BACKGROUND:There is a lack of safe and effective modular lentivectors in differentiation regulation of embryonic stem cel s. OBJECTIVE:To construct a modular lentivector, Puro-Nanog-hrGFP, with Nanog promoter-control ed humanized renil a reniformis green fluorescent protein (hrGFP) and puromycin and to observe the expression of Nanog during the differentiation of Nanog-hrGFP-transfected mouse embryonic stem cel lines. METHODS:After PCR amplification, Nanog, hrGFP and entry vector were recombined into the pDest-puro vectors to generate the lentiviral expression vector, Puro-Nanog-hrGFP, by the LR reaction. Through lentivirus production, transduction and puromycin screening, the transduced cel lines with Nanog-hrGFP gene were generated and identified. Expression of Nanog during the differentiation of transgenic mouse cel lines was detected. RESULTS AND CONCLUSION:The lentiviral expression vectors Puro-Nanog-hrGFP were constructed successful y by Gateway technology, and then the transducted cel lines were obtained by lentiviral infection. The expression of Nanog was gradual y decreased during the process of transgenic cel lines differentiation, which provides a new tool for further investigation on regulation of stem cel differentiation.

Journal of Audiology and Speech Pathology 2010;18(2):167-169
Objective To study the middle ear mucosa of guinea pigs with tympanosclerosis by electron microscope and try to obtain some insights into the feature and pathogenesis of tympanosclerosis. Methods Eight healthy variegated guinea pigs were devided into 2 groups.Six(8 ears) guinea pigs were subjected to inoculation of 1×10~8/L of staphylococcus aureus solution 100 μl into the middle ear cavities under the microscope. All the guinea pigs were observed for more than 6 months with no farther treatment. For electron-microscopic studies, the mucosa tissues were taken from the tympanic mucosa in 6 guinea pigs (8 ears) with tympanosclerosis from various sites, while the middle ear mucosa of two healthy guinea pigs (4 ears) were taken as a control. Results Uhrastructural examination of the normal middle ear mucosa revealed a few collagen fibers, normal morphous of fibrocyte, rough endoplasmic reticulum and mitochondria, and there was no lysosome. However, the tympanosclerosis specimens showed that irregular deformation, elongation, and degeneration of fibrocytes and oval nucleus were darkly stained, lots of mitochondria and lysosomes gathered into the cytoplasm around the nuclear and cystic expansion of the endoplasmic reticulum. In the submucosa extracellular matrix, there were a large number of collagen fibers containing lots of amorphous high-density electron-rich body. Conclusion Electron-microscopic studies of the middle ear mucosa of guinea pigs with tympanosclerosis revealed evident proliferation of collagen fibers, and calcifications were seen in the structures such as extracellular matrix vesicle, lysosomes, myelin structures within lipid granules, which mainly in extracellular matrix vesicles.

Objective To explore the clinical efficacy of ebridement and suturing combined with double vacuum sealing drainage (VSD) technology for the treatment of pressure ulcer. Methods Totally 32 patients of pressure ulcer (from July 2011 to October 2015) re-ceived debridement and suturing after infection control. Then F14 silicone ventricular drainage tube was placed in the wound for drainage while VSD was placed outside the wound. Both tubes inside and outside wound were connected to the central vacuum (20~40 kPa). Three days after the drainage,F14 silicone ventricular drainage tube was removed, and VSD material outside wound was replaced to continuous vacu-um treatment for another 3 to 4 days. Results All of 32 cases were healed up without hematoma, dead space and flap margin necrosis,inclu-ding 4 cases of poor healing, which were healed after active dressing. Conclusion Small area of the pressure ulcer can be closed up by de-bridement and suturing combined with double VSD technology, which is a simple operation with little injury and high clinical application value.
