3.The Expression and Significance of CRISP2 Gene in Different Types of Spermatogenic Failure

Objective To study sperm mRNA transcript expression of CRISP2 gene in the semen of patients with different types of spermatogenetic failure. Methods A total of 150 male infertility patients were divided into normal group, asthenozoospermia group and oligozoospermia group(n=50 for each group). Total RNA was extracted from sperm cells. Re-al-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) method was used to detect CRISP 2 gene mRNA levels in three groups. Results There was a significantly higher expression of CRISP2 in normal group (10.281 ± 2.173) than that of asthenozoospermia group (2.092±0.969, P<0.05). There was no significant difference in the expression of CRISP2 between normal group and oligozoospermia group (9.420±2.794, P>0.05). The relative expression of CRISP2 was sig-nificantly lower in asthenozoospermia group than that of oligozoospermia group (P<0.05). Conclusion The CRISP2 expres-sion was significantly different in patients with different types of spermatogenic failure. The reduced expression of CRISP 2 may lead to the decreased sperm motility.
4.Clinical observation of intravitreal injection of Conbercept treating diabetic macularedema

AIM: To observe the clinical efficiency of intravitreal conbercept on diabetic macular edema(DME).METHODS: This was a single arm, open-babel prospective study.Twenty eyes from 20 patients (12 males and 8 females) with DME diagnosed by fundus fluorescein angiography (FFA) and optical coherence tomography (OCT) were enrolled.Before the injection, best-corrected visual acuity (BCVA) of early treatment of diabetic retinopathy study (ETDRS), non-contact tonometer, ophthalmoscope, fundus photography, fundus fluoresein angiograph (FFA), and OCT were examined.All affected eyes were treated with intravitreal conbercept 0.05mL (10mg/mL).Patients were followed up for 6 to 11mo, with a mean duration of 8.55±1.96mo.Post-treatment BCVA, CMT, leakage of macular edema and complications were compared with baseline using repeat analysis.RESULTS: The initial average visual acuity (ETDRS letters) were 43.35±17.45, range from 9 to 70.The initial average central macular thickness (CMT) was 576.30±167.92μm, range from 337 to 987μm.The mean BCVA showed significant improvement during 1, 3, 6mo post-treatment and the latest follow up, with a mean increase of 11.2±5.9, 13.8±7.9, 15.7±6.8 and 14.7±8.6, respectively (P<0.01).The changes of BCVA between before and at 1mo after treatment were different compared with the changes between before and at 6mo (P<0.01).During the latest follow up, the mean BCVA was obviously improved in 10 eyes (50%), improved in 7 eyes (35%), stable in 3 eyes (15%).Likewise, the mean CMT significantly decreased during the follow-up period with a mean CMT reduction of 183.8±159.5, 292.9±169.0, 271.4±167.2 and 286.4±166.9μm respectively (P<0.001).The CMT at 1mo were different with that 3, 6mo and final follow-up (P<0.01).During the latest follow up, macula lutea leakage disappeared in 6 eyes (30%), decreased in 12 eyes (60%) and increased in 2 eyes (10%).No adverse events such as secondary retinal detachment or endophthalmitis were found during the follow-up.CONCLUSION: Intravitreal conbercept significantly improve visual acuity and macular edema exudation.
5.Polarization of retinal macrophages and (or) microglial cells and common ocular fundus diseases

Retinal macrophages and (or) microglial cells play important roles in regulating inflammation,angiogenesis and tissue repairing,thus affect the development and prognosis of ischemic retinal disease,ocular immune diseases and ocular tumors.Reversing the polarization imbalance of these cells may provide new therapeutic strategies for ischemic retinal disease and ocular immune diseases.The duality of the polarization direction of these cells is still controversial in the inflammatory reaction and pathological angiogenesis of ischemic retinal disease.Meanwhile,the plasticity and diversity of the function need to be further studied and discussed.
6.Establishment of fingerprint of processing Gentianae scraba Bge and its mutual mode by HPLC

AIM:To establish the HPLC fingerprint of methanol-resoluble component of processing Gentiana scraba Bge collected from different habits to provide evidence for quality control and quality evaluation of Gentiana scraba Bge.METHODS:Ten batches of processing Gentiana scraba Bge were measured by RP-HPLC,and the mutual mode was made by the similarity degree evaluating system 2004 of Chromatographic Fingerprint of Traditional Chinese Medicine promulgated by Chromatographic condition,consisted of Agilent-Eclipse-XDB-C_ 18-column and gradient elution with methanol-water,the detection wavelength was at 254 nm,flow rate was at 1.0 mL/min and the column temperature was at 20 ℃.RESULTS:The mutual mode of processing Gentiana scraba Bge was set up and the similarity of fingerprints of processing Gentiana scraba Bge from different habits was gained.CONCLUSION:According to the analysis of statistics,this method is steady,accurate with good repeatability and can be used as standardization implantation and quality control of Gentiana scraba Bge.
7.Significance of bFGF,FGFR-2 and Hpa expression in gastric precancerous lesions and gastric carcinoma tissues

9.Protective effect of fluvastatin on ischemic reperfused myocardium in rabbits

AIM: To investigate the protective effect of fluvastatin and its influence on ICAM-1 mRNA expression in ischemia/reperfusion myocardium of normocholesterolemic rabbits. METHODS: 24 rabbits were divided into three groups randomly and myocardial ischemia/reperfusion model in the rabbit was made. Rabbits were subjected to 45 min of regional myocardial ischemia and 2 h of reperfusion. 10 mg?kg~ -1 ?d~ -1 fluvastatin were administered for one week. Dynamic index of blood flow was recorded and analyzed. Serum activity of CK, CKMB, LDH and LDH-1 were measured. The expression of ICAM-1 mRNA in ischemic myocardium was detected with semi-quantitative RT-PCR. RESULTS: In comparison with control group, pretreatment with fluvastatin decreased LVEDP at the whole observed duration, and spontaneously increased ?dp/dt_ max . Serum activities of CK, CKMB and LDH-1 in control group were significantly higher than those in sham group, but heavily reduced in fluvastatin group. Increased expression of ICAM-1 mRNA due to ischemia reperfusion was reduced significantly in fluvastatin group compare to control group. CONCLUSION: Pretreatment of fluvastatin may reduce inflammation reaction in reperfused myocardium, and this may contribute to its protective effect against experimental myocardial ischemia reperfusion injury.
10.Optimization of the Inclusion Process of Essential Oil in Xiaoyu Anshen Capsules

OBJECTIVE:To optimize the inclusion process of the volatile oil of Pogostemon cablin and Acorus tatarinowii in Xiaoyu anshen capsule.METHODS:The inclusion process was carried out by saturated water solution method.And the inclusion process was optimized by orthogonal experiment with the inclusion ratio and yield of the volatile oil as indexes taking the ratio of volatile oil to ?-CD,the temperature and the time of inclusion as factors.The physical phase of the inclusion complex was detected by differential scanning calorimetry(DSC).RESULTS:The optimum inclusion process was established as follows:the ratio of oil to ?-CD=1∶8;the ratio of water to ?-CD=10∶1;the inclusion time was 1h and the inclusion temperature was 50 ℃.The inclusion rate was as high as 90%.The DSC revealed that a new physical phase has been formed for the inclusion complex.CONCLUSION:The intermolecular inclusion complex with good stability and high yield of volatile oil could be obtained from the optimized inclusion process.
