provides an overview of the functional properties of MSC in cancer drug resistance.

Objective To investigate the serum levels of anti-BRAF antibody in patients with rheumatoid arthritis (RA) and its clinical significance.Methods Blood samples from 129 patients with RA,71 patients with other rheumatic diseases and 68 healthy controls were measured by enzyme-linked immunosorbent assays (ELISA).The serum levels of anti-BRAF antibody in patients with RA were divided into high,medium and low groups,each group had 43 cases.The clinical symptoms,immunological findings and related imaging data of all cases were recorded.We investigated the relationship between the serum levels of anti-BRAF antibody and the clinical symptoms,immunological findings and related imaging changes of the RA group.Chi-square test,Mann-Whitney U test,Spearman's correlation test were used for statistical analysis.Results ① The serum levels of anti-BRAF antibody were significantly higher in RA group [A:1.093 (0.150-3.210)] than in other rheumatic diseases group [A:0.278 (0.093-0.455)] and healthy controls [A:0.202 (0.121-0.443)] (Z=-11.223,P=0.021 ; Z=-7.839,P=0.008 7).The serum levels of anti-BRAF antibody was not significantly different between patients with other rheumatic diseases group and healthy controls (Z=-0.905,P=0.445).② The difference in positive rate of rheumatoid factor (RF) in different groups of anti-BRAF antibody levels was statistically significant (λx2=10.14,P=0.006),namely the positive rate was significantlyhigher in the low-level group (88％) than in the high-level group (58％) (x2=10.03,P=0.002).Thepositive rate of anti-cyclic citrullinated peptide antibody and anti-keratin antibody of the three groups was not significantly different (x2=0.721,P=0.697; x2=1.735,P=0.188).③ There was significant negative correlation between the levels of anti-BRAF antibody and the course of disease (r=-0.233,P=0.047),RF(r=-2.84,P=0.039),IgM (r=-0.234,P=0.038),and hemoglobin (r=-0.287,P=0.032).There was no significant correlation between the level of anti-BRAF antibody and the joint swelling index (r=-0.133,P=0.230),joint pain index (r=-0.173,P=0.100),other clinical and laboratory parameters.There was significantly negative correlation between the ESR and hemoglobin level.④ There was no significant difference of the level of anti-BRAF antibody between the normal group and patients with bone destruction in the RA group (Z=-0.642,P=0.521).⑤ There was no significant difference in the level of anti-BRAF antibody between the normal group and patients with interstitial pulmonary fibrosis group in the RA group (Z=-0.121,P=0.904).Conclusion The anti-BRAF antibody can be used for the diagnosis of RA as a new autoantibody,and it can also be used in the diagnosis of early or RF-negative RA.There is no correlation between the level of anti-BRAF antibody and bone destruction and interstitial pulmonary fibrosis.

Objective To establish a method for the content determination of baicalin in Jian' er Xiaosi Oral Liquid.Method A HPLC method was adopted.The chromatography conditions were as follows:Prodigy ODS(150 mm? 4.6 mm,5 ? m)served as stationary phase and methanol-water-glacial acetic acid(v/v,47:53:1.5)as mobile phase,the detection wavelength was 280 nm,column temperature 35 ℃ and flow rate 1.2 mL/min.Results The amount of inlet baicalin had a good linearity with the response value of peak area in the range of 0.284～ 3.540 ? g,r=0.999 9.The average recovery of baicalin was 100.33 % and RSD was 0.58 %(n=6).Conclusion This method is simple,rapid,reliable,reproducible,and can be used for the quality control of Jian' er Xiaosi Oral Liquid.

OBJECTIVE: To investigate the correlation between C-erbB-2 and angiogenesis of breast cancer and its clinical significance.METHODS: The expression of C-erbB-2,VEGF(vascular endothelial growth factor) and MVD(microvessel density) in breast cancer tissues were detected by two-step immunohistochemical method.RESULTS: For the 50 breast cancer specimens,the positive rate was 44 %(22/50) for C-erbB-2 vs.64%(32/50) for VEGF,and the MVD value was 25.96?11.59.The positive expression rates of C-erbB-2 and VEGF and MVD value in patients with lymph node metastasis of breast cancer were all higher than in those without lymph node metastasis of breast cancer(P0.05).C-erbB-2 expression was positively correlated with VEGF expression(P

Type 2 diabetes mellitus (T2DM) usually becomes complicated and unmanageable when combing with metabolic syndromes of obesity,hypertension and hyperlipidemia.Medical treatment generally adopts non-surgical management,such as diet control,exercise and oral medications; surgical treatment can improve the condition of patients with T2DM and the metabolic syndromes.In this article,the multi-disciplinary treatment of a patient with T2DM and obesity in the Third Xiangya Hospital of Central South University was analyzed,so as to provide a new way for the treatment of this disease.

Objective To evaluate the predictive value of diffusion weighted imaging ( DWI) for progres-sion free survival(PFS)in patients with cervix cancer after neoadjuvant chemotherapy (NACT).Methods Thirty two consecutive patients with pathologically confirmed cervical cancer underwent MRI including DWI before and after NACT.Pathologic results and MR images were reviewed .Univariate analysis was applied by Kaplan -Meier method.The Cox proportional hazard model was used to evaluate prognostic variables for multivariate analysis .Re-ceiver operating characteristic ( ROC) curves were used to find optimal cutoff values of independent prognostic fac -tors for disease progression.Results Of 32 patients,10 had disease progression during follow -up period.In uni-variate analysis,FIGO stage,tumor size,the depth of tumor invasion,lymph node metastasis and ADC changes be-tween before and after neoadjuvant chemotherapy (ΔADC) were significantly associated with the disease progres-sion.In multivariate analysis , FIGO stage and ΔADC were the independent prognostic factors for PFS .Optimal cutoff values that determined by ROC curves analyses were FIGO stage ⅡandΔADC 0.31.Area under the ROC curve(AUC)of FIGO stage and ΔADC were 0.841(sensitivity 90.0%,specificity 68.2%)and 0.864(sensitivity 80.0%,specificity 81.8%),respectively.Conclusion In patients with cervix cancer after NACT ,FIGO stage andΔADC are significant predictive values for PFS .

Objective To study the effect of PUMA gene mediated by recombinant adenovirus vector combined with radiation on the pancreatic carcinoma. Methods The PANC-1 cells were infected with Ad-PUMA (MOI = 10, 50 and 100, respectively) for 48 h. The expression of PUMA mRNA and protein was detected by RT-PCR and Western blot, respectively. PANC-1 cells were divided into 4 groups: control group, transfection group, irradiation group and combined treatment group. The cell growth inhibition rate and apoptotic rate of PANC-1 cells were assessed by MTT assay and flow cytometry. Human pancreatic carcinomas were transplanted subcutaneously in nude mice, which were randomized into 4 groups: control group, transfection group, irradiation group and combined treatment group. Tumor growth rate and apoptotie index at different time points were recorded in 35 days. Results The expression of PUMA mRNA and protein was increased with the increase of MOI of Ad-PUMA, which was does-dependant (MOI = 10, mRNA = 0.46 ± 0.02, protein = 0. 75 ±0.09;MOI=50, mRNA= 1.12±0.09, protein = 1.01 ±0.18;MOI= 100, mRNA= 1.50±0.08, protein=1.80 ± 0.15 ;P < 0.05). The proliferation of PANC-1 cells was suppressed significantly when transfected by Ad-PUMA in a dose-dependent manner(r = -0.986 55), which was more significant combined with radiation (r = -0.971 26, P < 0.05). Meanwhile, the apoptotie rate was increased in the same manner [for pre- and post-irradiation,which was (45.4 ± 5.26) % and (73.2 ± 6.62) %, respectively, P < 0.05]. From 7 to 35 d after PUMA gene transfection and radiotherapy, the tumor growth was significantly slower than those of irradiation group, transfection group and control group [35 d after therapy, the volume of tumor was (19.82 ± 6.45)mm3 ,(39.5 ± 9.23)mm3 , (33.6 ±3 10.3)mm3 and (52.0 ± 11.43)mm3 , respectively, P < 0.05]. And the apoptotic index was increased in the same manner (AI = 0.43 ± 0.05, 0.29 ± 0.10, 0.24 ± 0.05 and 0.00 ± 0.00, respectively, P < 0.05). Conclusions Recombinant adenoviral-mediated PUMA gene combined with irradiation could increase the cell-killing effect on pancreatic carcinoma. It is better than that of either one kind of therapy.

Objective To characterize the progression of segmental vitiligo.Methods Clinical data were collected using questionnaires from 387 patients with segmental vitiligo at the Department of Dermatology,Third People's Hospital of Hangzhou,between October 2011 and October 2012.The progression of segmental vitiligo was analyzed.Results Among the 387 patients,329 initially sufferred from focal vitiligo that evolved into segmental vitiligo later,58 began with segmental vitiligo.Vitiligo progressed most rapidly in the initial three months,and tended to be stable after three years in 220 (66.9％) of the 329 patients experiencing the evolution from focal to segmental vitiligo.By contrast,vitiligo developed most rapidly in the first month,and tended to be quiescent after one year in 40 (69.0％) of the 58 patients presenting with segmental vitiligo only.The condition still remained active in 27.6％ (107/387) of these patients after several years of progression.Totally,333 (86.0％) patients showed the involvement of single ganglion at the onset of vitiligo,including 173 (52.0％) patients with the involvement of trigeminal ganglion.Autologous melanocyte transplantation was conducted for 62 patients whose condition had been quiescent for more than one year,and 56 (90.3％) patients achieved more than 80％ repigmentation.Conclusions Segmental vitiligo,which tends to remain quiescent after three-year progression,seems to have a more rapid onset than focal vitiligo.Autologous melanocyte transplantation appears to be an effective treatment for segmental vitiligo in stable stage.
