Clinical Medicine of China 2017;33(3):242-245
Objective To investigate the clinical value of thromboelastogram(TEG) in predicting the deep venous thrombosis(DVT) formation of bedridden senile patients and further explore the risk factors for hypercoagulability in those patients. Methods Patients lying on bed for at least 1 year because of alzheimer's disease, stroke and fracture, aged between 70 to 90 years old, were selected in the Geriatrics Department of Shougang Hospital of Peking University from February 2013 to February 2014.Those patients were divided by Clotting composite index(CI) measured with TEG,CI less than and equal to 3 was control group,CI more than 3 was hypercoagulable group. The patients were followed-up for one year to identify the formation of DVT.Logistic regression analysis was used to analyze the independent risk factors of elderly patients with high blood coagulation state in bed. Results Two groups of baseline blood clots elastic graph indicators:Angle(α) of solidification,CI,coagulation time(K),the biggest thrombosis(MA),blood coagulation reaction time(R) comparative differences were statistically significant(P<0.05).In view of the bedridden elderly patients of high coagulation state, according to the result of single factor analysis of related factors, high coagulation state revealed with age(P=0.047),history of diabetes(P=0.001),bed time(P=0.043),tumor(P=0.001),fracture(P=0.002),but unrelated with sex, blood lipids, blood pressure, alcohol consumption, left ventricular ejection fraction. The multivariate logistic regression analysis indicated that fracture(OR(95%CI):5.634(1.114~28.496),P=0.037),tumor(OR(95%CI):4.652(1.142~18.955),P=0.032) and diabetes mellitus(OR(95%CI):6.269(1.528~25.725),P=0.011) were independent risk factors for DVT.Conclusion The incidence of DVT in bedridden senile patients with hypercoagulability identified by CI is higher than those with normal coagulability.Tumor, fracture and diabetes mellitus are independent risk factors for DVT.

Objective To screen related gene of endometrial cancer by mRNA differential display (DD) polymerase chain reaction (PCR). Methods Individual mRNA from normal and neoplastic endometrium obtained from the same patient were comparatively studied by DD methodology. The mRNAs were isolated from the tissues, reverse transcribed to cDNA, and then amplified by PCR. The PCR products were displayed on a polyacrylamid gel by silver staining, and the differentially expressed bands were retrieved and reamplified. Subsequently we selected four bands for sequencing and making further investigation. Results cDNA homology was analysed by searching through GenBank sequence databases and indicated that tumor fragment (T1 7) has no significant sequence homology with any sequence of known function. Acession number is AF315581. Further study showed that T1 7 expressed more abundantly in endometrial cancer than hyperplasia endometrium, but not expressed in normal endometrium. Conclusions T1.7 is a new expressed sequence tag. It may be related gene fragment of endometrial cancer.

Objective To investigate the radiosensitization effect of the simultaneous silence of bcl-2 and XIAP on human head and neck cancer cell line in vitro. Methods Four groups of UMSCC12 cells were transfected with siRNA-bcl-2,siRNA-XIAP,siRNA-bcl-2 plus siRNA XIAP,and siRNA control, respectively. The silence efficiency was tested by Western blot assay. Apoptosis was evaluated with the activities of caspase-3 and caspase-9,and radiosensitization effect was evaluated with clonogenic assay. Results Bcl-2 and XIAP protein expressions were effectively eliminated in the cells simultaneously transfected with siRNA-bcl-2 and siRNA-XIAP.Transfection of cells with bcl-2 siRNA increased radiationinduced apoptosis ( t =5.32,6.27 ; P < 0.05 ),but transfection with XIAP siRNA did not impact cell apoptosis.Since the simultaneous transfection of the above two siRNAs,the activities of caspase-3 and caspase-9 were increased by 1.36 and 1.34 times ( t =11.47,6.22 ; P < 0.05 ) in nonirradiated cells and increased by 1.72 and 1.98 times ( t =12.02,20.14; P < 0.05 ) in irradiated cells, respectively. The colonic assay showed that the SERs were 1.06,1.15 and 1.41 for the cells transfected with XIAP siRNA,bcl-2 siRNA and both siRNAs,respectively. Conclusions Compared to single silence of XIAP or bcl-2,simultaneous silence of XIAP and bcl-2 offers a potential approach to improving the radiosensitivity of the head and neck cancer cells, and apoptosis might contribute to the enhancement of radiosensitization.
