1.Study of Apoptosis in Different Types of Renal Pathology in Children with Lupus Nephritis

Objective To detect the apoptosis and the intensity of proliferation in different types of renal pathology in children with lupus nephritis (LN), analyzed the relationship between apoptosis and proliferation in LN. Methods Twenty - seven children (aged 7-16 years old, 21 type IV and 6 type V ) with biopsy- proven LN and nine as controls were included in the study. Apoptosis was detected by in situ nick- end labeling techniques (TUNEL) in renal biopsy samples. Immunohistochemistry was employed to detect the proliferating cells identified by proliferating cell nuclear antigen (PCNA) and detect the expressions of proteins of apoptosis associated gene PDCD5 and Caspase - 3 in these patients. Results 1. Compared to type V LN, the patients with type IV LN had more apoptolic cells,more PCNA positive cells and higher ratios of PCNA/apoptosis (P/A) in glomeruli. 2. There were no difference in expression of PDCD5 in glomeruli in type IV LN compared with those in type V LN. Numbers of apoptotic cells were observed in glomeruli. The expression of Caspase- 3 in type IV LN increased in glomeruli compared with that in type V LN. Conclusions The up- regulation of mechanism of apoptosis in type IV LN was less than that of type V LN. Caspase- 3 participated in the apoptosis of glomeruli of LN, but PDCD5 did not play a role during apoptosis of glomeruli of LN or the effect of PDCD5 promoting apoptosis was depressed.
2.Expression changes of activin A in the process of hepatic fibrosis

Objective Activin A, a member of the transforming growth factor (TGF ?) superfamily, has been reported to overexpress in liver cirrhosis. The aim of this study was to examine the expression changes of activin A in the process of carbon tetrachloride induced rat hepatic fibrosis. Methods Hepatic fibrosis was induced in rats by subcutaneous injections of 40% carbon tetrachloride oily solution for a period of 1 to 7 weeks. 6~10 rats were killed at week 1,2,3,4,5,6 and 7 weeks. The kinetics of activin A messenger RNA expression and its protein localization were assayed by semi quantity reverse transcription polymerase chain reaction (RT PCR) and immunohistochemistry. Results RT PCR and immunohistochemistry showed normal rat liver could express activin A mRNA and protein, and its expression was transiently decreased and became undectable after carbon tetrachloride injections for 2 or 3 weeks, then increased grandually. 3.Effects of Cervical Physical Therapy and Excisional Therapy on the Prognosis of Cervical Precancerous Lesions

Objective To investigate the prognosis of cervical intraepithelial neoplasia (CIN) Ⅱ and Ⅲ after being treated by microwave coagulation, loop electrosurgical excision procedure (LEEP) and cold knife conization. Methods From March 2000 to February 2006, 289 patients with CIN Ⅱ or Ⅲ diagnosed by colposcopic biopsy were treated and followed up according to consensus guidelines for the management of women with CIN. Results Totally 233 of the 289 patients (80.6%) completed the follow-up (3-56 months with a mean of 11.8). Among them, 77.7% (181/233) received treatment; the other 22.3% (52/233) did not. In the 181 patients who received treatment, the lesion disappeared in 160 (88.4%) cases, during the follow-up; while in those who did not received treatment, the CIN was cured spontaneously in 63.5% (33/52; ?2=17.664, P=0.000). The prognosis of CIN Ⅱ was similar among the patients who were treated with microwave coagulation, LEEP, or cold knife conization (?2=0.008,P=0.996). In addition, in the patients with CIN Ⅲ, the prognosis after cold knife conization was not significantly different from that after LEEP (?2=0.017,P=0.897). Conclusions The management of CIN Ⅱ and Ⅲ may increase the cure rate. However, the three therapeutic methods produce similar prognosis.
4.Determination of Puerarin in Jiangzhijianfei Granules by HPLC

Objective To determine the content of Puerarin in Jiangzhijianfei Granules by high performance liquid chromatography.Methods The determination was conducted by HPLC using a Shim-pack-C18 column(5?m,150 mm?4.6 mm)and a mobile phase of methanol-0.1%Phosphoric acid water(25.3∶74.7).The flow rate was 0.9 mL/min and the detection wavelength was 250 nm.Results Good linear relationship between area and amount was noted for 0.114~0.570?g of Puerarin,with a correlation coefficient of 0.999 6.The average recovery of was 98.74%,RSD was 0.67%.Conclusion The established method is simple,accurate,sensitive and can be applied to the quality control of the preparation.
5.Investigation of Fluoride Source in Coal-burning Endemic Fluorosis Areas in Hunan Province

Objective To explore the origin of fluoride in the coal-combustion endemic fluorosis areas in the middle part of Hunan Province and to provide basic data for making preventive measure in the regions. Methods In November, 2006, the prevalence rate of dental fluorosis of children aged 10-12 years were examined, the villages were divided into three groups based on the prevalence rate of dental fluorosis. The fluoride content in the drinking water, coal, briquettes, briquette ash, clay and indoor air was determined by fluoride ion selective electrode. The quantity of fluoride release was calculated. Results There was no significant difference in the concentration of fluoride in water among the three groups, and it was below 1.0 mg/L in fluorosis and non-fluorosis areas. The fluoride levels in the coal, briquette ash and indoor air showed no statistical difference among groups. Objective To explore the adverse effect of endemic fluorosis caused by fluoride from coal burning on the intelligence of children. Methods Four primary schools were selected respectively from slight, moderate, severe endemic fluorosis areas and the control area in Xinhua county of Hu'nan Province, and twenty children of grade 5 were randomly chosen in each school. The seriousness of dental fluorosis was examined by Dean's method and intelligence level was evaluated by CRT-RC. The morning urine was collected and the fluoride content in urine was measured by fluoride ion selection electrode method. Auricular points of Lung (MA-IC1), Fengxi (MA-SF), EarShenmen (MA-TF1), Subcortex (MA-AT1), Adrenal Gland (MA-T) and Large Intestine (MA-SC4) were selected in the present paper, 25cases of skin pruritus were treated by combination of the auricular acupuncture and routine method, andanother 25 cases were treated with the routine method as the control group. The result showed better therapeutic effect in the former than the latter with statistical difference.

10.Analysis of HLA-DR expression in relation to the development of oral squamous cell carcinoma

Objective: To observe HLA DR expression in different s tages of oral squamous cell carcinoma (OSCC) and to study the clinic significan ce of the abnormal HLA DR expression in primary OSCC. Method: HLA DR expression was detected by immunohistochemistry method in 26 cas es of histologically normal oral epithelia, 8 leukoplakia, 32 primary focuses an d 12 metastasis focuses of OSCC. Results: HLA DR express ion in primary OSCC focuses was significantly higher than that in normal epithel ium( P 0.05). Conclusion: Although HLA DR expression is frequently obs erved in the development of OSCCs, it can not be regarded as one of the indepen dent prognosis factors.
